Erythropoietin is the principal hormone involved in the regulation of erythropoiesis by stimulating the proliferation and differentiation of erythroid progenitor cells and the maintenance of a physiological level of circulating erythrocyte mass. It is a glycoprotein produced primarily by the kidney. The biological effects of EPO are mediated by the erythropoietin receptor (Epo R). Genetic variations in EPO is associated with susceptibility to microvascular complications of diabetes type 2 (MVCD2) (including diabetic retinopathy, diabetic nephropathy leading to end-stage renal disease and diabetic neuropathy). It is used for the treatment of anemia and misused as a performance-enhancing drug in endurance athletes.

Description:
Epidermal growth factor (EGF) is a growth factor and the founding member of the EGF family. All EGF family members are synthesised as type I transmembrane precursor proteins that may contain several EGF domains in the extracellular region. The mature proteins are released from the cell surface by regulated proteolysis. EGF is present in various body fluids, including blood, milk, urine, saliva, seminal fluid, pancreatic juice, cerebrospinal fluid, and amniotic fluid. Four ErbB (HER) family receptor tyrosine kinases including EGFR/ErbB1, ErbB2, ErbB3 and ErbB4, mediate responses to EGF family members. These receptors undergo a complex pattern of ligand induced homo or heterodimerisation to transduce EGF family signals. EGF binds to the receptor EGFR stimulating the intrinsic protein-tyrosine kinase activity of the receptor. The tyrosine kinase activity initiates a signal transduction cascade that results in a variety of biochemical changes within the cell, including a rise in intracellular calcium levels, increased glycolysis and protein synthesis, and increases in the expression of certain genes including the gene for EGFR, which lead to DNA synthesis, cell growth, proliferation and differentiation. Other biological activities ascribed to EGF include epithelial development, angiogenesis, inhibition of gastric acid secretion, fibroblast proliferation, and colony formation of epidermal cells in culture. Defects in EGF are the cause of hypomagnesemia type 4 (HOMG4), also known as renal hypomagnesemia normocalciuric. HOMG4 is a disorder characterised by massive renal hypomagnesemia and normal levels of serum calcium and calcium excretion. Clinical features include seizures, mild-to mederate psychomotor retardation, and brisk tendon reflexes.

CD137 (4-1BB) is a member of the tumor necrosis factor (TNF) receptor family. CD137 is expressed by activated T cells, dendritic cells, NK cells, granulocytes and cells of blood vessel walls at sites of inflammation. CD137 has costimulatory activity for activated T cells. Crosslinking of CD137 enhances T cell proliferation, IL-2 secretion survival and cytolytic activity. CD137 was shown to enhance immune activity to eliminate tumours in mice.

B-cell activating factor belonging to the TNF family receptor (BAFF-R), B-cell maturation antigen (BCMA), and transmembrane activator and calcium modulator and cyclophilin ligand interactor (TACI) play critical functions in promoting B-cell survival by engaging the ligands a proliferation-inducing ligand (APRIL) and/or BAFF. TACI is predominantly expressed on peripheral blood B cells and interacts with the B cell activating factor (BAFF) of ligands and a proliferation inducing ligand (APRIL), which are expressed by numerous cell types, including dendritic cells, macrophages and neutrophils. TNFRSF13B, the gene, which encodes TACI is mutated in nearly 10% of patients with common variable immune deficiency (CVID), an antibody deficiency syndrome characterised by loss of memory B cells and plasma cells.

IL-2RB is the subunit critical for receptor-mediated signalling via physically or functionally coupling to other signalling molecules, such as the Jak-STAT and Src-family protein tyrosine kinase although it lacks apparent catalytic motifs.

Ubiquitin-Conjugating Enzyme E2 C (UBE2C) is a 179 amino acid enzyme that belongs to the Ubiquitin-Conjugating Enzyme family. UBE2C is highly expressed in tumour tissues and at low levels in most adult normal tissues. UBE2C is required for the destruction of mitotic cyclins and for cell cycle progression. UBE2C accepts Ubiquitin from the E1 complex and catalyses its covalent attachment to other proteins. It acts as an essential factor of the anaphase promoting complex/cyclosome (APC/C), which has E3 ubiquitin ligase activity, and targets for destruction substrates from the preceding mitosis (Cyclin A, Cyclin B, Securin, Geminin).

Hyaluronan-binding protein 2（HABP2） is an extracellular serine protease which binds hyaluronic acid. It secreted as an inactive single-chain precursor and is then activated to a heterodimeric form, which consists of a 50 kDa heavy and a 27 kDa light chain linked by a disulfide bond. HABP2 is involved in cell adhesion, it can cleave the alpha-chain at multiple sites and the beta-chain between 'Lys-53' and 'Lys-54' , but not the gamma-chain of fibrinogen. As a result of this, it does not initiate the formation of the fibrin clot and does not cause the fibrinolysis directly. It does not cleave prothrombin and plasminogen but converts the inactive single chain urinary plasminogen activator to the active two chain form, activates coagulation factor VII.

T-cell-specific surface glycoprotein CD28, contains an Ig-like V-type domain. CD28 is one of the proteins expressed on T cells that provide co-stimulatory signals, which are required for their activation. It is the receptor for CD80 and CD86. When activated by Toll-like receptor ligands, the CD80 expression is upregulated in antigen presenting cells (APCs). The CD86 expression on antigen presenting cells is constitutive. CD28 is the only B7 receptor constitutively expressed on naive T cells. It involved in T-cell activation, the induction of cell proliferation and cytokine production and promotion of T-cell survival.

FGF-acidic is a non-glycosylated heparin binding growth factor that is expressed in the brain, kidney, retina, smooth muscle cells, bone matrix, osteoblasts, astrocytes and endothelial cells. FGF-acidic has the ability to signal through all the FGF receptors. Recombinant murine FGF-acidic is a 15.9 kDa protein consisting of 141 amino acid residues.
