Description:
Coronin 6, a newly identified member of the coronin family, is highly enriched at adult NMJs and regulates AChR clustering via modulating the interaction between receptors and the actin cytoskeletal network. Coronins are a family of conserved actin-binding proteins originally identified in the actin-rich structure of the amoeba Dictyostelium discoideum . To date, seven members of coronins have been identified in mammals, and most exhibit tissue-specific distribution patterns. Coronin 6 is prominently expressed in adult muscle and enriched at the NMJ. Studies with cultured myotubes reveal that Coronin 6 regulates both agrin- and laminin-induced AChR clustering and is important for anchoring AChRs onto the actin cytoskeleton. Also, both the C-terminal region and a conserved Arg29 residue at the N terminus of Coronin 6 are essential for its actin-binding activity and stabilisation of AChR–cytoskeleton linkage. Importantly, in vivo knockdown of Coronin 6 in mouse skeletal muscle fibers leads to destabilisation of AChR clusters, which demonstrates that Coronin 6 is a critical regulator of AChR clustering at the postsynaptic region of the NMJs through modulating the receptor-anchored actin cytoskeleton. The human Coronin 6 has five isoforms produced by alternative splicing, and tissue-specific expression of these isoforms are unclear.

Neutrophil cytosol factor 1( NCF1) is a 47 kDa cytosolic subunit of neutrophil NADPH oxidase. This oxidase is characterised as a multicomponent enzyme which is activated to produce superoxide anion. NCF2, NCF1, and a membrane bound cytochrome b558 are required for the activation of the latent NADPH oxidase. The human NCF1 gene encodes a 390 amino acids protein without a signal peptide. The NCF1 gene interacts with other subunits of nicotinamide adenine dinucleotide phosphate-oxidase (NADPH) and plays an important role in innate immunity, producing reactive oxygen species and reducing the severity and duration of parasitic infection and autoimmune disease. NCF1 also has a role in T cell activation.

PROCR / EPCR / CD201 is a N-glycosylated protein, with a soluble form exists. PROCR is probably released by a metalloprotease. PROCR seems to have the same activity as the membrane-bound form. PROCR / EPCR / CD201 is expressed strongly in the endothelial cells of arteries and veins in heart and lung. PROCR binds activated protein C and enhances protein C activation by the thrombin - thrombomodulin complex. PROCR plays a role in the protein C pathway controlling blood coagulation.

Cluster of differentiation 14 (CD14), is a cell surface glycoprotein, and is a is a component of the innate immune system. CD14 is a myelomonocytic differentiation antigen preferentially expressed on monocytes, macrophages, and activated granulocytes. CD14 exists in two forms. Either it is anchored into the membrane by a glycosylphosphatidylinositol tail (mCD14) or it appears in a soluble form (sCD14). Soluble CD14 either appears after shedding of mCD14 (48 kDa) or is directly secreted from intracellular vesicles (56 kDa). CD14 acts as a co-receptor (along with the Toll-like receptor TLR 4 and MD-2) for the detection of bacterial lipopolysaccharide (LPS). CD14 can bind LPS only in the presence of lipopolysaccharide-binding protein (LBP). CD14 has been proposed to be involved in various biological processes, including transportation of other lipids, cell-cell interaction during different immune responses, as well as recognition of apoptotic cells. Although LPS is considered its main ligand, CD14 also recognizes other pathogen-associated molecular patterns. CD14+ cells are monocytes that can differentiate into a host of different cells. CD14 has been shown to interact with Lipopolysaccharide-binding protein.

CTLA-4 (Cytotoxic T-Lymphocyte Antigen 4) is also known as CD152 (Cluster of differentiation 152), is a protein receptor that downregulates the immune system. CTLA4 is a member of the immunoglobulin superfamily, which is expressed on the surface of Helper T cells and transmits an inhibitory signal to T cells. The protein contains an extracellular V domain, a transmembrane domain, and a cytoplasmic tail. Alternate splice variants, encoding different isoforms. CTLA4 is similar to the T-cell co-stimulatory protein, CD28, and both molecules bind to CD80 and CD86, also called B7-1 and B7-2 respectively, on antigen-presenting cells. CTLA4 transmits an inhibitory signal to T cells, whereas CD28 transmits a stimulatory signal. Intracellular CTLA4 is also found in regulatory T cells and may be important to their function. T cell activation through the T cell receptor and CD28 leads to increased expression of CTLA-4, an inhibitory receptor for B7 molecules. Fusion proteins of CTLA4 and antibodies (CTLA4-Ig) have been used in clinical trials for rheumatoid arthritis.

C-X-C chemokine receptor type 4 is also known as fusin or CD184 (cluster of differentiation 184), CXCR4, CD184, D2S201E, FB22, HM89, HSY3RR, LAP3, LCR1, LESTR, NPY3R, NPYR, NPYRL, NPYY3R or WHIM. CXCR-4 is an alpha-chemokine receptor specific for stromal-derived-factor-1 (SDF-1 also called CXCL12), a molecule endowed with potent chemotactic activity for lymphocytes. This receptor is one of several chemokine receptors that HIV isolates can use to infect CD4+ T cells. HIV isolates that use CXCR4 are traditionally known as T-cell tropic isolates. Typically, these viruses are found late in infection. It is unclear as to whether the emergence of CXCR4 using HIV is a consequence or a cause of immunodeficiency.CXCR4 is upregulated during the implantation window in natural and hormone replacement therapy cycles in the endometrium, producing, in presence of a human blastocyst, a surface polarization of the CXCR4 receptors suggesting that this receptor is implicated in the adhesion phase of human implantation. SDF-1 and CXCR4 were believed to be a relatively "monogamous" ligand-receptor pair (other chemokines tend to use several different chemokine receptors in a fairly "promiscuous" manner). Recent evidence demonstrates ubiquitin is also a natural ligand of CXCR4. Chronic exposure to THC increased T lymphocyte CXCR4 expression on both CD4+ and CD8+ T lymphocytes. Drugs that block the CXCR4 receptor appear to be capable of "mobilizing" hematopoietic stem cells into the bloodstream as peripheral blood stem cells.

Betatrophin (RIFL; Lipasin; Angiopoietin-like protein 8 (ANGPTL8)) is a newly discovered secreted protein of 198 aa that was proposed to promote beta cell proliferation and improve glucose tolerance in mice. Betatrophin may also function in inhibition of lipase activity and on serum triglyceride regulation. Betatrophin is expressed in the liver and in white and brown adipose tissue of mice. In humans, betatrophin is found to be predominantly expressed in the liver. Betatrophin levels are reduced by fasting and are elevated upon insulin resistance and during pregnancy. Betatrophin, according to preliminary data could bind to the macrophage receptor Marco and also to RTN4R, a neuronal receptor. Recently, a study using ANGPTL8 KO mice showed that ANGPTL8/Betatrophin does not play a role in beta cell proliferation nor in glycemic control as previously thought, but regulates plasma triglyceride levels.

The mature human FGFRL1 consists of a 354 amino acid extracellular domain (ECD) with 3 Ig-like C2-type domains, a 21 amino acid transmembrane segment, and a 134 amino acid cytoplasmic domain. FGFR1 expressed in various tissues, preferentially in cartilaginous tissues and pancreas. It highly expressed in the liver, kidney, heart, brain and skeletal muscle, weakly expressed in the lung, small intestine and spleen. FGFRL1 has a negative effect on cell proliferation.
