MHC class I polypeptide-related sequence A, also known as MIC-A, PERB11.1 and MICA, is a single-pass type I membrane protein which belongs to the MHC class I family of MIC subfamily. MICA contains one Ig-like C1-type domain and is expressed on the cell surface, although unlike canonical class I molecules does not seem to associate with beta-2-microglobulin. It is thought that MICA functions as a stress-induced antigen that is broadly recognised by NK cells, NKT cells, and most of the subtypes of T cells. MICA is the ligand for NK cell activating receptor KLRK1/NKG2D. MICA seems to have no role in antigen presentation. MICA leads to cell lysis by binding to KLRK1.

Chloride Intracellular Channel Protein 1 (CLIC1) belongs to the Chloride Channel CLIC family and contains one GST C-terminal domain. CLIC1 can be expressed in various cell types, but it is especially prominent in the heart, placenta, liver, kidney, and pancreas. CLIC1 can insert into membranes and form chloride ion channels. The channel activity depends on the pH. CLIC1 membrane insertion seems to be redox-regulated and may occur only under oxidising conditions. CLIC1 is also involved in the regulation of the cell cycle.

Tumour Necrosis Factor Receptor Superfamily Member 21 (TNFRSF21) is a type I transmembrane receptor that includes four extracellular cysteine-rich motifs and a cytoplasmic death domain. DR6 is highly expressed in heart, brain, placenta, pancreas, lymph node, thymus and prostate. DR6 may activate NF-kappa-B and JNK to promote apoptosis and T-cell differentiation. In addition, DR6 binds with N-APP, which is released by the deprivation of Trophic-factor. It triggers caspase activation and degeneration of both neuronal cell bodies (via caspase-3) and axons (via caspase-6). DR6 is also expressed on the tumour cell lines and can be induced by TNF- alpha.

BAFF is mainly produced by innate immune cells such as neutrophils, monocytes, macrophages, dendritic cells, follicular dendritic cells. T cells, activated B cells, some malignant B cells and also non-lymphoid cells like astrocytes, synoviocytes and epithelial cells can also produce BAFF. BAFF binds three distinct receptors (BAFF-R, TACI and BCMA) expressed predominantly on B cells, although activated T cells also express BAFF-R. BAFF is a master regulator of peripheral B cell survival, and together with IL-6, promotes Ig class-switching and plasma cell differentiation . Besides its major role in B cell biology, BAFF co-stimulates activated T cells. Deregulated expression of BAFF leads to autoimmune disorders in mice. In humans, elevated levels of soluble BAFF have been detected in the serum of patients with various autoimmune diseases, such as Sjögren’s syndrome , Rheumatoid Arthritis (RA) , Multiple sclerosis (MS) and Systemic Lupus Erythematosus (SLE) . BAFF is also increased levels in some lymphoid cancers.

Human Fatty Acid-Binding Protein 1 (FABP1) is a cytoplasm protein, which belongs to the calycin superfamily and Fatty-acid binding protein (FABP) family. Fatty acid binding proteins are a family of small, highly conserved, cytoplasmic proteins that bind long-chain fatty acids. FABP1 forms a beta-barrel structure that accommodates hydrophobic ligands in its interior. FABP1 can bind free fatty acids and their coenzyme A derivatives, bilirubin, and some other small molecules in the cytoplasm, so it can be involved in intracellular lipid transport.

Fatty Acid Binding Protein 3 (FABP3) is a small cytoplasmic protein (15 kDa) that is released from cardiac myocytes following an ischemic episode. Like the nine other distinct FABPs that have been identified, FABP3 is involved in active fatty acid metabolism where it transports fatty acids from the cell membrane to mitochondria for oxidation. FABPs are divided into at least three distinct types, namely the hepatic-, intestinal- and cardiac-types. They form 14-15 kDa proteins and are thought to participate in the uptake, intracellular metabolism and/or transport of long-chain fatty acids. They may also be responsible in the modulation of cell growth and proliferation. The FABP3 gene contains four exons and its function is to arrest growth of mammary epithelial cells. This gene is also a candidate tumor suppressor gene for human breast cancer. FABP3 is a sensitive biomarker for myocardial infarction and can be detected in the blood within one to three hours of onset of pain.

Recently, it was shown to suppress inflammasome activation by inhibiting caspase-1 activation, thus preventing processing of pro-IL-1beta into IL-1beta and the consequent phagosome maturation.

Protein Delta Homolog 1 (DLK-1) is a transmembrane protein which contains a signal peptide, an extracellular domain with six tandem epidermai growth factor (EGF)-like domains, a single pass transmembrane domain, and a short cytoplasmic tail. It is found within the stromal cells in close contact to the vascular structure of placental villi, yolk sac, fetal liver, adrenal cortex and pancreas and in the beta cells of the islets of Langerhans in the adult pancreas. In addition, it is detected in some forms of neuroendocrine lung tumor tissue. DLK-1 may have a improtant role in neuroendocrine differentiation.

Growing evidence suggests that many Kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen Kallikrein subfamily members located in a cluster on chromosome 19. Its encoded protein is secreted and may play a role in suppression of tumorigenesis in breast and prostate cancers. Alternate splicing of this gene results in multiple transcript variants encoding the same protein.
