ATP-binding cassette sub-family B member 5(ABCB5) is a plasma membrane-spanning protein. ABCB5 is principally expressed in physiological skin and human malignant melanoma. ABCB5 has been suggested to regulate skin progenitor cell fusion and mediate chemotherapeutic drug resistance in stem-like tumour cell subpopulations in human malignant melanoma. It is commonly over-expressed on circulating melanoma tumour cells. Furthermore, the ABCB5+ melanoma- initiating cells were demonstrated to express FLT1 (VEGFR1) receptor tyrosine kinase which was functionally required for efficient xenograft tumour formation, as demonstrated by shRNA knockdown experiments.

Sialic acid-binding Ig-like lectin 5 (SIGLEC5) is also known as CD33 antigen-like 2 (CD33L2), Obesity-binding protein 2 (OB-BP2), CD antigen CD170, which belongs to the immunoglobulin superfamily and SIGLEC (sialic acid binding Ig-like lectin) family. SIGLEC5 / CD170 is expressed by monocytic / myeloid lineage cells. SIGLEC-5 / CD33-L2 binds equally to alpha-2,3-linked and alpha-2,6-linked sialic acid. The sialic acid recognition site may be masked by cis interactions with sialic acids on the same cell surface.

CD160 antigen is a Lipid-anchor that exists as a disulfide-linked homomultimer. CD160 contains one Ig-like V-type domain. The human CD160 precursor is a cysteine-rich, glycosylphosphatidylinositol-anchored protein of 181 amino acids with a single Ig-like domain. It is weakly homologous to KIR2DL4. CD160 is expressed in the spleen, peripheral blood, and small intestine. Its expression is tightly associated with peripheral blood NK cells and CD8 T lymphocytes with cytolytic effector activity. CD160 is a receptor showing broad specificity for both classical and non-classical MHC class I molecules.

Cathepsin H (CTSH), which can act both as an aminopeptidase and as an endopeptidase, is a lysosomal cysteine protease of the papain family. CTSH is composed of a dimer of disulfide-linked heavy and light chains, both produced from a single protein precursor. CTSH is associated with various pathological conditions like human fibrous meningioma, colorectal cancer, arthritis, human prostate tumour and lung cancer. CTSH is associated with cancer progression because of their ability to degrade extracellular matrices facilitating invasion, angiogenesis and metastasis as is evident from numerous clinical reports and experimental models. The expression of CTSH is significantly increased in disease states such as in prostate tumours, sera of asthmatic patients, and mucosa of colorectal cancer patients.

OX40, also termed CD134 and TNFRSF4, is a T cell co-stimulatory molecule of the TNF receptor superfamily which plays a key role in the survival and homeostasis of effector and memory T cells. OX40 is expressed on CD4+ and CD8+ T cells upon engagement of the TCR by antigen presenting cells along with co-stimulation by CD40-CD40 Ligand and CD28-B7. The interaction between OX40 and OX40 ligand (OX40 L) will occur when activated T cells bind to professional antigen-presenting cells (APCs). The T-cell functions, including cytokine production, expansion, and survival, are then enhanced by the OX40 costimulatory signals. OX40 signals are critical for controllling the function and differentiation of Foxp3+ regulatory T cells. OX40-OX40L interaction regulates T-cell tolerance, peripheral T-cell homeostasis, and T-cell-mediated inflammatory diseases.

Bcl-2-like protein 1 (BCL2L1), is also known as Apoptosis regulator Bcl-X (Bcl-XL). BCL2L1 / Bcl-XL is a homodimer protein and belongs to the Bcl-2 family. BCL2L1 is expressed at high levels in cells that undergo a high rate of turnover, such as developing lymphocytes. BCL2L1 is a potent inhibitor of cell death. BCL2L1 Inhibits activation of caspases. ICOS ligand (ICOSLG) is also known as B7 homolog 2 (B7-H2), B7-related protein 1 (B7RP-1) and CD antigen CD275, which belongs to the immunoglobulin superfamily and BTN/MOG family. ICOSLG contains one Ig-like C2-type (immunoglobulin-like) domain and one Ig-like V-type (immunoglobulin-like) domain. Isoform 1 is widely expressed, while isoform 2 is detected only in lymph nodes, leukocytes and spleen. B7-H2 is ligand for the T-cell-specific cell surface receptor ICOS. B7-H2 acts as a costimulatory signal for T-cell proliferation and cytokine secretion and induces also B-cell proliferation and differentiation into plasma cells. B7-H2 could play an important role in mediating local tissue responses to inflammatory conditions, as well as in modulating the secondary immune response by co-stimulating memory T-cell function.

When the DNA glycosylase remove the damaged bases, APE1 cleaves the AP site to allow resynthesis and ligation to complete repair.
