Bone Morphogenetic Protein Receptor Type-1A (BMPR1A) belongs to the TKL Ser/Thr protein kinase family and TGFB receptor subfamily, including the type I receptors BMPR1A and BMPR1B and the type II receptor BMPR2. BMPR1A is a single-pass type I membrane protein and highly expressed in skeletal muscle. BMPR1A contains one GS domain and one protein protein kinase domain. BMPR1A is necessary for the extracellular matrix depostition by osteoblasts. BMPR1A can activate SMAD transcriptional regulators, binding with ligands. Defects in BMPR1A are a cause of juvenile polyposis syndrome, Cowden disease and hereditary mixed polyposis syndrome 2 (HMPS2).

Description:
Mouse Cxcl12 is a secreted and highly conserved protein which belongs to the intercrine alpha (chemokine CxC) family.CXCL12 is widely expressed in various organs including brain, kidney, skeletal muscle, heart, liver, and lymphoid organs. Cxcl12 activates the C-X-C chemokine receptor CXCR4 to induce a rapid and transient rise in the level of intracellular calcium ions and chemotaxis. It also binds to atypical chemokine receptor ACKR3 which activates the beta-arrestin pathway and acts as a scavenger receptor for SDF-1. Cxcl12 has several critical functions during embryonic development such as B-cell lymphopoiesis, myelopoiesis in bone marrow and heart ventricular septum formation. Cxcl12 plays an important role in acting as a positive regulator of monocyte migration and a negative regulator of monocyte adhesion via the LYN kinase. It stimulates migration of monocytes and T-lymphocytes through its receptors, CXCR4 and ACKR3, and decreases monocyte adherence to surfaces coated with ICAM-1, a ligand for beta-2 integrins. SDF1A/CXCR4 signaling axis inhibits beta-2 integrin LFA-1 mediated adhesion of monocytes to ICAM-1 through LYN kinase. It also plays a protective role after myocardial infarction, induces down-regulation and internalization of ACKR3 expressed in various cells and stimulates the proliferation of bone marrow-derived b progenitor cells in the presence of IL-7 as well as growth of the stromal cell-dependent B-cell clone DW34 cells.

Progranulin (PGRN) is a widely expressed pluripotent growth factor which plays a role in processes such as development, wound repair and inflammation by activating signalling cascades that control cell cycle progression and cell motility. Its function in the central nervous system is of interest, as mutations in the PGRN gene were found in cases of frontotemporal degeneration (FTLD). In addition, PGRN has also been linked to tumorigenesis. Progranulin is a biomarker for FTLD, other types of Alzheimer‘s Disease (AD) and potentially for MCI (Mild Cognitive Impairment). Additionally, PGRN is described as a new ligand of TNF receptors and a potential therapeutic against inflammatory disease like arthritis.

Mouse Il10 is the prototypic member of the IL-10 cytokine family, including IL-10, IL-19, IL-20, IL-22 (IL-TIF), IL-24 and IL-26. Many viruses encode viral members of the IL-10 family, such as Epstein-Barr virus (EBV) and human cytomegalovirus (HCMV).Its main function is inhibiting the synthesis of a number of cytokines, including IFN-gamma, IL-2, IL-3, TNF and GM-CSF produced by activated macrophages and by helper T-cells. Although human and mouse IL-10 are 81% identical at the nucleotide and amino acid level, mouse IL-10 is species-specific and does not act on human cells. Interestingly, Human IL-10 is active on mouse cells.

Mouse CCL24 is a secreted protein, which is a member of the CC chemokine subfamily. Mouse Ccl24 cDNA encodes a 119 amino acid residue precursor protein, shares approximately 58% amino acid sequence identity with human Ccl24. It is predominantly expressed in the jejunum and spleen and also be induced in the lung by allergen challengeand IL4. Mouse ccl24 has lower chemotactic activity for neutrophils but none for monocytes and activated lymphocytes. Ccl24 is chemotactic for resting T-lymphocytes, eosinophils and can bind to CCR3. LPS and IL4 also differentially regulate the expression of Ccl24 in monocytes and macrophages.

TNF-related apoptosis-inducing ligand (TRAIL; Apo2L;CD253; TNFSF10) is a type II transmembrane protein of about 34kDa. Like most members of the tumour necrosis factor (TNF) superfamily of cytokines TRAIL can be cleaved at the cell surface by metalloproteases to form a soluble molecule. Active TRAIL forms trimers and specifically binds to fi ve distinct receptors: TRAIL-R1 (DR4; Apo2;CD261; TNFRSF10A), TRAIL-R2 (DR5; KILLER; TRICK2A;TRICK2B; CD262; TNFRSF10B), TRAIL-R3 (DcR1;LIT; TRID; CD263; TNFRSF10C), TRAIL-R4 (DcR2;TRUNDD; CD264; TNFRSF10D), and osteoprotegerin (OPG; OCIF; TNFRSF11B). Interleukin-2 (IL-2) is a 133 amino acid glycoprotein with one intramolecular disulfide bond and variable glycosylation. It is secreted by activated T cells and induces proliferation and maturation of activated T cells, natural killer cells, and lymphokine activated killer cells. IL-2 also stimulates proliferation of antibody-producing B cells, activates neutrophils, and induces mononuclear cells to secrete IFN-gamma and TNF-alpha and -beta. Moreover, studies have shown that IL-2 is required for activation-induced apoptosis, an important hemeostatic mechanism in the immune system, which is involved in the maintenance of peripheral tolerance to self-antigens.
