Sonic hedgehog protein (SHH) is also known as HHG-1, which belongs to the hedgehog family. SHH is expressed in fetal intestine, liver, lung, and kidney and not expressed in adult tissues. Sonic hedgehog protein can be cleavage into sonic hedgehog protein N-product and sonic hedgehog protein C-product by autolysis. SHH binds to the patched (PTC) receptor, which functions in association with smoothened (SMO), to activate the transcription of target genes. In the absence of SHH, PTC represses the constitutive signaling activity of SMO. Also regulates another target, the gli oncogene. Intercellular signal essential for a variety of patterning events during development. The C-terminal domain displays an autoproteolysis activity and a cholesterol transferase activity. The N-product is the active species in both local and long-range signaling, whereas the C-product has no signaling activity.

Mouse interleukin 13 (mIL-13) is a pleiotropic cytokine produced by activated Th2 cells. IL-13 induces B cell proliferation and immunoglobin production. It contains a four helical bundle with two internal disulfide bonds. Mouse IL13 shares 58% sequence identity with human protein and exhibits cross-species activity. IL13 signals via receptor IL13R (type2, IL4R) and activates STAT-6. IL13 initially binds IL-13R alpha1 with low affinity and triggers association of IL4R alpha, generating a high affinity heterodimeric receptor IL13R and eliciting downstream signals. IL13 also binds IL-13R alpha2 with high affinity, which plays a role in a negative feedback system of IL13 signaling. IL13 is an important mediator of allergic inflammation and disease.

Chemokine (C-X-C Motif) Ligand 2 (CXCL2) is a small secreted cytokine which belongs to the CXC chemokine family. CXCL2 is 90% identical in amino acid sequence as a related chemokine, CXCL1. CXCL2/MIP2-alpha is secreted by monocytes and macrophages and is chemotactic for polymorphonuclear leukocytes and hematopoietic stem cells. The gene for CXCL2 is located on human chromosome 4 in a cluster of other CXC chemokines. CXCL2 mobilizes cells by interacting with a cell surface chemokine receptor called CXCR2. CXCL2/MIP2-alpha has been known to regulate immune functions mainly by chemo-attracting neutrophils. It is produced by activated monocytes and neutrophils and expressed at sites of inflammation. CXCL2 is a hematoregulatory chemokine, which suppresses hematopoietic progenitor cell proliferation. CXCL2 can be induced by receptor activator of NF-kappaB ligand, the osteoclast (OC) differentiation factor, through JNK and NF-kappaB signaling pathways in OC precursor cells. CXCL2 in turn enhanced the proliferation of OC precursor cells of bone marrow-derived macrophages (BMMs) through the activation of ERK. Knockdown of CXCL2 inhibited both the proliferation of and the ERK activation in BMMs. During osteoclastogenesis CXCL2 stimulated the adhesion and the migration of BMMs. CXCL2 is a novel therapeutic target for inflammatory bone destructive diseases.
