Ectodysplasin A receptor (EDAR) is a type I transmembrane protein of the TNF- alpha receptor superfamily which plays a key role in ectodermal differentiation. EDAR was encoded by the mouse down less gene and defective in human dominant and recessive forms of autosomal hypo hidrotic ectodermal dysplasia (EDA) syndrome. The extracellular domain of EDAR contains 14 cysteine residues, six of which approximate the TNFRSF cysteine-rich region, the cytoplasmic domain contains a region with homology to the death domains found in other TNFRSF members. EDAR has been suggested to be an early and important promoter of placode development in all ectodermal organs, such as uch as hair follicles, teeth and sweat glands. EDA-A1, the A1 isoform of EDA, is the EDAR ligand. EDA and EDA are implicated in appendage development by the cloning of a gene underlying hypo hidrotic ectodermal dysplasia (HED) in mouse and human. HED is Characterised by agenesis or malformation of ectoderm-derived appendages, such as teeth, sweat glands and hair follicles, while the skin itself develops normally.

Interleukin-4 (IL-4) is a pleiotropic cytokine that regulates diverse T and B cell responses including cell proliferation, survival and gene expression. IL-4 is produced by mast cells, T cells, and bone marrow stromal cells. IL-4 regulates the differentiation of naive CD4+ T cells into helper Th2 cells, Characterised by their cytokine-secretion profile that includes secretion of IL-4, IL-5, IL-6, IL-10, and IL-13, which favour a humoral immune response. Another dominant function of IL-4 is the regulation of immunoglobulin class switching to the IgG1 and IgE isotypes. Excessive IL-4 production by Th2 cells has been associated with elevated IgE production and allergic response.

Description:
Follistatin-like 1 (FSTL1) is a secreted glycoprotein that has been grouped into the follistatin family of proteins. FSTL1 is composed of a follistatin domain and two non-functional calcium-binding motifs. It was originally cloned as a TGF beta 1 inducible factor but subsequently shown to regulate diverse developmental pathways and tissue homeostasis. Ablation of the FSTL1 gene in the mouse results in several structural developmental defects and neonatal lethality due to respiratory failure. FSTL1 suppresses BMP signalling, but the precise mechanism of its action has not been elucidated. FSTL1 is expressed in the human placenta, mainly in extra villous trophoblasts.

Tumour necrosis factor receptor superfamily member 9(TNFRSF9), also known as CD137 and 4-1BB, is an inducible T cell surface protein belonging to the tumour necrosis factor receptor superfamily. It is a single-pass type I membrane protein which contains 4 TNFR-Cys repeats. The human and mouse proteins share 60% amino acid sequence identity. CD137 is expressed by mesenchymal cells, including endothelial cells, chondrocytes, and cells of the central nervous system. CD137 is also broadly expressed by cells of the human immune system, is broadly expressed by cells of the human immune system, including activated CD8+ and CD4+ T cells, activated natural killer (NK) cells, follicular dendritic cells (FDCs) and monocytes. CD137 has diverse roles in the immune response, the one key function is to promote the survival of both T cells and dendritic cells by binding the cognate ligand CD137L (4-1BBL).

Cadherin-16(CDH16) is a single-pass type I membrane protein which contains six cadherin domains. Mature cadherin proteins consist of a large N-terminal extracellular domain, a single membrane-spanning domain, and a small highly conserved C-terminal cytoplasmic domain. Cadherins are calcium-dependent cell adhesion proteins and may contribute to the sorting of heterogeneous cell types. They preferentially interact with themselves in a homophilic manner in connecting cells. Three calcium ions are usually bound at the interface of each cadherin domain and rigidify the connections, imparting a strong curvature to the full-length ectodomain. CDH16 is exclusively expressed in kidney, where the protein functions as the principal mediator of homotypic cellular recognition. It plays a role in the morphogenic direction of tissue development. CDH16 is composed of an extracellular domain containing 6 cadherin domains, a transmembrane region and a truncated cytoplasmic domain. However, it lacks the prosequence and tripeptide HAV adhesion recognition sequence typical of most classical cadherins.

Interleukin-12 subunit beta (IL-12B) belongs to the type I cytokine receptor family. It contains 1 fibronectin type-III domain and 1 Ig-like C2-type domain. IL-12B is a cytokine that acts on T and natural killer cells, and has a broad array of biological activities. IL-12 is a disulfide-linked heterodimer composed of the 40 kD cytokine receptor encoded by IL12B and a 35 kD subunit encoded by IL12A. IL12 is expressed by activated macrophages that serve as an essential inducer of Th1 cells development. It has been found to be important for sustaining a sufficient number of memory/effector Th1 cells to mediate long-term protection to an intracellular pathogen.
