Description:
CD200 is a transmembrane immunoregulatory protein that belongs to the immunoglobulin superfamily. It contains one Ig like V type domain and one Ig like C2 type domain in its extracelluar domain. CD200 is widely but not ubiquitously expressed. Its receptor (CD200R) is restricted primarily to mast cells, basophils, macrophages, and dendritic cells, which suggests myeloid cell regulation as the major function of CD200. CD200 and CD200R associate via their respective N-terminal Ig-like domains. In myeloid cells, CD200R initiates inhibitory signals following receptor-ligand contact. In T cells, CD200 functions as a co-stimulatory molecule independent of the CD28 pathway. In addition, CD200 also plays an important role in prevention of graft rejection, autoimmune diseases and spontaneous abortion.

Description:
Saporin is a ribosome-inactivating protein (RIP) of type I. This monomeric RNA N-glycosidase purified from seeds of the plant Saponaria officinalis also known as Soapwort, is capable of specific depurination of eukaryotic ribosomes thus arresting protein synthesis. No ligand has been identified in saporin hence its inability to transverse the cell membrane. Due to its toxicity and stability of the structure, saporin has proven extremely useful for construction of immunotoxins.

Ephrin type-B receptor 4(EPHB4) is also known as HTK, MYK1 and TYRO11,is a member of Eph family. The Eph family of receptors are divided into 2 groups based on the similarity of their extracellular domain sequences and their affinities for binding ephrin-A and ephrin-B ligands. Ephrin receptors make up the largest subgroup of the receptor tyrosine kinase (RTK) family. The protein encoded by EPHB4 binds to Ephrin-B2 and plays an essential role in vascular development. EPHB4 and its ligand ephrin-B2 are specifically expressed on venous and arterial endothelial cells, respectively, and play an essential role in vascular development via bidirectional signals. The forward EPHB4 signaling inhibits cell adhesion, chemotaxis, angiogenesis and tumor growth. Incontrast, the reverse Ephrin-B2 signaling exerts the opposite effect. It has been reported that aberrant expression of EPHB4 is associated with prostate cancer and highly malignant breast cancers, accordingly, EPHB4 has potential application as a therapeutic candidate.

Endothelial Differentiation-Related Factor 1 (EDF1) is a 148 amino acid transcriptional coactivator that contains 1 HTH cro/C1-type DNA-binding domain. It has been postulated that the protein functions as a bridging molecule that interconnects regulatory proteins and the basal transcriptional machinery, thereby modulating the transcription of genes involved in endothelial differentiation. When endothelial cells are induced to differentiate in vitro, EDF1 is downregulated, leading to inhibition of cell growth and cell polarization. EDF1 binds calmodulin thorough its IQ domain and regulates nitric oxide synthase activity through calmodulin sequestration in the cytoplasm. Though ubiquitously expressed, EDF1 is most abundant in adult liver, heart, adipose tissues, intestine and pancreas. In fetal tissues, EDF1 is most abundant in kidney. There are two isoforms of EDF1 that are produced as a result of alternative splicing events.

Chloride Intracellular Channel Protein 4 (CLIC4) is a 253 amino acid single-pass membrane protein that localizes to both the nucleus and the cytoplasm and contains one GST C-terminal domain. CLIC4 is expressed in various tissues and exhibits an intracellular vesicular pattern in Panc-1 cells (pancreatic cancer cells). CLIC4 acts as a monomer which is able to form selective ion channels in target proteins, thus facilitating the transport of chloride and other ions. CLIC4 is believed to have a role in apoptosis and is able to translocate to the nucleus under stress conditions. CLIC4 has alternate cellular functions like a potential role in angiogenesis or in maintaining apical-basolateral membrane polarity during mitosis and cytokinesis.

Nectin-3 is a type I transmembrane glycoprotein that belongs to the nectin family. Its precursor is 549 amino acids in length and contains an extended signal sequence of 57 amino acids, an extracellular domain (ECD) of 347 amino acids, a transmembrane segment of 21 amino acids, and a cytoplasmic region of 124 amino acids. It is predominantly expressed in testis and placenta as well as in various cell lines, including epithelial cell lines. Nectin-3 plays a role in cell-cell adhesion through heterophilic trans-interactions with nectin-like proteins or nectins, such as trans-interaction with PVRL2/Nectin-2 at Sertoli-spermatid junctions. Nectin-3 is also involved in the formation of cell-cell junctions, including adherens junctions and synapses.

Nogo Receptor (NgR) is a glycosylphosphoinositol (GPI)-anchored protein that belongs to the Nogo recptor family. Human NgR is predominantly expressed in neurons and their axons in the central nervous systems. As a receptor for myelin-derived proteins Nogo, myelin-associated glycoprotein (MAG) and myelin oligodendrocyte glycoprotein (OMG), NgR mediates axonal growth inhibition and may play a role in regulating axonal regeneration and plasticity in the adult central nervous system. LIF has the capacity to induce terminal differentiation in leukemic cells. Its activities also include the induction of hematopoietic differentiation in normal and myeloid leukaemia cells, the induction of neuronal cell differentiation and the stimulation of acute-phase protein synthesis in hepatocytes. LIF activates JAK & STAT signalling in human embryonic stem (ES) cells, but this pathway does not maintain pluripotency in these cells, which instead rely on FGF2-mediated ERK signalling. By contrast, mouse ES cells can be maintained by LIF-mediated JAK & STAT signalling. LIF binds to a high affinity heterodimeric receptor complex consisting of two proteins: LIF-R alpha that binds LIF with low affinity and the 130kDa (gp130) subunit that by itself does not bind LIF, but is required for high affinity binding of LIF.

It is implicated in the regulation of tumour cell growth, is involved in monocyte/macrophage-mediated immunological processes and functions as an important survival factor for plasmablasts and bone marrow plasma cells. MultimericAPRIL™ is a high activity construct in which two trimeric APRIL ligands are artificially linked via the collagen domain of ACRP30. This construct very effectively stimulates proliferation B cell. A basic amino acid sequence (QKQKKQ) close to the N-terminus of APRIL is required for binding to negatively charged sulphated glycosaminoglycan side chains of proteoglycans. Proteoglycans mediate binding of APRIL to tumour cells as well as primary lymphoid cells.
