Anthrax Toxin Receptor 1 (ANTXR1) is a single-pass type I membrane protein that belongs to the ATR family. ANTXR1 contains one VWFA domain and binds PA through the VWA domain. ANTXR1 is highly expressed in tumor endothelial cells. ANTXR1 plays a role in cell attachment and migration. ANTXR1 interacts with extracellular matrix proteins and the actin cytoskeleton, it mediates adhesion of cells to type 1 collagen and gelatin, reorganization of the actin cytoskeleton and promotes cell spreading. It is also involved in the angiogenic response of cultured umbilical vein endothelial cells, up-regulated in cultured angiogenic umbilical vein endothelial cells. Defects in ANTXR1 are associated with susceptibility to hemangioma capillary infantile (HCI).

neuregulin-1 (heregulin-1，NRG1) is a member of neuregulin family, which is comprised of four genes that encode a large number of secreted or membrane-bound isoforms. All family members share an EGF-like domain that interacts with the ErbB family of tyrosine kinase receptors. NRG1 isoforms can be classified into type I, type II and type III isoforms. NRG1 directs ligand for ERBB3 and ERBB4 tyrosine kinase receptors, concomitantly recruits ERBB1 and ERBB2 coreceptors, resulting in ligand-stimulated tyrosine phosphorylation and activation of the ERBB receptors. NRG proteins show distinct spatial and temporal expression patterns and play important roles during development of both the nervous system and the heart.

Cystatin-M is a typical secretory protein. It is synthesized as a preprotein with a patent N-terminal signal sequence.It belongs to the cystatin family. The most widely accepted function of cystatins is that of protease inhibitors. Most cysteine proteases are confined within cells where optimal pH and redox conditions favor their enzymatic activity. Thus, the majority of intracellular cysteine proteases are inactivated by oxidizing conditions outside the cells. Among the various types of intracellular cysteine proteases, cystatins seem to target preferentially endosomal/lysosomal cysteine proteases of the papain family, such as cathepsin B, cathepsin K/O2, cathepsin L, cathepsin L2/V and cathepsin S. Another important function of Cst6 seems to be in the terminal differentiation of stratified squamous epithelial cells and in the formation of cornified envelops. Cst6 is believed to be important in fine-tuning the enzymatic activities of endosomal/lysosomal cysteine proteases such as cathepsin L, cathepsin L2/V and AEP/mammalian legumain. Deregulated activity of these proteases could lead to abnormal activation of transglutaminases and disorders in cornification.

Members of the dickkopf-related protein family (DKK-1, -2, -3, and -4) are secreted proteins with two cysteine-rich domains separated by a linker region. Decorin is also known as bone proteoglycan II, PGS2, SLRR1B, DCN, DSPG2 and PG40, is a secreted chondroitin /dermatan sulfate proteoglycan in the family of small leucine-rich proteoglycans (SLRPs). Decorin is a small cellular or pericellular matrix proteoglycan and is closely related in structure to biglycan protein. Decorin and biglycan are thought to be the result of a gene duplication. This protein is a component of connective tissue, binds to type I collagen fibrils, and plays a role in matrix assembly. Decorin appears to influence fibrillogenesis, and also interacts with fibronectin, thrombospondin, the complement component C1q, epidermal growth factor receptor (EGFR) and transforming growth factor-beta (TGF-beta). Defects in DCN are the cause of congenital stromal corneal dystrophy (CSCD).

Description:
The IL-17 family include IL-17A, IL-17B, IL-17C, IL-17D, IL-17E (also called IL-25), and IL-17F. The family is comprised of at least six proinflammatory cytokines that share a conserved cysteine-knot structure but diverge at the N-terminus. All members of the IL-17 family have a similar protein structure, with four highly conserved cysteine residues critical to their 3-dimensional shape, yet they have no sequence similarity to any other known cytokines. IL-17 family members are glycoproteins secreted as dimers that induce local cytokine production and recruit granulocytes to sites of inflammation. IL-17 is induced by IL-15 and IL-23, mainly in activated CD4+ T cells distinct from Th1 or Th2 cells. IL-17F is the most homologous to IL-17, but is induced only by IL-23 in activated monocytes.
