CD83 antigen is a single-pass type I membrane protein which contains one Ig-like V-type (immunoglobulin-like) domain. CD83 is expressed by activated lymphocytes, Langerhans cells and interdigitating reticulum cells. It contains one Ig-like V-type (immunoglobulin-like) domain. , the soluble CD83 has the opposite effect and has an immune inhibitory capacity. Due to its immune inhibitory function, CD83 may play a significant role in antigen presentation or the cellular interactions that follow lymphocyte activation.

Cystatin SA is a member of family 2 of the Cystatin superfamily. Together with Cystatins S and SN, it is produced by the salivary gland and secreted largely in the submandibular/sublingual saliva. Cystatin SA inhibits Cathepsin L, but may not inhibit Cathepsins B and H. The functions of Cystatin SA are largely unknown. However, it has been suggested to play a role in the control of periodontal tissue destruction.

Human Kallikrein 11 (KLK11) is a member of tissue kallikrein family which are extracellular serine proteases consisting of 15 members. Two isoforms of KLK11 are differentially expressed. Isoform 1 is predominantly expressed in brain and isoform 2 is preferentially expressed in prostate. Isoform 1 consists of a signal peptide,a short pro peptide and the mature chain.Isoform 2 contains an extra 32 amino acid N terminal to full-length isoform 1.KLK11 is a novel marker for ovarian and prostate cancer carcinomas. KLK11 can be activated by thermolysin and is active against a thioester substrate.

Description:
Endothelial Protein C Receptor (EPCR) is a Vitamin K-dependent Serine Protease that plays a major role in blood coagulation. Binding of Protein C to EPCR leads to the proteolytic activation of PAR1 (Protease-Activated Receptor 1) on endothelial cells and subsequent up-regulation of Protein C-induced genes. EPCR is a type I transmembrane glycoprotein in the CD1/MHC family. It is expressed most strongly in the endothelial cells of arteries and veins in heart and lung. Membrane bound EPCR is released by metalloproteolytic cleavage to generate the soluble receptor. The extracellular domain of human and mouse EPCR shares approximately 61% amino acid sequence homology. EPCR plays an important role in augmenting Protein C activation by the Thrombin-Thrombomodulin complex and in regulating blood coagulation and inflammation. EPCR is found primarily on endothelial cells. Deletion of EPCR function results in embryonic death, at least in part due to placental thrombosis.

Tnfsf11 is widely expressed in cells including T cells and T cell rich organs, such as thymus and lymph nodes. This cytokine can bind to TNFRSF11B/OPG andTNFRSF11A/RANK. Tnfsf11 is involved in a number of fundamental biological processes such as acting as regulator of interactions between T-cells and dendritic cells, the regulation of the T-cell-dependent immune response and enhancing bone-resorption in humoral hypercalcemia of malignancy. It augments the ability of dendritic cells to stimulate naive T-cell proliferation.

OX40, also termed CD134 and TNFRSF4, is a T cell co-stimulatory molecule of the TNF receptor superfamily which plays a key role in the survival and homeostasis of effector and memory T cells. OX40 is expressed on CD4+ and CD8+ T cells upon engagement of the TCR by antigen presenting cells along with co-stimulation by CD40-CD40 Ligand and CD28-B7. The interaction between OX40 and OX40 ligand (OX40L) will occur when activated T cells bind to professional antigen-presenting cells (APCs). The T-cell functions, including cytokine production, expansion, and survival, are then enhanced by the OX40 costimulatory signals. OX40 signals are critical for controllling the function and differentiation of Foxp3+ regulatory T cells. OX40-OX40L interaction regulates T-cell tolerance, peripheral T-cell homeostasis, and T-cell-mediated inflammatory diseases.

The tetrameric receptor complex for IFN gamma consists of two subunits, IFNGR1 (IFN gamma R alpha) and IFNGR2 (IFN gamma R beta ), through which the dimeric IFN- gamma exerts its biological functions, including antiviral, antiproliferation and immune-modulatory activity in mammals. Both IFNGR1 and IFNGR2 are single transmembrane proteins belonging to the class II cytokine family. FNGR1, widely expressed in most host cells, is essential for IFN gamma binding, receptor trafficking, and signal transduction. IFNGR1 possesses an intracellular Janus tyrosine kinase (JAK) 1 binding site, a signal transducer and activator of transcription 1 (STAT1) binding site. The resulting STAT1 homodimers translocate from the cytoplasm to the nucleus and bind to the interferon-gamma activated sequence (GAS) promoter to induce expression of downstream interferon stimulated genes (ISGs).

Interleukin-3 (IL-3) is a potent growth promoting cytokine. IL-3 can stimulate the proliferation and differentiation of pluripotent hematopoietic stem cells as well as various lineage committed progenitors. IL-3 exerts its biological function through binding to specific cell surface receptors. The amino acid sequences of this protein among different species share relatively low identity and its activity is highly species-specific. The protein may activate NF-kappa-B and promote apoptosis and it may activate JNK and be involved in T-cell differentiation.It is required for both normal cell body death and axonal pruning. Trophic-factor deprivation triggers the cleavage of surface APP by beta-secretase to release sAPP-beta which is further cleaved to release an N-terminal fragment of APP (N-APP). N-APP binds TNFRSF21 triggering caspase activation and degeneration of both neuronal cell bodies (via caspase-3) and axons (via caspase-6).

RSPO1 is a secreted protein,containing 2 FU(furin-like) repeats and 1 TSP type-1 domain and belonging to the R-spondin family. RSPO1 is required for the early development of gonads, regardless of sex. It has been found in mice only eleven days after fertilization. To induce cell proliferation, it acts synergistically with WNT4. They help stabilize beta catenin, which activates downstream targets. RSPO1 is necessary in female sex development. It augments the WNT/ beta catenin pathway to oppose male sex development. In critical gonadal stages, between six and nine weeks after fertilization, the ovaries upregulate it while the testes downregulate it. RSPO1 can potentially aid in the treatment of mucositis, which is characterized by inflammation of the oral cavity. This unfortunate condition often accompanies chemotherapy and radiation in cancer patients with head and neck tumors.

P-selectin glycoprotein ligand 1 is the high affinitycounter-receptor for P-selectin on expressed on activated endothelial cells and platelets. As such, it plays a critical role in the tethering of these cells to activated platelets or endothelia expressing P-selectin. As cell adhesion molecules, multiple studies have shown that PSGL-1/ P-selectin interaction is required for the normal recruitment of leukocytes during inflammatory reactions, and also participates in hemostatic responses. PSGL-1 can also bind to other two members of the selectin family, E-selectin (endothelial) and L-selectin (leukocyte), but binds best to P-selectin.

T-cell-specific surface glycoprotein CD28(CD28) is a single-pass typeI membrane protein which contains one Ig-likeV-type (immunoglobulin-like) domain. It belongs to the immunoglobulin(Ig) superfamily. CD28 is one of the molecules expressed on T cells that provide co-stimulatory signals, which are required for T cell activation.CD28 co-stimulation is necessary for CD4 positive T-cell proliferation and survival, interleukin-2 production, and T-helper type-2 development. Human post-thymic regulatory T cells require CD28 co-stimulation to expand and maintain potent suppressive function in vivo. Apoptosis plays a key role in the age-related decline of CD28 expression and in immunosenescence. CD28 is the receptor for CD80 (B7.1) and CD86 (B7.2). When activated by Toll-like receptor ligands, the CD80 expression is upregulated in antigen presenting cells (APCs). The CD86 expression on antigen presenting cells is constitutive. CD28 is the only B7 receptor constitutively expressed on naive T cells.

Homeobox B4 (HOXB4) is encoded by the HOXB4 gene which is a member of the the class I homeobox (HOX) gene family and encodes a nuclear protein with a homeobox DNA-binding domain. These genes are master control regulators of developmental programs including embryonic and adult hematopoiesis. Multiple HOX genes, including HOXB4, are highly expressed in the hematopoietic stem cells (HSC) compartment. HOXB4 gene can act in opposite ways when expressed by different cells, promoting the proliferation of stem cells whilst activating the apoptotic pathway in some embryonic structures. The protein HOXB4, as a homeodomain transcription factor, has been shown to be an important regulator of stem cell renewal and hematopoiesis. Incellular or ectopic expression of HOXB4 expands hematopoietic stem and progenitor cells in vivo and in vitro, making it a potential candidate for therapeutic stem cell expansion.
