tumour necrosis factor receptor superfamily member 25 (TNFRSF25) contains 1 death domain and 4 TNFR-Cys repeats. TNFRSF25 is a cell surface receptor of the tumour necrosis factor receptor superfamily which mediates apoptotic signalling and differentiation, activated by a monogamous ligand, known as TL1A (TNFSF15), which is rapidly upregulated in antigen presenting cells and some endothelial cells following Toll-Like Receptor or Fc receptor activation. This receptor has been shown to signal both through the TRADD adaptor molecule to stimulate NF-kappa B activity or through the FADD adaptor molecule to stimulate caspase activation and regulate cell apoptosis. It may play a role in regulating lymphocyte homeostasis.

Interleukin-2 receptor subunit alpha (IL2RA) is a single-pass type I membrane protein, contains 2 Sushi (CCP/SCR) domains. The interleukin 2 (IL2) receptor alpha (IL2RA) and beta (IL2RB) chains, together with the common gamma chain (IL2RG), constitute the high-affinity IL2 receptor. Homodimeric alpha chains (IL2RA) result in low-affinity receptor, while homodimeric beta (IL2RB) chains produce a medium-affinity receptor. Normally an integral-membrane protein, soluble IL2RA has been isolated and determined to result from extracellular proteolyisis.

Asialoglycoprotein Receptor 1 (ASGPR1) is an endocytic recycling receptor, belongs to the long-form subfamily of the C-type/Ca2+-dependent lectin family. ASGPR consists of two noncovalently-linked subnits, ASGPR1 and ASGPR2. ASGPR1 mediates the endocytosis of plasma glycoproteins, recognizes terminal galactose and N-acetylgalactosamine units. When the ligand binds to to ASGPR1, results in the complex is internalized and transported to a sorting organelle, then ASGPR1 and ligand can be disassociated, ASGPR1 returns to the cell membrane surface.

Cytochrome C (CYCS) is a small heme protein that belongs to the cytochrome c family. It is found loosely associated with the inner membrane of the mitochondrion. Cytochrome C is a highly soluble protein that functions as a central component of the electron transport chain in mitochondria. CYCS transfers electrons between Complexes III (Coenzyme Q - Cyt C reductase) and IV (Cyt C oxidase). CYCS plays a role in apoptosis. Suppression of the anti-apoptotic members or activation of the pro-apoptotic members of the Bcl-2 family leads to altered mitochondrial membrane permeability resulting in release of cytochrome c into the cytosol. Binding of Cytochrome C to Apaf-1 triggers the activation of caspase-9, which then accelerates apoptosis by activating other caspases.

As protease inhibitors, serpins have an array of functions including regulating blood coagulation, fibrinolysis, the complement pathway, angiogenesis, inflammation, tumour suppression, extracellular matrix remodelling, and cell motility. SERPINB1 regulates the activity of the neutrophil proteases elastase, cathepsin G, proteinase-3, chymase, chymotrypsin, and kallikrein-3. Reactive bond 1 of SerpinB1 is specific for reaction with chymotrypsin-like protease such as cathepsin G, chymotrypsin or chymase. Reactive bond 2 of SerpinB1 is specific for reaction with elastase-like protease such as neutrophyl elastase, proteinase-3, pancreatic elastase or PSA. In addition, SERPINB1 also functions as a potent intracellular inhibitor of granzyme H.

Follistatin-like 1 (FSTL1) is a secreted glycoprotein that has been grouped into the follistatin family of proteins. FSTL1 is composed of a follistatin domain and two non-functional calcium-binding motifs. It was originally cloned as a TGF beta 1 inducible factor but subsequently shown to regulate diverse developmental pathways and tissue homeostasis. Ablation of the FSTL1 gene in the mouse results in several structural developmental defects and neonatal lethality due to respiratory failure. FSTL1 suppresses BMP signalling, but the precise mechanism of its action has not been elucidated. FSTL1 is expressed in the human placenta, mainly in extra villous trophoblasts.

Tumour necrosis factor receptor superfamily member 9(TNFRSF9), also known as CD137 and 4-1BB, is an inducible T cell surface protein belonging to the tumour necrosis factor receptor superfamily. It is a single-pass type I membrane protein which contains 4 TNFR-Cys repeats. The human and mouse proteins share 60% amino acid sequence identity. CD137 is expressed by mesenchymal cells, including endothelial cells, chondrocytes, and cells of the central nervous system. CD137 is also broadly expressed by cells of the human immune system, is broadly expressed by cells of the human immune system, including activated CD8+ and CD4+ T cells, activated natural killer (NK) cells, follicular dendritic cells (FDCs) and monocytes. CD137 has diverse roles in the immune response, the one key function is to promote the survival of both T cells and dendritic cells by binding the cognate ligand CD137L (4-1BBL).
