TNF-related apoptosis-inducing ligand (TRAIL; Apo2L;CD253; TNFSF10) is a type II transmembrane protein of about 34kDa. Like most members of the tumor necrosis factor (TNF) superfamily of cytokines TRAIL can be cleaved at the cell surface by metalloproteases to form a soluble molecule. Active TRAIL forms trimers and specifically binds to five distinct receptors: TRAIL-R1 (DR4; Apo2;CD261; TNFRSF10A), TRAIL-R2 (DR5; KILLER; TRICK2A;TRICK2B; CD262; TNFRSF10B), TRAIL-R3 (DcR1;LIT; TRID; CD263; TNFRSF10C), TRAIL-R4 (DcR2;TRUNDD; CD264; TNFRSF10D), and osteoprotegerin (OPG; OCIF; TNFRSF11B). Trimerised TRAIL triggers apoptosis upon ligation of cell surface TRAIL-R1 and/or TRAIL-R2 by inducing the formation of the so-called multiprotein death-inducing signalling complex (DISC).

Description:
Folate receptor alpha(FOLR) belongs to the folate receptor family, and is primarily expressed in tissues of epithelial origin. It is also expressed in kidney, lung and cerebellum. The secreted form is derived from the membrane-bound form either by cleavage of the GPI anchor, or/and by proteolysis catalyzed by a metalloprotease. FOLR1 binds to folate and reduced folic acid derivatives and mediates delivery of 5-methyltetrahydrofolate and folate analogs into the interior of cells. It has high affinity for folate and folic acid analogs at neutral pH. Exposure to slightly acidic pH after receptor endocytosis triggers a conformation change that strongly reduces its affinity for folates and mediates their release. It is required for normal embryonic development and normal cell proliferation.

CD83 is expressed by activated lymphocytes, Langerhans cells and interdigitating reticulum cells. It contains one Ig-like V-type (immunoglobulin-like) domain, the soluble CD83 has the opposite effect and has an immune inhibitory capacity. Due to its immune inhibitory function, CD83 may play a significant role in antigen presentation or the cellular interactions that follow lymphocyte activation.

FNDC5 is a membrane protein comprised by a short cytoplasmic domain, a transmembrane segment, and an ectodomain consisting of a fibronectin type III (FNIII) domain. The ectodomain has been proposed to be cleaved to a soluble peptide hormone named “irisin”. Irisin is secreted from muscle in response to exercise, and may regulate energy metabolism, stem cell differentiation, and neuronal development. The circulating irisin converts white fat into the more thermogenic beige fat, and may mediate some beneficial effects of exercise in humans and the potential for generating weight loss. In mice, expression of Irisin has been shown to regulate obesity and diabetes. A similar function is suggested in human.

TNFRSF10B is a member of the TNF-receptor superfamily, and contains an intracellular death domain. This receptor can be activated by tumor necrosis factor-related apoptosis inducing ligand (TNFSF10/TRAIL/APO-2L), and transduces apoptosis signal. The adapter molecule FADD recruits caspase-8 to the activated receptor and is required for the apoptosis mediated by TNFRSF10B. TNFRSF10B is expressed in a number of cell types, and to particularly high levels in lymphocytes and spleen. This single-pass transmembrane protein contains two cysteine-rich repeat units in its extracellular region, followed by a transmembrane segment and a cytoplasmic tail containing a typical “death domain”. TNFRSF10B expression is regulated by the tumor suppressor p53. It is also indicated that the activation of NF-kappa-B can be promoted by TNFRSF10B.

Calreticulin-3 belongs to the calreticulin family, members of which are calcium binding chaperones localised mainly in the endoplasmic reticulum. It can be divided into a N-terminal globular domain, a proline-rich P-domain forming an elongated arm-like structure and a C-terminal acidic domain. During spermatogenesis process, Calreticulin-3 may act as a lectin-independent chaperone for specific client proteins such as ADAM3. Defects in CALR3 are the cause of familial hypertrophic cardiomyopathy type 19 (CMH19), it is a hereditary heart disorder characterised by ventricular hypertrophy, which is usually asymmetric and often involves the interventricular septum. The symptoms include dyspnea, syncope, collapse, palpitations, and chest pain.
