Cartilage oligomeric matrix protein (COMP) is also known as Thrombospondin-5 (TSP5), EDM1, EPD1, MED, PSACH, THBS5, which belongs to the thrombospondin family. COMP / TSP5 contains 4 EGF-like domains, 1 TSP C-terminal (TSPC) domain, 8 TSP type-3 repeats. Abundantly expressed in the chondrocyte extracellular matrix, and is also found in bone, tendon, ligament and synovium and blood vessels. COMP may play a role in the structural integrity of cartilage via its interaction with other extracellular matrix proteins such as the collagens and fibronectin. COMP can mediate the interaction of chondrocytes with the cartilage extracellular matrix through interaction with cell surface integrin receptors. Thrombospondin-5 could play a role in the pathogenesis of osteoarthritis. COMP is a marker of cartilage turnover.

Galectin-3(LGALS3) is also known as Galactose-specific lectin 3, Mac-2 antigen, Carbohydrate-binding protein 35, Laminin-binding protein and Galactoside-binding protein. LGALS3 is highly expressed in early stages of papillary carcinoma, and lowly during tumor progression. The inflammasome is a multiprotein complex that mediates the activation of caspase-1, which promotes, amongst others, the secretion of the proinflammatory cytokines interleukin (IL)-1beta and IL-18. Frizzled proteins can activate canonical Wnt/beta-catenin signalling as well as non-canonical pathways (Wnt/Ca2+ pathways and planar cell polarity). The cysteine-rich domain (CRD) of the frizzled receptors situated on the extracellular part of the receptor is sufficient to bind the Wnt ligands.

Protein-Tyrosine Phosphatase 1C (PTP1C) belongs to the protein-tyrosine phosphatase family.which is known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. PTP1C is highly expressed in leukocyte cell type. It contains two SH2 domains and one tyrosine-protein phosphatase domain. The SH2 regions may interact with other cellular components to modulate its own phosphatase activity against interacting substrates.
