Acidic Fibroblast Growth Factor is a member of the fibroblast growth factor (FGF) family. FGF family members possess broad mitogenic and cell survival activities, and are involved in a variety of biological processes, including embryonic development, cell growth, morphogenesis, tissue repair, tumor growth and invasion. FIBP is an intracellular protein that binds selectively to acidic fibroblast growth factor (aFGF). It is postulated that FIBP may be involved in the mitogenic action of aFGF.

Mouse interleukin 13 (mIL-13) is a pleiotropic cytokine produced by activated Th2 cells. IL-13 induces B cell proliferation and immunoglobin production. It contains a four helical bundle with two internal disulfide bonds. Mouse IL13 shares 58% sequence identity with human protein and exhibits cross-species activity. IL13 signals via receptor IL13R (type2, IL4R) and activates STAT-6. IL13 initially binds IL-13R alpha1 with low affinity and triggers association of IL4R alpha, generating a high affinity heterodimeric receptor IL13R and eliciting downstream signals. IL13 also binds IL-13R alpha2 with high affinity, which plays a role in a negative feedback system of IL13 signaling. IL13 is an important mediator of allergic inflammation and disease.

Chemokine (C-X-C Motif) Ligand 2 (CXCL2) is a small secreted cytokine which belongs to the CXC chemokine family. CXCL2 is 90% identical in amino acid sequence as a related chemokine, CXCL1. CXCL2/MIP2-alpha is secreted by monocytes and macrophages and is chemotactic for polymorphonuclear leukocytes and hematopoietic stem cells. The gene for CXCL2 is located on human chromosome 4 in a cluster of other CXC chemokines. CXCL2 mobilizes cells by interacting with a cell surface chemokine receptor called CXCR2. CXCL2/MIP2-alpha has been known to regulate immune functions mainly by chemo-attracting neutrophils. It is produced by activated monocytes and neutrophils and expressed at sites of inflammation. CXCL2 is a hematoregulatory chemokine, which suppresses hematopoietic progenitor cell proliferation. CXCL2 can be induced by receptor activator of NF-kappaB ligand, the osteoclast (OC) differentiation factor, through JNK and NF-kappaB signaling pathways in OC precursor cells. CXCL2 in turn enhanced the proliferation of OC precursor cells of bone marrow-derived macrophages (BMMs) through the activation of ERK. Knockdown of CXCL2 inhibited both the proliferation of and the ERK activation in BMMs. During osteoclastogenesis CXCL2 stimulated the adhesion and the migration of BMMs. CXCL2 is a novel therapeutic target for inflammatory bone destructive diseases.

Human Chemokine (C-C Motif) Ligand 23 (CCL23) is a small cytokine belonging to the CC chemokine family. CCL23 is also known as myeloid progenitor inhibitory factor MPIF-1, CK8 and SCYA23. CCL23 cDNA encodes a 120 amino acid residue precursor protein with a putative 21 amino acid residue signal peptide that is cleaved to generate a 99 amino acid residue mature CCL23 (amino acids 22 -120). Additional N-terminal Processing of the 99 amino acid residue variant can generate a 75 amino acid residue peptide (amino acid 46-120) that is significantly more active than the 99 amino acid residue variant. CCL23 binds to CCR1 with high affinity and has chemotactic activity for monocytes, dendritic cells, and osteoclast precursors. CCL23 enhances angiogenesis of endothelial cells, but reduces the proliferation of progenitor cells giving rise to granulocyte and monocyte lineages.

Interleukin-33 (IL-33) was initially discovered as a nuclear factor NF-HEV abundantly expressed in high endothelial venules. It is a 30-32 kD pro-inflammatory protein with intracellular and extracellular activities and a chromatin-associated cytokine of the IL-1 family with high sequence and structural similarity to IL-1 and IL-18. IL-33 is highly and selectively expressed by high endothelial venule endothelial cells (HEVECs) in human tonsils, Peyers's patches, and lymph nodes. It contains a bipartite nuclear localization signal at the C-terminus, and is targeted to the nucleus when ectopically expressed in human umbilical vein endothelial cells (HUVECs) and HeLa cells. The C-terminal fragment, corresponding to mature IL-33, binds and triggers signaling. IL-33 mediates its biological effects via Toll-interleukin 1 (IL-1) receptor (TIR) domain-containing receptor ST2, activates NF-kappaB and MAP kinases, and drives production of T(H)2-associated cytokines from in vitro polarized T(H)2 cells. In vivo, IL-33 induces the expression of IL-4, IL-5, and IL-13 and leads to severe pathological changes in mucosal organs. Human IL-33 is 270 amino acids in length.

Proteoglycan 3, also known as Eosinophil major basic protein homolog, Prepro-major basic protein homolog, PRG3 and MBPH, contains one C-type lectin domain. Proteoglycans are a major component of the animal extracellular matrix. PRG3 localizes to the eosinophil secondary granule and is expressed in bone marrow, not detected in placenta. PRG3 has similar cytotoxic and cytostimulatory activities to PRG2/MBP. In vitro, PRG3 can stimulate neutrophil superoxide production and IL8 release, histamine and leukotriene C4 release from basophils.
