TESK2 is a nuclear protein that belongs to the protein kinase superfamily and is expressed in testis and prostate. Functioning as a dual-specificity protein kinase, TESK2 catalyzes the ATP-dependent phosphorylation of substrates and autophosphorylation on tyrosine and serine/threonine residues, thereby mediating intracellular signal transduction pathways. TESK2 requires magnesium as a cofactor and its catalytic activity is thought to play an important role in meiotic events such as spermatogenesis. TESK2 contains one protein kinase domain that is 65% identical to the kinase domain found in TESK1 (testicular protein kinase 1), suggesting a similar role for these proteins in phosphorylation events. Three isoforms of TESK2 are expressed due to alternative splicing.

Tom1L-1 is a 476 amino acid Golgi apparatus protein belonging to the TOM1 family and is a member of the multivesicular body (MVB) sorting machinery. Containing a GAT domain and a VHS domain, Tom1L-1 interacts with Fyn, GRB2, PI 3-kinase p85?and various signaling proteins when phosphorylated. GAT domain of Tom1L1 binds ubiquitin, suggesting participation in the sorting of ubiquitinated proteins into MVBs. Tom1L-1 may act as an adapter protein involved in signaling pathways and may promote Fyn activation, possibly by disrupting intramolecular SH3-dependent interactions. As an interactor and a substrate of Src tyrosine kinases (SFK), Tom1L1 is considered a novel mechanism involved in negative regulation of SFK mitogenic and transforming signals. Tom1L1 modulates SFK partitioning at the plasma membrane and downregulates Src kinases in an endosomal-dependent manner. It is suggested that Tom1L-1 functions as an anti-oncogene by inhibiting the formation of squamous cell carcinomas in skin.

Calcyclin, also known as Protein S100-A6, growth factor-inducible protein 2A9, S-100 calcium-binding protein A6 (S-100A6) or MLN 4, is a homodimeric member of the S-100 calcium-binding protein family whose expression is upregulated in proliferating and differentiating cells. Calcyclin is inducible by growth factors and overexpressed in acute myeloid leukemias. It is expressed in a cell-specific manner in subpopulations of neurons and astrocytes and in epithelial cells and fibroblasts. Calcyclin is a specific target of S-100B protein in vivo. The binding of Calcyclin to S-100B is stabilized by S-100B-bound calcium and zinc. Calcyclin associates with both Annexin XI and CacyBP (calcyclin-binding protein). It functions to activate several processes along the calcium signal transduction pathway including the regulation of cell growth, proliferation, secretion and exocytosis.

CSP contains a J-domain that binds HSP 70/HSC 70 chaperone ATPases and a membrane-targeting, palmitoylated cysteine-rich string region. CSPs may act as molecular chaperones in synapses, and mediate conformational folding of components of the vesicular exocytotic machinery. CSP is involved in the fine tuning of neurotransmission through its interaction with receptor-coupled trimeric GTP binding proteins (G proteins) and N-type Ca2+ channels. Two variants of CSP have been described: CSP1; and the 31 amino acid, C-terminally truncated isoform, CSP2. Subcellular fractionation of insulinoma cells shows CSP1 in granular fractions, while the membrane and cytosol fractions contain predominantly CSP2. The fractions also contain additional proteins, presumably CSP dimers. Furthermore, in various mammalian cell lines (including rat brain) CSP1 expression predominates CSP2 expression.

Synaptojanins exist as two isoforms, synaptojanin 1 and 2, which differ in the C-terminal domain, and each isoform has multiple variants produced by alternative splicing. Synaptojanin 1 is expressed as two major forms: the shorter is found in brain while the longer is expressed in peripheral tissues. Eight splice variants of synaptojanin 2 have been detected, including a brain specific isoform. Synaptojanins are thought to participate in the endocytosis of synaptic vesicles and the regulation of the actin cytoskeleton.
