Omschrijving:
This gene is a member of a gene family that encodes enzymes involved in both the metabolism of glutathione and in the transpeptidation of amino acids. Changes in the activity of gamma-glutamyltransferase may signal preneoplastic or toxic conditions in the liver or kidney. The protein encoded by this gene consists of a heavy and a light chain, and it can interact with CT120, a plasma membrane-associated protein that is possibly involved in lung carcinogenesis. Omschrijving:
Chromosome 1 is the largest human chromosome spanning about 260 million base pairs and making up 8% of the human genome. There are about 3,000 genes on chromosome 1, and considering the great number of genes there are also a large number of diseases associated with chromosome 1. Notably, the rare aging disease Hutchinson-Gilford progeria is associated with the LMNA gene which encodes lamin A. When defective, the LMNA gene product can build up in the nucleus and cause characteristic nuclear blebs. The mechanism of rapidly enhanced aging is unclear and is a topic of continuing exploration. The MUTYH gene is located on chromosome 1 and is partially responsible for familial adenomatous polyposis. Stickler syndrome, Parkinsons, Gaucher disease and Usher syndrome are also associated with chromosome 1. A breakpoint has been identified in 1q which disrupts the DISC1 gene and is linked to schizophrenia. Aberrations in chromosome 1 are found in a variety of cancers including head and neck cancer, malignant melanoma and multiple myeloma. The C1orf111 gene product has been provisionally designated C1orf111 pending further characterization.

Omschrijving:
Regulator of histone methyltransferase complexes that plays an essential role in embryonic development, including heart and liver development, neural tube fusion process and hematopoiesis. Acts by modulating histone methyltransferase activity and promoting the recruitment of histone methyltransferase complexes to their target genes. Binds DNA and mediates the recruitment of the PRC2 complex to target genes in embryonic stem cells. Does not have histone demethylase activity but regulates activity of various histone methyltransferase complexes. In embryonic stem cells, it associates with the PRC2 complex and inhibits trimethylation of 'Lys-27' of histone H3 (H3K27me3) by the PRC2 complex, thereby playing a key role in differentiation of embryonic stem cells and normal development. In cardiac cells, it is required to repress expression of cyclin-D1 (CCND1) by activating methylation of 'Lys-9' of histone H3 (H3K9me) by the GLP1/EHMT1 and G9a/EHMT2 histone methyltransferases. Also acts as a transcriptional repressor of ANF via its interaction with GATA4 and NKX2-5. Participates in the negative regulation of cell proliferation signaling.

HAP1 (huntingtin-associated protein 1) binds to huntingtin (1). Huntingtin is a protein that contains a polyglutamine region and when the number of glutamine repeats exceeds 35, the gene encodes a version of huntingtin that leads to Huntington’s disease (HD) (2,3). The ability of HAP1 to bind to huntingtin is enhanced by an expanded polyglutamine repeat region (1). HAP1 shows neuronal localization and moves with huntingtin in nerve fibers (4,5). HAP1 is primarily expressed in brain tissue, with greater expression in the olfactory bulb and brain stem (1). Mouse HAP1 is localized to membrane-bound organelles including large endosomes, tubulovesicular structures and budding vesicles in neurons (6). Duo, also designated huntingtin-associated protein interacting protein or HAPIP, binds Huntingtin-associated protein 1 (HAP1) and may have a role in vesicle trafficking and cytoskeletal function.

Albumin is a soluble, monomeric protein which comprises about one-half of the blood serum protein. Albumin functions primarily as a carrier protein for steroids, fatty acids, and thyroid hormones and plays a role in stabilising extracellular fluid volume. Mutations in this gene on chromosome 4 result in various anomalous proteins. Albumin is a globular unglycosylated serum protein of molecular weight 65000. The human albumin gene is 16961 nucleotides long from the putative & apos; cap & apos; site to the first poly(A) addition site. It is split into 15 exons which are symmetrically placed within the 3 domains that are thought to have arisen by triplication of a single primordial domain. Albumin is synthesised in the liver as preproalbumin which has an N-terminal peptide that is removed before the nascent protein is released from the rough endoplasmic reticulum. The product, proalbumin, is in turn cleaved in the Golgi vesicles to produce the secreted albumin.

COPA (alpha-coat protein) is processed to produce Xenin. Xenin stimulates exocrine pancreatic secretion to affect small and large intestinal motility, and inhibits pentagastrin-stimulated secretion of acid. In the gut, Xenin interacts with the neurotensin receptor. Membrane and vesicular trafficking in the early secretory pathway are mediated by non-Clathrin COP (coat protein) I-coated vesicles. COPI-coated vesicles mediate retrograde transport from the Golgi back to the ER and intra-Golgi transport. The cytosolic precursor of the COPI coat, the heptameric coatomer complex, is composed of two subcomplexes. The first consists of the COPB, COPG, COPD and COPZ subunits (also known as b-, g-, d- and z-COP, respectively), which are distantly homologous to AP Clathrin adaptor subunits. The second consists of the COPA, b'-COP and COPE subunits (also known as a-COP, COPP and e-COP, respectively).

Omschrijving:
This gene encodes an enzyme belonging to the protein tyrosine kinase family, and it plays a role in T-cell development and lymphocyte activation. This enzyme, which is phosphorylated on tyrosine residues upon T-cell antigen receptor (TCR) stimulation, functions in the initial step of TCR-mediated signal transduction in combination with the Src family kinases, Lck and Fyn. This enzyme is also essential for thymocyte development. Mutations in this gene cause selective T-cell defect, a severe combined immunodeficiency disease characterised by a selective absence of CD8-positive T-cells. Two transcript variants that encode different isoforms have been found for this gene.

Guanine nucleotide-binding proteins (G proteins) are involved as a modulator or transducer in various transmembrane signaling systems. The beta and gamma chains are required for the GTPase activity, for replacement of GDP by GTP, and for G protein-effector interaction. Plays a role in the regulation of adenylyl cyclase signaling in certain regions of the brain. Plays a role in the formation or stabilzation of a G protein heterotrimer (G(olf) subunit alpha-beta-gamma-7) that is required for adenylyl cyclase activity in the striatum.
