Molecular cloning and analysis of several PSG genes has indicated that the PSGs form a subgroup of the carcinoembryonic antigen (CEA) gene family, Members of the CEA family consist of a single N domain, with structural similarity to the immunoglobulin variable domains, followed by a variable number of immunoglobulin constant-like A and/or B domains.

Glioma Pathogenesis-Related Protein 1 (GLIPR1) is a member of the CRISP family. It is a single-pass membrane protein with 266 amino acids. GLIPR1 is highly expressed in the lung and kidney and lowly expressed in the heart and liver. It is also highly expressed in cell lines that are derived from nervous system tumors arising from glia; conversely, it is lowly expressed in non-glial-derived nervous system tumor cell lines. GLIPR1 is only expressed in brain tumor glioblastoma multiforme/astrocytoma and not expressed in other nervous system tumors nor normal adult or fetal tissues.

Description:
LIG4 encodes a DNA ligase that joins single-strand breaks in a double-stranded polydeoxynucleotide in an ATP-dependent reaction. This protein is essential for V (D)J recombination and DNA double-strand break (DSB) repair through nonhomologous end joining (NHEJ). This protein forms a complex with the X-ray repair cross complementing protein 4 (XRCC4), and further interacts with the DNA-dependent protein kinase (DNA-PK). Both XRCC4 and DNA-PK are known to be required for NHEJ. The crystal structure of the complex formed by this protein and XRCC4 has been resolved. Defects in this gene are the cause of LIG4 syndrome.

DPP3 has post-proline dipeptidyl aminopeptidase activity, cleaving Xaa-Pro dipeptides from the N-termini of proteins. Cadherin-8 (CDH8) is a type II classical cadherin from the cadherin superfamily. Member of the Cadherin superfamily are integral membrane proteins that mediate calcium-dependent cell-cell adhesion. Cadherin proteins are composed of a large N-terminal extracellular domain, a single membrane-spanning domain, and a small highly conserved C-terminal cytoplasmic domain. Cadherins are calcium dependent cell adhesion proteins. They preferentially interact with themselves in a homophilic manner in connecting cells. The extracellular domain of CDH8 contains five cadherin domains. CDH8 is expressed in brain and is putatively involved in synaptic adhesion, axon outgrowth and guidance.

Neuroligin 4, X-Linked (NLGN4X) is a single-pass type I membrane protein that belongs to the type-B carboxylesterase/lipase family. NLGN4X is detected at higher levels in heart and at lower levels in the liver, skeletal muscle, and pancreas. NLGN4X is a putative neuronal cell surface protein involved in cell-cell-interactions. NLGN4X may act as splice site-specific ligands for beta -neurexins. It has been shown that NLGN4X is involved in the formation and remodeling of central nervous system synapses. NLGN4X also interacts with discs, large (Drosophila) homolog 4 (DLG4). Defects in NLGN4X have been associated with autism and Asperger syndrome.

CD83 is expressed by activated lymphocytes, Langerhans cells and interdigitating reticulum cells. It contains one Ig-like V-type (immunoglobulin-like) domain, the soluble CD83 has the opposite effect and has an immune inhibitory capacity. Due to its immune inhibitory function, CD83 may play a significant role in antigen presentation or the cellular interactions that follow lymphocyte activation.
