Description:
B- and T-Lymphocyte Attenuator (BTLA) is a single-pass type I membrane protein containing 1 Ig-like V-type (immunoglobulin-like) domain. BTLA expression is induced during activation of T cells, and BTLA remains expressed on Th1 cells but not Th2 cells. Like PD1 and CTLA4, BTLA interacts with a B7 homolog, B7H4. However, unlike PD-1 and CTLA-4, BTLA displays T-Cell inhibition via interaction with tumor necrosis family receptors (TNF-R), not just the B7 family of cell surface receptors. BTLA is a lymphocyte inhibitory receptor that inhibits lymphocytes during immune response. BTLA also is a ligand for tumor necrosis factor (receptor) superfamily, member 14 (TNFRSF14), also known as herpes virus entry mediator (HVEM). BTLA-HVEM complexes negatively regulate T-cell immune responses.

EGF is a single-pass type I membrane protein,containing 8 LDL-receptor class B repeats and 9 EGF-like domains. EGF results in cellular proliferation, differentiation, and survival.EGF is a low-molecular-weight polypeptide first purified from the mouse submandibular gland, but since then found in many human tissues including submandibular gland, parotid gland. Salivary EGF, which seems also regulated by dietary inorganic iodine, also plays an important physiological role in the maintenance of oro-esophageal and gastric tissue integrity. The biological effects of salivary EGF include healing of oral and gastroesophageal ulcers, inhibition of gastric acid secretion, stimulation of DNA synthesis as well as mucosal protection from intraluminal injurious factors such as gastric acid, bile acids, pepsin, and trypsin and to physical, chemical and bacterial agents.

Programmed cell death protein 1(PDCD1) is a single-pass type I membrane protein and contains 1 Ig-like V-type domain. PD-1 is a member of the extended CD28/CTLA-4 family of T cell regulators. PDCD1 inhibits the T-cell proliferation and production of related cytokines including IL-1, IL-4, IL-10 and IFN- gamma by suppressing the activation and transduction of PI3K/AKT pathway. In addition, coligation of PDCD1 inhibits BCR-mediating signal by dephosphorylating key signal transducer. PDCD1 has been suggested to be involved in lymphocyte clonal selection and peripheral tolerance, and thus contributes to the prevention of autoimmune diseases. As a cell surface molecule, PDCD1 regulates the adaptive immune response. Engagement of PD-1 by its ligands PD-L1 or PD-L2 transduces a signal that inhibits T-cell proliferation, cytokine production, and cytolytic function.

The OKT4 monoclonal antibody specifically binds to the CD4 receptor for the human immunodeficiency virus (HIV). CD4 is a 59 kDa single-chain transmembrane glycoprotein that expressed on the surface of most of the thymocytes, T-helper cells, and in low levels on monocytes and macrophages. CD4 is a co-receptor in the antigen-induced T cell activation (together with the MHC class II). Recognises an 85-115 kDa protein (variation with cell type), identified as intercellular adhesion molecule (ICAM-1). It has 7 potential N-linked glycosylation sites. ICAM1 is a single chain glycoprotein of Ig supergene family, present on unstimulated endothelial cells (EC) and on a variety of other cell types including activated fibroblasts, EC, macrophages, and lymphocytes. ICAM1 mediates cell adhesion by binding to integrins CD11a/CD18 (leukocyte adhesion molecule, LFA-1) and to CD11b/CD18 (Mac-1). This interaction enhances antigen-specific T-cell activation. ICAM1 also binds to CD43 and to Plasmodium falciparum infected RBCs and may also be related to progression and metastasis of tumors.

RPA Interacting Protein Antibody: Replication protein A (RPA) is a single-stranded-DNA binding protein involved in numerous eukaryotic DNA processes including replication, repair and recombination. RPA interacting protein (RPA IP) has been identified as an adapter protein that is involved in RPA nuclear import instead of the prototypical importin proteins that normally mediate nuclear import. Multiple isoforms of RPA IP are known to exist, with the longest isoform localized to the cytoplasm. Isoform 2 is sumoylated and is located in the PML nuclear body within the nucleus. It has been suggested that this isoform mediates the localization of the RPA complex into the PML nuclear body, thereby participating in RPA function in DNA metabolism.

Nicotinic acetylcholine receptors (nAChRs) are ligand-gated ion channels formed by a pentameric arrangement of alpha and beta subunits to create distinct muscle and neuronal receptors. Neuronal receptors are found throughout the peripheral and central nervous system where they are involved in fast synaptic transmission. This gene encodes an alpha subunit that is widely expressed in the brain. The proposed structure for nAChR subunits is a conserved N-terminal extracellular domain followed by three conserved transmembrane domains, a variable cytoplasmic loop, a fourth conserved transmembrane domain, and a short C-terminal extracellular region. Mutations in this gene cause autosomal dominant nocturnal frontal lobe epilepsy type 4. Single nucleotide polymorphisms (SNPs) in this gene have been associated with nicotine dependence.

KCTD15 Antibody: Childhood and adult obesity in the United States and to a lesser extent the rest of the world has increased dramatically over the past decade. Both environmental and genetic factors are involved in the onset and progression of weight gain. Recently, the potassium channel KCTD15 was identified as a genetic loci associated with higher than normal body mass index (BMI) in humans along with genes such as GNPDA2, MTCH2, FTO, and TMEM18. Further studies on single nucleotide polymorphisms (SNPs) in non-diabetic and diabetic patients showed that FTO was most strongly associated with obesity while MTCH2 and GNPDA2 were still significantly associated with higher than normal BMI levels. At least two isoforms of KCTD15 are known to exist.

Description:
This antibody recognises a single chain, transmembrane, heavily glycosylated protein of 90 to 120 kDa, which is identified as CD34. Its expression is a hallmark for identifying pluripotent hematopoietic stem or progenitor cells. Its expression is gradually lost as lineage committed progenitors differentiate. CD34 is a marker of choice for staining blasts in acute myeloid leukemia. In addition, it is expressed by soft tissue tumors, such as solitary fibrous tumor and gastrointestinal stromal tumor. CD34 expression is also found in vascular endothelium. Additionally, it appears that proliferating endothelial cells overexpress this molecule than the non-proliferating endothelial cells. Anti-CD34 labels > 85% of angiosarcoma and Kaposi’s sarcoma, but shows low specificity.

Akirin1 Antibody: The highly conserved, nuclear-localized Akirin1 and Akirin2 proteins critically regulate the transcription of NF-kappa B-dependent genes and are required for defense against Gram-negative bacteria in the immune deficiency and NF-kappa B pathways. Akirin1 is dispensable in the mouse, and neither knockout mice nor cells derived from them have obvious distinctive phenotypes. In contrast, Akirin2 is required for development in the mouse and knockout of both Akirin homologs in mice show that Akirin2 is required downstream of toll-like receptor (TLR), TNF-alpha and IL-1 beta signaling, and for the production of IL-6. Akirin2 is functionally closer to the single gene in Drosophila, as the homozygous null D. melanogaster Akirin mutants show a similar, mid-to-early embryonic death.

This antibody recognises an epitope encoded by exon v9 on the variant portion of human CD44. The CD44 molecule belongs to a family of cellular adhesion molecules found on a wide range of normal and malignant cells in epithelial, mesothelial and hemopoiesis tissues. CD44 is a single gene with 20 exons, of which 10 are normally expressed to encode the basic CD44 (H-CAM) molecule. The additional 10 exons (v1 to v10) are only expressed by alternative splicing of the nuclear RNA. The expression of specific cell adhesion molecule CD44 splice variants has been reported to be associated with metastasis in certain human malignancies.
