Description:
High Mobility Group Protein B2 (HMGB2) belongs to the non-histone chromosomal high-mobility group protein family. Members of this family are chromatin-associated and widely spread in the nucleus of higher eukaryotic cells. HMGB2 contains 2 HMG box DNA-binding domains. It is associated with chromatin and has the ability to bend DNA, preferentially single-stranded DNA. It is shown that HMGB2 is able to efficiently bend DNA and form DNA circles. In addition, HMGB2 is involved in the final ligation step in DNA end-joining processes of DNA double-strand breaks repair and V(D)J recombination.

CD300C is a single-pass type I membrane protein which belongs to the immunoregulatory signaling (IRS) family. CD300C contains one Ig-like V-type domain and is present on the surface of natural killer cells, granulocytes, most myeloid cells, dendritic cells, and a subpopulation of T and B lymphocytes. The CD300C (CMRF-35A) and CD300A (CMRF-35H) molecules are homologous leukocyte surface proteins. CD300a and CD300C play an important role in the cross-regulation of TNF-alpha and IFN-alpha secretion from pDCs. CD300A and CD300C are indistinguishable on the surface of NK cells. The ligand for CD300C is presently unknown.

The B- and T-lymphocyte attenuator (BTLA) protein is a member of the immunoglobulin superfamily containing a single immunoglobulin (Ig) domain. Like other coihibitory receptors such as CTLA-4 and PD-1, BTLA functions as a receptor that relays inhibitory signals to suppress the immune response, and can inhibit LPS-induced endotoxic shock by suppressing TLR4 signaling in innate immune cells (1,2). Polymorphisms in this gene have also been associated with an increased risk of rheumatoid arthritis.

RBMS3 is a member of a small family of proteins which bind single stranded DNA/RNA. These proteins are characterized by the presence of two sets of ribonucleoprotein consensus sequence (RNP-CS) that contain conserved motifs, RNP1 and RNP2, originally described in RNA binding proteins, and required for DNA binding. These proteins have been implicated in such diverse functions as DNA replication, gene transcription, cell cycle progression and apoptosis.The protein encoded by this gene is a member of a small family of proteins which bind single stranded DNA/RNA. These proteins are characterized by the presence of two sets of ribonucleoprotein consensus sequence (RNP-CS) that contain conserved motifs, RNP1 and RNP2, originally described in RNA binding proteins, and required for DNA binding. These proteins have been implicated in such diverse functions as DNA replication, gene transcription, cell cycle progression and apoptosis. The encoded protein was isolated by virtue of its binding to an upstream element of the alpha2 (I) collagen promoter. The observation that this protein localizes mostly in the cytoplasm suggests that it may be involved in a cytoplasmic function such as controlling RNA metabolism, rather than transcription. Multiple alternatively spliced transcript variants encoding different isoforms have been found for this gene.

Cytotoxic T-lymphocyte protein 4, is a single-pass type I membrane protein. It is widely expressed with highest levels in lymphoid tissues. CD28 and CTLA-4, together with their ligands, B7-1 and B7-2, constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD28 and CTLA-4 are structurally homologous molecules that are members of the immunoglobulin (Ig) gene superfamily. CTLA4 transmits an inhibitory signal to T cells, whereas CD28 transmits a stimulatory signal. Intracellular CTLA4 is also found in regulatory T Cells and may play an important role in their functions. Tcell activation through the Tcell receptor and CD28 leads to increased expression of CTLA4.

TNFRSF10B is a member of the TNF-receptor superfamily, and contains an intracellular death domain. This receptor can be activated by tumor necrosis factor-related apoptosis inducing ligand (TNFSF10/TRAIL/APO-2L), and transduces apoptosis signal. The adapter molecule FADD recruits caspase-8 to the activated receptor and is required for the apoptosis mediated by TNFRSF10B. TNFRSF10B is expressed in a number of cell types, and to particularly high levels in lymphocytes and spleen. This single-pass transmembrane protein contains two cysteine-rich repeat units in its extracellular region, followed by a transmembrane segment and a cytoplasmic tail containing a typical “death domain”. TNFRSF10B expression is regulated by the tumor suppressor p53. It is also indicated that the activation of NF-kappa-B can be promoted by TNFRSF10B.

Junctional Adhesion Molecule B (JAM-B) is a single-pass type I membrane protein that belongs to the juctional adhesion molecules family. JAM-B includes a signal sequence (aa 1-28), an extracellular region (aa 29-238) with one Ig-like C2-type domain and one Ig-like V-type domain, a transmembrane segment (aa 239-259), and a cytoplasmic domain (aa 260 - 298). JAMB is localized to the tight junctions between endothelial cells or epithelial cells. JAM-B is prominently expressed in the heart, placenta, lung, foreskin and lymph node.
