The OKT4 monoclonal antibody specifically binds to the CD4 receptor for the human immunodeficiency virus (HIV). CD4 is a 59 kDa single-chain transmembrane glycoprotein that expressed on the surface of most of the thymocytes, T-helper cells, and in low levels on monocytes and macrophages. CD4 is a co-receptor in the antigen-induced T cell activation (together with the MHC class II). The OKT4 and the RPA-T4 monoclonal antibodies recognize different epitopes of CD4 and they do not exhibit cross-block binding.

IRS-1 Antibody: Following tyrosine phosphorylation, the insulin receptor substrate 1 and 2 (IRS-1 and IRS-2) can form a protein scaffolding for the assembly of a host of Src homology 2 (SH2) domain-containing proteins. IRS-1 tyrosine phosphorylation can occur through the activity of several cytokine and growth factor receptors such as interleukin (IL)-4, IL-9, interferon-gamma, in addition to the insulin and insulin-like growth factor 1 receptors. The scaffolding provided by IRS-1 and IRS-2 is necessary for insulin signal transduction across the cell membrane. IRS-1 tyrosine phosphorylation, and thus formation of the IRS scaffolding is inhibited by tumor necrosis factor (TNF), and this inhibition can itself be blocked by rapamycin, an inhibitor of the mammalian target of rapamycin (TOR). TNF activity could also be blocked by inhibition of the Akt kinase and the PTEN tumor suppressor, suggesting that TNF impairs insulin signaling through IRS-1 by activation of the TOR signaling pathway.

The RDR5 monoclonal antibody specifically reacts with human CD127, a 60-90 kDa type I transmembrane glycoprotein also known as the IL-7 receptor alpha chain. CD127 binds with CD132 (gamma c) to forms the IL-7 receptor that is expressed on mature T cells and immature B cells. Since CD127 expression is downregulated in Treg cells, it can be used as a marker for differentiating them from common T cells. The binding of the RDR5 antibody can be blocked by pre-incubating the cells with human IL-7.

AHSG is an important circulating inhibitor of calcification in vivo, and is downregulated during the acute-phase response. BATF Antibody: BATF is a nuclear basic leucine zipper protein that belongs to the AP-1/ATF superfamily of transcription factors. The leucine zipper of this protein mediates dimerization with members of the Jun family of proteins. This protein is thought to be a negative regulator of AP-1/ATF transcriptional events and blocks cellular transformation by Ras and Fos. BATF also is required for the differentiation of IL-17-producing T helper (TH17) cells, a CD4+ subset of T cells that coordinates the inflammatory response in host defense. In both T cells and B cells, BATF is required for the appropriate regulation of the class-switch recombination.

CCL4, (C-C motif chemokine 4; macrophage inflammatory protein 1beta) is a chemokine secreted at sites of inflammation by activated leukocytes, lymphocytes, vascular endothelial cells and pulmonary smooth cells. CCL4 attracts a variety of immune cells to sites of microbial infection as well as to pathologic inflammation, such as allergic asthma and ischemic myocardium. Full-length and truncated CCL4 exert biological activity through the receptor CCR5. The truncated form of CCL4 interacts additionally with CCR1 and CCR2b. The ability of CCL4 to bind CCR5 inhibits the cellular entry of M-tropic HIV. Both CCL4 forms block HIV-1 infection of T cells by inducing the downregulation of CCR5.
