3- (4,5-Dimethyl-2-thiazolyl-2)-2,5-diphenyl-tetrazolium bromide is used as a colorimetric metabolic activity indicator in cell viability assays. Thiazolyl Blue Tetrazolium Blue (MTT) is a dye used in measurement of cell proliferation. MTT produces a yellowish solution that is converted to dark blue, water-insoluble MTT formazan by mitochondrial dehydrogenases of living cells. The blue crystals are solubilized with acidified isopropanol and the intensity is measured colorimetrically at 570 nm. MTT has been used as a histochemical/cytochemical reagent and for the detection of NAD. ADP-linked enzyme systems in tissue cannot be detected with MTT, due to binding of the cation by the cyanide trap used. MTT is rapidly reduced to the formazan, which chelates with nickel, copper, and cobalt; the cobalt chelate has been used in oxidative systems.

Description:   Serpin A5 is a member of the human Serpin superfamily consists of at least 35 members. It is synthesized in the liver and has been detected in saliva, cerebral spinal fluid, amniotic fluid, tears and semen. As a potent inhibitor of the protein C anticoagulant pathway at the levels of both zymogen activation and enzyme inhibition, Serpin A5 additionally inhibits a variety of serine protease including thrombin, factor Xa, several kallekreins and acrosin. It plays a critical role in the processes of blood of blood coagulation and fertilization. Serpin A5 also inhibits urinary plasminogen activator (uPA), a mediator of tumor cell invasion, and regulates tumor growth and metastasis by inhibiting angiogenesis. Furthermore, recent studies have identified PCI as a potent and direct inhibitor of activated HGFA (hepatocyte growth factor activator), suggesting a novel function in the regulation of tissue repair and regeneration. Similar to Serpins C1 and D1, the thrombin inhibitory activity of serpinA5 is enhanced by heparin.

SCGN contains six EF-hand domains, related to calbindin D-28K and calretinin. Description:   JPH4 Antibody: Junctional complexes between the plasma membrane (PM) and endoplasmic/sarcoplasmic reticulum (ER/SR) are a common feature of all excitable cell types and mediate cross talk between cell surface and intracellular ion channels. Junctophilins (JPs) are important components of the junctional complexes. JPs are composed of a carboxy-terminal hydrophobic segment spanning the ER/SR membrane and a remaining cytoplasmic domain that shows specific affinity for the PM. Four JPs have been identified as tissue-specific subtypes derived from different genes: JPH1 is expressed in skeletal muscle, JPH2 is detected throughout all muscle cell types, and JPH3 and JPH4 are predominantly expressed in the brain. In the CNS, both JPH3 and JPH4 are expressed throughout neural sites and contribute to the subsurface cistern formation in neurons. Mice lacking both JPH3 and JPH4 subtypes exhibit serious symptoms such as impaired learning and memory and are accompanied by abnormal nervous functions.

Description:   The antibody HEB-29 reacts with human blood group B. The specificity of the antibody HEB-29 was confirmed by comparison of specificity and reactivity to standard reagent using >5,000 samples of blood. mAb HEB-29 shows specific staining of erythrocytes and vascular epithelium of blood group B controls and no staining in group A controls. It is applicable for tissue staining in tumor patients with blood groups B and AB. Blood group antigens are generally defined as molecules formed by sequential addition of saccharides to the carbohydrate side chains of lipids and proteins detected on erythrocytes and certain epithelial cells. The A, B and H antigens are reported to undergo modulation during malignant cellular transformation. Blood group related antigens represent a group of carbohydrate determinants carried on both glycolipids and glycoproteins. They are usually mucin type, and are detected on erythrocytes, certain epithelial cells, and in secretions of certain individuals. Sixteen genetically and biosynthetically distinct but inter related specificities belong to this group of antigens, including A, B, H, Lewis A, Lewis B, Lewis X, Lewis Y, and precursor type 1 chain antigens.

Description:   Nop30 Antibody: Apoptosis, also known as programmed cell death, plays major roles in development and normal tissue turnover in addition to tumor formation. Apoptosis is regulated by death domain (DD) and/or caspase recruitment domain (CARD) containing molecules and the caspase family of proteases. CARD domain containing cell death regulators include RAIDD, Apaf-1, caspase-9, and caspase-2. A novel CARD domain containing protein was recently identified and designated ARC for apoptosis repressor with CARD. An alternate splicing isoform of ARC was identified as Nop30. While ARC interacts with caspase-2 and -8 and suppresses apoptosis induced by cell death adapters FADD and TRADD and by cell death receptors Fas, TNFR-1 and DR3, Nop30 multimerizes and binds to the splicing factor SRp30c and may act to influence alternative splice site selection in vivo. The Nop30 antibody will not detect ARC protein.

Description:   The 2F1 monoclonal antibody is specific for the mouse Killer cell Lectin-like Receptor G1 (KLRG1), a homodimer consisting of two N-glycosylated subunits of 30-38 kDa, also known as MAFA (Mast cell Function-associated Antigen). The antigen contains a cytoplasmic motif similar to ITIM (the immunoreceptor tyrosine-based inhibitory motif). KLRG1 is a receptor for cadherin, a family of transmembrane glycoproteins that mediate cell adhesion, and a common marker of T cell senescence. The receptor is believed to play an important role in the innate and adaptive immune system through the regulation of leukocytes. It is expressed on lymphokine-activated killer (LAK) cells, adherent LAK (A-LAK) cells, a sub-set of natural killer (NK) cells, T cells. In NK cells, it inhibits cytokine production and cytotoxicity activity.The receptor expression was not detected on the mouse peritoneal mast cells, or bone marrow mast cells.

Description:   KappaB ras Antibody: KappaB ras-1 ( kappa B-ras-1) and kappaB-ras-2 are two small proteins that similar to Ras-like small GTPases that associate with I kappa B (I kappa B), an inhibitor of the transcription factor NF-kappa B. I kappa B exists in two homologous forms, I kappa B-alpha and I kappa B-beta, although I kappa B-beta contains a unique 47-amino acid region within its ankyrin domain. While inactive I kappa B-alpha-NF-kappa B complexes can shuttle in and out of the nucleus, I kappa B-beta-NF-kappa B complexes are retained exclusively in the cytoplasm. It is suggested that kappaB-ras proteins preferentially bind to the I kappa B-beta form through this unique insert within the ankyrin region, thus modulating the cellular location of I kappa B-beta and regulating the rate of degradation of I kappa B-beta. This antibody detects both kappaB-ras1 and kappaB-ras2.
