Description:
The M1/70 monoclonal antibody specifically reacts with the 170 kDa alpha M integrin chain of mouse CD11b from the Mac-1 integrin (CD11b/CD18). Mac-1 binds to C3bi, CD54 (ICAM-1), and fibrinogen, and it is expressed by granulocytes, macrophages, NK cells, myeloid-derived dendritic cells, microglia, activated lymphocytes, and mouse B-1 cells. The expression is up-regulated on activated neutrophils at the same time that L-selectin is shed from the cell surface. The M1/70 antibody is used for the detection of monocytes, granulocytes, and a subset of natural killer cells in human peripheral blood.M1/70 blocks C3bi binding and cell adherence, but not cell-mediated lysis and it cross-reacts with human CD11b.

Description:
Ractopamine is a phenolethanolamine beta-adrenoceptor agonist that is used for the improvement of weight gain, carcass leanness and feed efficiency in pigs. It has been known for years that ractopamine enhances growth in meat animals by repartitioning nutrients. This means that the nutrients that the animal consumes in its feed are shifted away from being stored as fat and are instead used in lean tissue production. This shift occurs as the animal reaches the top of its growth curve and lean tissue production drops off and fat deposition increases. Fat levels also appear to decrease, due to an increase in lypolysis (fat breakdown). Using the ractopamine monoclonal antibody, it is possible to detect ractopamine and other agonists in urine, muscle and liver both rapidly and with accuracy.

Recognises a protein of 40 kDa, identified as cytokeratin-19 (CK19), which is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. Anti-CK19 is a useful marker for detection of tumor cells in lymph nodes, peripheral blood, bone marrow and breast cancer.

This antibody recognises proteins in MW range of 265-400 kDa, identified as different glycoforms of MUC1 (Mucin-1) or EMA (epithelial membrane antigen). The alpha subunit has cell adhesive properties. It can act both as an adhesion and an anti-adhesion protein. MUC1/Mucin-1 may provide a protective layer on epithelial cells against bacterial and enzyme attack. The beta subunit contains a C-terminal domain, which is involved in cell signaling through phosphorylations and protein-protein interactions. In immunohistochemical assays, the MUC1/EMA antibody superbly stains routine formalin/paraffin carcinoma tissues. MUC1 antibody is useful as a pan-epithelial marker for detecting early metastatic loci of carcinoma in bone marrow or liver.

This antibody is specific to Complement 4d (C4d) and it reacts with the secreted as well as cell-bound protein. C4d is a degradation product of the activated complement factor C4b. Complement 4b is typically activated by binding of antibodies to specific target molecules. Following activation and degradation of the C4 molecule, thio-ester groups are exposed, which allow transient, covalent binding of the degradation product C4d to endothelial cell surfaces and extracellular matrix components of vascular basement membranes near the sites of C4 activation. The presence of C4d in peritubular capillaries is a key indicator for acute humoral (i.e. antibody-mediated) rejection of kidney, heart, pancreas and lung allografts. As an established marker of antibody-mediated acute renal allograft rejection and its proclivity for endothelium, this component can be detected in peritubular capillaries in chronic renal allograft rejection as well as hyperacute rejection, acute vascular rejection, acute cellular rejection, and borderline rejection. C4d has been shown to be a significant predictor of transplant kidney graft survival. C4d antibody, combined with antibody to C3d, can be utilized as a tool for diagnosis of allograft rejection that may warrant a prompt and aggressive anti-rejection treatment.

MEK6 is a member of MAPKK protein kinase family. By using degenerate oligonucleotide primers from the conserved kinase domains of MKK3 and MKK4 two human cDNAs and 1 murine cDNA encoding closely related proteins of the MKK family were cloned. The two human clones appear to be different isoforms of the same gene generated by differential splicing: the shorter clone was designated MKK6, encodes a 278-amino acid protein, while the longer clone, designated MKK6b, encodes a 334-amino acid protein. MKK6 is about 80% identical to MKK3 and 40% identical to MKK4. 1.7-kb human MKK6 transcript is highly expressed in skeletal muscle, while an MKK6b-specific probe detected mRNA bands of 1.8, 2.4, and 4.5 kb that are enriched in heart, skeletal muscle, pancreas and liver. MKK6 plays an important role in intracellular signaling pathways leading toward activation of the p38 MAP kinase. MEK6 phosphorylates and activates p38 in response to inflammatory cytokines or environmental stress. As an essential component of p38 MAPK mediated signal transduction pathway, this gene is involved in many cellular processes such as stress induced cell cycle arrest, transcription activation and apoptosis.

Cadherin-6 (CDH6) is a type-II classic cadherin cell-cell adhesion molecules, which are expressed in graded or areal patterns, as well as layer-specific patterns, in the cortical plate. Human Cadherin-6 is synthesized as a 790 aa type I transmembrane glycoprotein that contains a 18 aa signal peptide, a 35 aa propeptide, a 562 aa extracellular region, a 21 aa transmembrane segment, and a 154 aa cytoplasmic domain. There are five cadherin domains of approximately 110 aa each in the extracellular region. Cadherin-6 has high expression in kidney, brain, and cerebellum, and may contribute to the formation of the segmental structure of the early brain, as well as the development of renal proximal tubules. Weak expression is also detected in lung, pancreas, gastric mucosa and cytotrophoblasts. As a classic cadherin, Cadherin-6 will form homodimers and promote intercellular adhesion with itself and, possibly, Cadherin-9 and -14.

Recognises a protein of 42-46 kDa, identified as MAGE-1. This mAb does not cross-react with MAGE-2, -3, -4, -6 -9, -10, -or -12 protein. Human malignant neoplasms carry rejection antigens that are recognized by the patients' autologous, tumor directed and specific, cytolytic, CD8+ T lymphocyte clones (CTL). The MAGE family of genes codes an important group of antigens. It was identified that melanomas and primary glial brain tumors express common melanoma associated antigens (MAAs). Because MAGE-1 is expressed on a significant proportion of human neoplasms of various histological types (melanoma, brain tumors of glial origin, neuroblastoma, non-small cell lung cancer, breast, gastric, colorectal, ovarian, renal cell carcinomas) and not on normal tissues, the encoded antigen may serve as a marker of early detection and target for cancer immunotherapy.

LRFN5 Antibody: LRFN5 is one of a family of five transmembrane glycoproteins that are highly expressed in neuronal tissues. LRFN proteins share leucine-rich repeat (LRR)-immunoglobulin-like (Ig)-fibronectin type III (Fn)-transmembrane domain structure with other members of the LRR-Ig-Fn protein superfamily such as the Slitrk family of proteins. Expression of LRFN1, -3, and -4 mRNA was detected in embryonic neuronal cells, while LRFN2 and LRFN5 expression was primarily restricted to more mature cells. LRFN1, -2, and -4 bound to PDZ domains of postsynaptic PSD95, re-distributing PSD95 to the cell periphery. It has been suggested that the Lrfn proteins play a role in the developing and/or mature vertebrate nervous system.

LGP2 Antibody: Anti-viral innate immune responses are triggered by pathogen-associated molecular patters (such as the accumulation of intracellular nucleic acids resulting from virus infections) and represent the first line of defense against numerous infectious organisms. The Toll-like receptors (TLR) 3, 7, 8, and 9 are expressed in immune cells and function as transmembrane pattern recognition receptors to detect foreign nucleic acids. Other proteins that play similar roles, such as RIG-1 and MDA5 are members of a CARD-helicase family and are expressed in the cytoplasm. A third protein, LGP2, is similar to RIG-1 and MDA5, except for lacking a homologous CARD domain. It is thought to act as an element of negative-feedback regulation of intracellular antiviral signaling. At least four isoforms of LGP2 are known to exist.

The COL-1 antibody recognises proteins of 80-200 kDa, identified as different members of the Carcinoembryonic Antigen family. CEA is synthesised during development in the fetal gut and is re-expressed in increased amounts in intestinal carcinomas and several other tumors. This antibody does not react with nonspecific cross-reacting antigen (NCA) and with human polymorphonuclear leucocytes. It shows no reaction with a variety of normal tissues and is suitable for staining of formalin/paraffin tissues. CEA is not found in benign glands, stroma, or malignant prostatic cells. CEA antibody is useful in detecting early foci of gastric carcinoma and in distinguishing pulmonary adenocarcinomas (60-70% are positive) from pleural mesotheliomas (rarely or weakly positive). CEA antibody positivity is seen in adenocarcinomas from the lung, colon, stomach, esophagus, pancreas, gallbadder, urachus, salivary gland, ovary, and endocervix.

Reacts with human Integrin beta 3, also called CD61. It associates with the alphaV-chain (CD51) to form vitronectin receptor, or with the alphaIIb-chain (CD41) to form the GpIIb/GpIIIa complex (CD41/CD61). The CD41/CD61 complex appears early in megakaryocyte maturation. The activated CD41/CD61 complex is a receptor for von Willebrand factor, soluble fibrinogen, fibronectin, vitronectin and thrombospondin. It plays a central role in platelet activation and aggregation. The CD51/CD61 is implicated in tumor metastasis and adenoviral infection. The antibody detects platelets in smears of blood and bone marrow, as well as megakaryocytes in frozen sections and cell smears. The antibody is useful for classification of megakaryoblastic leukemia.
