ZW10 Interactor is localized to the kinetochores from late Prophase to Anaphase and has a uniform distribution in the cytoplasm of Interphase cells. ZWINT interacts ZW10, MIS12 and NDC80 subunit of the NDC80 complex specifically during mitosis. ZWINT is a part of the MIS12 complex which is required for kinetochore formation and spindle checkpoint activity. In addition, ZWINT is required to target ZW10 to the kinetochore at prometaphase.

Ganglioside GM2 activator (GM2A) is a small glycolipid transport protein which acts as a substrate specific co-factor for the lysosomal enzyme beta -hexosaminidase A (HEXB). HEXB together with GM2A, catalyses the degradation of the ganglioside GM2, and other molecules containing terminal N-acetyl hexosamines. GM2A accommodate several single chain phospholipids and fatty acids, is a lipid transfer protein that stimulates the enzymatic processing of gangliosides, and also T-cell activation through lipid presentation. It extracts single GM2 molecules from membranes and presents them in soluble form to beta-hexosaminidase A for cleavage of N-acetyl-D-galactosamine and conversion to GM3.

Heat shock protein beta-2(HSPB2) is a protein that in humans is encoded by the HSPB2 gene. HSPB2 belongs to the superfamily of small heat-shock proteins containing a conservative alpha-crystallin domain at the C-terminal part of the molecule. It is expressed preferentially in the heart and skeletal muscle. HSPB2 has been shown to interact with TRAF6, HSPB8, Myotonic dystrophy protein kinase and CRYAB. HSPB2 regulates Myotonic Dystrophy Protein Kinase, which plays an important role in maintenance of muscle structure and function.

FAM19A4 is a secreted, 12 kDa member of the FAM19/TAFA family of chemokine-like proteins. Like other members of the FAM19/TAFA family, with the exception of TAFA5, mature FAM19A4 contains 10 regularly spaced cysteine residues. The FAM19A4 proteins are predominantly expressed in specific regions of the brain and the biological functions of FAM19A4 family members remain to be determined, but there are a few tentative hypotheses. First, FAM19A4 may modulate immune responses in the CNS by functioning as brain specific chemokines, and may act with other chemokines to optimize the recruitment and activity of immune cells in the CNS. Second, FAM19A4 may represent a novel class of neurokines that act as regulators of immune nervous cells. And third, FAM19A4 may control axonal sprouting following brain injury.

Growth Arrest and DNA Damage-Inducible Protein GADD45 alpha (GADD45A) is a member of the GADD45 family. GADD45A can be induced by UV irradiation, X-rays, growth arrest and alkylating agents, of which can be mediated by some kinases other than PKC. GADD45A can interact with MAPK14, GADD45GIP1, PCNA. In T-cells, GADD45A functions as a regulator of p38 MAPKs by inhibiting p88 phosphorylation and activity. GADD45A may affect PCNA interaction with some cell division protein kinase complexes, stimulating DNA excision repair in vitro and inhibits entry of cells into S phase.

It is a component of the ADA2A-containing complex (ATAC) complex, a complex with histone acetyltransferase activity on histones H3 and H4, and composed of CSRP2BP, KAT2A, TADA2L, TADA3L, ZZ3, MBIP, WDR5, YEATS2, CCDC101 and DR1. In the complex, it probably interacts directly with KAT2A, CSRP2BP and WDR5. It’s function to inhibit the MAP3K12 activity to induce the activation of the JNK/SAPK pathway.

Mouse stem cell factor (SCF), is the ligand for the receptor-type protein-tyrosine kinase KIT. It plays an essential role in the regulation of cell survival and proliferation, hematopoiesis, stem cell maintenance, gametogenesis, mast cell development, migration and function, and in melanogenesis. KITLG/SCF binding can activate several signaling pathways. It also promotes phosphorylation of PIK3R1, which is the regulatory subunit of phosphatidylinositol 3-kinase, and subsequent activation of the kinase AKT1.
