Description:
Sonic Hedgehog Homolog (SHH) belongs to a three-protein family called hedgehog. The other two family members are Indian Hedgehog (IHH) and Desert Hedgehog (DHH). Hedgehog proteins are key signaling molecules in embryonic development. SHH is expressed in various embryonic tissues and plays critical roles in regulating the patterning of many systems, such as limbs and brain. SHH also plays an important role in adult, including the division of adult stem cells and the development of certain cancers and other diseases. Human SHH is expressed as a 45kDa precursor, and undergoes a series of processing during secretion. After the removal of the signal peptide, a protease within the C-terminal domain catalyzes the cleavage of SHH into a 20 kDa N-terminal signaling domain (SHH-N) and a 25 kDa C-terminal domain (SHH-C). SHH-N has the “all signaling” capability. SHH-N binds to the 12 pass transmembrane protein Patched (Ptc) on cell surface, which releases the repression of the activity of Smoothened (Smo), a G-protein coupled receptor, by Ptc.

Bcl-2-Like Protein 1 (BCL2L1) is a member of the Bcl-2 family. BCL2L1 is expressed at high levels in cells that undergo a high rate of turnover, such as developing lymphocytes. BCL2L1 is a mitochondrial membrane protein. BCL2L1 contains four motifs, BH1, BH2 and BH4. The BH4 motif is required for anti-apoptotic activity. The BH1 and BH2 motifs are required for both heterodimerization with other Bcl-2 family members and for repression of cell death. BCL2L1 regulates cell death by blocking the voltage-dependent anion channnel (VDAC) and preventing the release of the caspase activator, CYC1, from the mitochondrial membrane. In addition, BCL2L1 promotes apoptosis.

Bcl-2-Related Protein A1 (BCL2A1) is a member of of the BCL-2 protein family which act as anti- and pro-apoptotic regulators that are involved in a wide variety of cellular activities such as embryonic development, homeostasis and tumorigenesis. BCL2A1 is a cytoplasm protein and interacts directly with BAK1, BID, BMF and BBC3. BCL2A1 is induced by phorbol ester and inflammatory cytokines, such as TNF or IL1B/interleukin-1 beta, but not by growth factors. Bone Morphogenetic Protein Receptor II (BMPR-II) is a Type II Serine/Threonine Kinase that mediates cellular responses to BMPs. BMPR-II is characterized by lacking of a GS domain, and presence of a C-terminal extension typical of type II receptors. BMPRII binds BMP2, BMP4 and BMP7 weakly in the absence of type I receptor, and the binding can be facilitated by the presence of the type I receptor, including BMPR-IA/Brk1, BMPR-IB, and ActR-I. BMPR-II plays a key role in cell growth. Defects in BMPR-II have been linked to primary pulmonary hypertension. Mouse interleukin 13 (mIL-13) is a pleiotropic cytokine produced by activated Th2 cells. IL-13 induces B cell proliferation and immunoglobin production. It contains a four helical bundle with two internal disulfide bonds. Mouse IL13 shares 58% sequence identity with human protein and exhibits cross-species activity. IL13 signals via receptor IL13R (type2, IL4R) and activates STAT-6. IL13 initially binds IL-13R alpha1 with low affinity and triggers association of IL4R alpha, generating a high affinity heterodimeric receptor IL13R and eliciting downstream signals. IL13 also binds IL-13R alpha2 with high affinity, which plays a role in a negative feedback system of IL13 signaling. IL13 is an important mediator of allergic inflammation and disease.

Chemokine (C-X-C Motif) Ligand 2 (CXCL2) is a small secreted cytokine which belongs to the CXC chemokine family. CXCL2 is 90% identical in amino acid sequence as a related chemokine, CXCL1. CXCL2/MIP2-alpha is secreted by monocytes and macrophages and is chemotactic for polymorphonuclear leukocytes and hematopoietic stem cells. The gene for CXCL2 is located on human chromosome 4 in a cluster of other CXC chemokines. CXCL2 mobilizes cells by interacting with a cell surface chemokine receptor called CXCR2. CXCL2/MIP2-alpha has been known to regulate immune functions mainly by chemo-attracting neutrophils. It is produced by activated monocytes and neutrophils and expressed at sites of inflammation. CXCL2 is a hematoregulatory chemokine, which suppresses hematopoietic progenitor cell proliferation. CXCL2 can be induced by receptor activator of NF-kappaB ligand, the osteoclast (OC) differentiation factor, through JNK and NF-kappaB signaling pathways in OC precursor cells. CXCL2 in turn enhanced the proliferation of OC precursor cells of bone marrow-derived macrophages (BMMs) through the activation of ERK. Knockdown of CXCL2 inhibited both the proliferation of and the ERK activation in BMMs. During osteoclastogenesis CXCL2 stimulated the adhesion and the migration of BMMs. CXCL2 is a novel therapeutic target for inflammatory bone destructive diseases.
