Methionine adenosyltransferase (EC 2.5.1.6) catalyzes the biosynthesis of S-adenosylmethionine (AdoMet) from methionine and ATP. AdoMet is the major methyl donor for many of the transmethylation reactions in the body. The MAT2A promoter is highly GC rich, especially in the first 300 bp. The promoter contains several potential SP1-, v-myb-, and GATA-binding sites, as well as potential binding sites for C/EBP, HSF2, c-myb, and STATx. It can efficiently drive expression from a reporter gene in both Jurkat and 293 cells.

Lumican is a 40 kD secreted protein which belongs to the small leucine-rich repeat proteoglycans (SLRPs) and the class II subfamily. Human Lumican is synthesized as a 338 amino acid precursor then cut the 18 aa signal sequence. The mature Human Lumican contains 12 leucine-rich repeats (LRRs), 4 potential sites of N-linked glycosylation, and a C- terminal with two conserved cyst-eines. Lumican can be existed in extracellular matrix of human articular cartilage. Lumican participates in the maintenance of tissue homeostasis and regulates cellular functions in vivo, such as cell proliferation, adhesion, migration, and differentiation. The overexpression of lumican has been correlated to colorectal tumor, breast, neuroendocrine, and pancreatic cancers.

FAS(TNFRSF6) is a receptor and contains three TNFR-Cys repeats and one death domain. It has been shown that FAS is involved in the physiological regulation of programmed cell death, and has been implicated in the pathogenesis of various malignancies and diseases of the immune system. FADD (adapter molecule) recruits caspase-8 to the activated receptor, the resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases mediating apoptosis. FAS-mediated apoptosis may play a role in the induction of peripheral tolerance, in the antigen-stimulated suicide of mature T-cells, or both.

Leukocyte-Associated Immunoglobulin-Like Receptor 2 (LAIR2) is a secreted, 131 amino acid protein that contains one Ig-like C2 type domain, making it a member of the Ig superfamily. When compared to LAIR-1, its transmembrane counterpart, it shares 83% amino acid identity across the signal sequence and extracellular domains; although one is secreted and one is membrane-bound, the two LAIR proteins are thought to have arisen from a common gene ancestor and appear to share similar adhesion profiles. This suggests that LAIR-2 may compete with LAIR-1 for ligand binding. A 114 amino acid alternate splice form of LAIR-2 is truncated at the C terminus, but retains the entire Ig domain. The expression profile of these splice forms, and the presence of orthologs in other species, have not been reported.

Fibronectin Leucine Rich Transmembrane Protein 1 (FLRT1) is a member of the Fibronectin Leucine Rich Transmembrane protein (FLRT) family. There are three fibronectin leucine-rich repeat transmembrane (FLRT) proteins: FLRT1, FLRT2 and FLRT3, all contain 10 leucine-rich repeats (LRR), a type III fibronectin (FN) domain, followed by the transmembrane region, and a short cytoplasmic tail. FLRT proteins have dual properties as regulators of cell adhesion and potentiators of fibroblast growth factor (FGF) mediated signalling. The fibronectin domain of all three FLRTs can bind FGF receptors. This binding is thought to regulate FGF signaling during development. The LRR domains are responsible for both the localization of FLRTs in areas of cell contact and homotypic cell association. FLRT1 is expressed at brain compartmental boundaries. FLRT1 is a target for tyrosine phosphorylation mediated by FGFR1 and implicate a non-receptor Src family kinase (SFK).

ESM-1, short from Endothelial cell-specific molecule 1, is expressed in lung, on the vascular capillary network within alveolar walls, and also at lower level in kidney. It is a secretory protein, and can be inducted by TNF and IL1B/interleukin-1beta, but not IL4/interleukin-4. This protein plays great roles in angiogenesis, sprouting, and may have potent implications in lung endothelial cell-leukocyte interactions.

Dectin-1 and dectin-2 are type II transmembrane proteins of the C-type lectin family. They contain single carbohydrate recognition domains (CRDs) in their extracellular region. Dectin-1 and dectin-2 are expressed in macrophages and dendritic cells. Dectin-1 recognises beta-glucans with its CRD and signals through its immunoreceptor tyrosine-based activation motif (ITAM)-like motif in the cytoplasmic domain, whereas dectin-2 recognises alpha-mannans and transduces its signal through association with the ITAM-containing Fc receptor gamma-chain.
