These proteins act through adaptor molecules such as MyD88 and TIRAP to activate various kinases and transcription factors. TLR7, like TLRs 3, 8, and 9, is localized in intracellular acidic compartments such as the phagolysosome and will recognize some single-stranded RNA viruses such as vesicular stomatitis virus (VSV) and influenza virus. Activation of TLR7 by VSV results in stimulation of the immune response including IFNalpha secretion, suggesting the importance of TLR7 in virus recognition.

TRIM5 gamma Antibody: TRIM5 is a member of a broad family of otherwise unrelated proteins defined by the presence of a tripartite motif containing a RING domain, a B-box type 1, and a B-box type 2, followed by a coiled-coil region. TRIM5 has five alternately spliced isoforms, the longest of which is the alpha variant which also contains a carboxy-terminal B30.2 (SPRY) domain. While a function has not yet been assigned to TRIM5gamma, it is known that expression of TRIM5 alpha variants from humans, rhesus monkeys, and African green monkeys enable resistance to infection by various retroviruses including HIV-1, albeit at differing efficiencies. Furthermore, the TRIM5 delta isoform appears to serve as a scaffold for the assembly of endogenous BTBD1 and BTBD2 proteins and also exhibits autoubiquitination activity in a RING finger- and UbcH5B-dependent manner.

PIST Antibody: Autophagy, the process of bulk degradation of cellular proteins through an autophagosomic-lysosomal pathway is important for normal growth control and may be defective in tumor cells. It is involved in the preservation of cellular nutrients under starvation conditions as well as the normal turnover of cytosolic components and is negatively regulated by TOR (Target of rapamycin). PIST, a PDZ-containing protein, was discovered in a yeast two-hybrid system as a binding partner to Beclin-1, a Bcl-2-interacting protein homologous to the yeast autophagy gene apg6. Experiments with mutant PIST proteins lacking the PDZ domain showed that PIST interaction with Beclin-1 through its coiled-coil domain can modulate Beclin-1 activity and suggest that PIST interactions with other proteins through its PDZ domain may regulate the activity of PIST and Beclin-1.

THEM4 Antibody: THEM4, also known as CTMP, binds specifically to the carboxy-terminal regulatory domain of PKB/Akt at the plasma membrane and acts as a negative regulator, reversing the phenotype of v-Akt-transformed cells. Hypermethylation of the THEM4 promoter and transcriptional downregulation of the gene has been reported in multiple glioblastomas, suggesting that epigenetic regulation of THEM4 may play a role in the progression of this cancer. Bioinformatic analysis, confirmed by in vitro testing, indicates that THEM4 is a broad-range, high activity acyl-CoA thioesterase. Recent reports have also indicated that TMEM4 is a mitochondrial protein whose overexpression is associated with an increase in mitochondrial membrane depolarization and caspase-3 and PARP cleavage, suggesting that THEM4 is involved in the apoptotic program.

APC7 Antibody: Cell cycle regulated protein ubiquitination and degradation within subcellular domains is thought to be essential for the normal progression of mitosis. APC7 is a highly conserved component of the anaphase promoting complex/cyclosome (APC/C), a cell cycle-regulated E3 ubiquitin ligase that controls progression through mitosis and the G1 phase of the cell cycle. APC/C is responsible for degrading anaphase inhibitors, mitotic cyclins, and spindle-associated proteins ensuring that events of mitosis take place in proper sequence. The individual APC/C components mRNA and protein levels are expressed at approximately the same levels in most tissues and cell lines, suggesting that they perform their functions as part of a complex. APC7 is required for proper protein ubiquitination function of APC/C and for the interaction of APC/C with various transcription coactivators.
