Description:
This antibody recognises a cell surface glycoprotein of 95/115/135 kDa (depending upon the extent of glycosylation), identified as CD43 [Workshop IV]. 70-90% of T-cell lymphomas and 22-37% of B-cell lymphomas express CD43. No reactivity has been observed with reactive B-cells. So a B-lineage population that co-expresses CD43 is highly likely to be a malignant lymphoma, especially a low-grade lymphoma, rather than a reactive B-cell population. When CD43 antibody is used in combination with CD20 antibody, effective immunophenotyping of the lymphomas in formalin-fixed tissues can be obtained. Co-staining of a lymphoid infiltrate with CD20 and CD42 antibody argues against a reactive process and favors a diagnosis of lymphoma.

This antibody recognises a ~90-95 kDa protein which is identified as cell surface transferrin receptor (CD71), a disulfide-bonded homodimeric glycoprotein of 180-190 kDa (Workshop IV). The antibody is highly specific to transferrin receptor and shows no cross-reaction with other related proteins. Ligand for transferrin receptor is the serum iron transport protein, transferrin. This receptor is broadly distributed in carcinomas, sarcomas, leukemias, and lymphomas. Transferrin receptor has been reported to be associated with cell proliferation in both normal and neoplastic tissues and useful in predicting clinical behavior or response to therapy in a number of malignancies including breast cancer.

The PAX gene family of nuclear transcription factors is comprised of nine members that function during embryogenesis to regulate the temporal and position-dependent differentiation of cells. In addition, the family is involved in a variety of signal transduction pathways in the adult organism. Mutations in the PAX family of proteins have been linked to disease and cancer in humans. PAX7 is a protein specifically expressed in cultured satellite cell- derived myoblasts. In situ hybridization reveals that PAX7 is also expressed in satellite cells residing in adult muscle. A chromosomal aberration in the gene encoding PAX7 causes rhabdomyosarcoma 2 (RMS2) (also called alveolar rhabdomyosarcoma).

An imbalance between MMPs and the associated TIMPs may play a significant role in the invasive phenotype of malignant tumors. TIMP’s inhibit the proteolytic invasiveness of tumor cells and normal placental trophoblast cells. TIMP3 may be involved in regulating trophoblastic invasion of the uterus as well as in regulating remodeling of the extracellular matrix during the folding of epithelia, and in the formation, branching and expansion of epithelial tubes.

This mAb recognises a protein of 40 kDa, identified as Thyroid transcription factor 1. TTF1 is a member of the NKx2 family of homeodomain transcription factors. It is expressed in epithelial cells of the thyroid gland and the lung. Nuclei from liver, stomach, pancreas, small intestine, colon, kidney, breast, skin, testes, pituitary, prostate, and adrenal glands are unreactive. antibody to TTF1 is useful in differentiating primary adenocarcinoma of the lung from metastatic carcinomas originating in the breast, mediastinal germ cell tumors, and malignant mesothelioma. The antibody can also be used to differentiate small cell lung carcinoma from lymphoid infiltrates. Loss of expression in non-small cell lung carcinoma has been associated with aggressive behavior of such neoplasms. TTF1 reactivity is also seen in thyroid malignancies.

Description:
Thyroglobulin is a 660 kDa dimeric pre-protein with mutiple glycosylation sites, detected at ~300 kDa in western blot. It is produced by and processed within the thyroid gland to produce the hormone thyroxine and triiodothyronine. Prior to forming dimers, thyroglobulin monomers undergo conformational maturation in the endoplasmic reticulation. The vast majority of follicular carcinomas of the thyroid will give positive immunoreactivity for thyroglobulin antibody even though sometimes only focally. Poorly differentiated carcinomas of the thyroid are frequently thyroglobulin antibody negative. Adenocarcinomas of other-than-thyroid origin do not react with this antibody. This antibody is useful in identification of thyroid carcinoma of the papillary and follicular types. Presence of thyroglobulin in metastatic lesions establishes the thyroid origin of tumor. Thyroglobulin antibody, combined with calcitonin antibody, can identify medullary carcinomas of the thyroid. Furthermore, thyroglobulin antibody, combined with TTF1 antibody, can be a reliable marker to differentiate between primary thyroid and lung neoplasms.

Melanocytes produce organelles called melanosomes which produce melanin, the pigment that gives color to skin, hair, eyes, scales and feathers. gp100 was identified in an attempt to clone the gene Tyrosinase, an enzyme required for melanin synthesis. Further testing determined that gp100 is a melanoma-specific protein and is responsible for melanosome maturation, facilitating the transition from amorphous rounded vesicles to fibrillary ellipsoid organelles.
Metastatic amelanotic melanoma can often be confused with a variety of poorly differentiated carcinomas, large cell lymphomas, and sarcomas using H & E stains alone. It is also difficult to differentiate melanoma from spindle cell carcinomas and various types of mesenchymal neoplasms. Clone HMB45 gp100 antibody stains fetal and neonatal melanocytes, junctional and blue nevus cells, and malignant melanoma. It also stains angiomyolipomas, tumors most commonly associated with the kidney. Intradermal nevi, normal adult melanocytes, and non-melanocytic cells are negative. This gp100 antibody does not stain tumor cells of epithelial, lymphoid, glial, or mesenchymal origin.

These enzymes catalyze the conversion of aldehydes and ketones to their corresponding alcohols by utilizing NADH and/or NADPH as cofactors. The 2F1 monoclonal antibody is specific for the mouse Killer cell Lectin-like Receptor G1 (KLRG1), a homodimer consisting of two N-glycosylated subunits of 30-38 kDa, also known as MAFA (Mast cell Function-associated Antigen). The antigen contains a cytoplasmic motif similar to ITIM (the immunoreceptor tyrosine-based inhibitory motif). KLRG1 is a receptor for cadherin, a family of transmembrane glycoproteins that mediate cell adhesion, and a common marker of T cell senescence. The receptor is believed to play an important role in the innate and adaptive immune system through the regulation of leukocytes. It is expressed on lymphokine-activated killer (LAK) cells, adherent LAK (A-LAK) cells, a sub-set of natural killer (NK) cells, T cells. In NK cells, it inhibits cytokine production and cytotoxicity activity.The receptor expression was not detected on the mouse peritoneal mast cells, or bone marrow mast cells.
