Description:
Nicastrin Antibody: Nicastrin, in addition to presenilin, PEN2, and APH-1 forms the gamma-secretase protein complex, a membrane-bound aspartyl protease that can cleave certain proteins at peptide bonds buried within the hydrophobic environment of the lipid bilayer. This cleavage is responsible for a key step in signaling from several cell-surface receptors and is thought to be required for the generation of the neurotoxic amyloid peptides that are central to the pathogenesis of Alzheimer's disease. Like the tumor necrosis factor-alpha-converting enzyme (TACE) and the beta-site cleavage enzyme (BACE) protease families, gamma-secretase will cleave the amyloid precursor protein (APP), but within the intramembrane region of APP, resulting in either the non-toxic p3 (from the alpha and gamma cleavage site) or the toxic Abeta amyloid peptide (from the beta and gamma cleavage site). It is thought that accumulation of the Abeta peptide is the precursor to Alzheimer's disease. Nicastrin is also thought to be involved in cell proliferation and signaling, especially in regards to activation of Notch receptors as loss of Nicastrin expression results in mouse embryonic lethality.

CCDC98 Antibody: CCDC98, also known as Abraxas 1, was identified through protein binding studies using the breast and ovarian predisposition protein BRCA1 as the binding target. CCDC98 recruits RAP80, a ubiquitin-binding protein, to BRCA1, allowing the formation of BRCA1 foci in response to DNA damage caused by ionizing radiation. Both CCDC98 and RAP80 are required for DNA damage resistance, G2-M checkpoint control, and DNA repair. Cells depleted of either CCDC98 or RAP80 exhibited increased sensitivity to ionizing radiation, although not as much as in BRCA1-depleted cells, suggesting that CCDC98 and RAP80 control only part of the DNA damage response role of BRCA1. At least two isoforms of CCDC98 are known to exist.

STEAP4 Antibody: The six-transmembrane epithelial antigen of prostate 4 (STEAP4) is a member of a family of metalloreductases identified as cell-surface antigens in prostate tissue. Similar to two other members of the STEAP family (STEAP 2 and STEAP3), STEAP4 promotes both iron and copper reduction. STEAP4 is highly expressed in placenta, lung, heart and prostate tissues, and overexpressed in prostate cancer cells compared to normal prostate cells. Overexpression of STEAP4 in prostate cells significantly increases cell growth and colony formation, suggesting STEAP4 may have a role in cell proliferation and prostate cancer progression.

Slug Antibody: Slug, a member of the Snail family of C2H2-type zinc finger transcription factors, was initially identified to be involved in epithelial-mesenchymal transitions as well as the formation of the neural tube during vertebrate embryogenesis. Like Snail, Slug transcription can be induced by growth factors such as FGF, BMP, and TGF-beta. Once expressed, Slug will bind E-box regions of promoters and repress transcription of genes such as E-cadherin and Claudin-1. More recently, its expression in breast, esophogeal, and colorectal carcinomas has been correlated with poor prognosis for survival. Furthermore, Slug can protect hemapoietic progenitor cells from radiation-induced apoptosis by repressing the p53-mediated transcription of Puma, a BH3-only antagonist of the anti-apoptotic members of the Bcl-2 family. Slug antibody has no cross-reactivity to Snail protein.

APC5 Antibody: Cell cycle regulated protein ubiquitination and degradation within subcellular domains is thought to be essential for the normal progression of mitosis. APC5 is a highly conserved component of the anaphase promoting complex/cyclosome (APC/C), a cell cycle-regulated E3 ubiquitin ligase that controls progression through mitosis and the G1 phase of the cell cycle. APC/C is responsible for degrading anaphase inhibitors, mitotic cyclins, and spindle-associated proteins ensuring that events of mitosis take place in proper sequence. The individual APC/C components mRNA and protein levels are expressed at approximately the same levels in most tissues and cell lines, suggesting that they perform their functions as part of a complex. While little is known of APC5, it is thought that APC5 associates with other APC/C components APC1, APC4, and CDC23 interdependently, such that loss of any one subunit reduces binding between the remaining three.

The KCNIP2 gene encodes a member of the family of voltage-gated potassium (Kv) channel-interacting proteins (KCNIPs), which belongs to the recoverin branch of the EF-hand superfamily. Members of the KCNIP family are small calcium binding proteins. They all have EF-hand-like domains, and differ from each other in the N-terminus. They are integral subunit components of native Kv4 channel complexes. They may regulate A-type currents, and hence neuronal excitability, in response to changes in intracellular calcium.
