Recognises a protein of 16-27 kDa, identified as human Interferon alpha 2 (IFNA2). Its epitope maps between amino acids 43-53 of the IFNA2. This mAb is specific for IFNA2 and does not cross-react with IFNA1. The epitopes of clones N27 and N39 mAbs are different and represent a good combination of antibodies to set up an ELISA assay for the quantitation of IFNA2 after viral infections. The IFNA family consists of 24 or more genes or pseudo-genes. The alpha interferons are mainly produced by lymphocytes, monocytes, macrophages, and cell lines such as Namalwa and KG1 following induction by viruses, nucleic acids, and glucocorticoid hormones. They are involved in virus resistance on target cells, inhibition of cell proliferation, induction of cytokines and regulation of expression of MHC class I antigens.

Description:
The superoxide-generating NADPH oxidase includes a membrane-bound flavocytochrome containing two subunits, gp91-phox and p22-phox, and the cytosolic proteins p47-phox and p67-phox. During activation of the NADPH oxidase, p47-phox and p67-phox migrate to the plasma membrane where they associate with the flavocytochrome, cytochrome b558, to form the active enzyme complex. The p22 and gp91-phox subunits also function as surface O2 sensors that initiate cellular signaling in response to hypoxic conditions. NOX4 is a renal gp91-phox homolog highly expressed at the site of erythropoietin production in the proximal convoluted tubule epithelial cells of the renal cortex. It is also expressed in fetal tissues, placenta, glioblastoma and vascular cells.

Description:
S100 belongs to the family of calcium binding proteins. S100A and S100B proteins are two members of the S100 family. S100A is composed of an alpha and beta chain whereas S100B is composed of two beta chains. S100 protein has been found in normal melanocytes, Langerhans cells, histiocytes, chondrocytes, lipocytes, skeletal and cardiac muscle, Schwann cells, epithelial and myoepithelial cells of the breast, salivary and sweat glands, as well as in glial cells. Neoplasms derived from these cells also express S100 protein, albeit non-uniformly. A large number of well-differentiated tumors of the salivary gland, adipose and cartilaginous tissue, and Schwann cell-derived tumors express S100 protein. Almost all malignant melanomas and cases of histiocytosis X are positive for S100 protein. Despite the fact that S100 protein is an ubiquitous substance, its demonstration is of great value in the identification of several neoplasms, particularly melanomas and their metastases.

Erythropoietin/EPO is a secreted, glycosylated cytokine hormone composed of four alpha helical bundles. It is the primary factor responsible for regulating erythropoiesis during steady-state conditions and in response to blood loss and hemorrhage in the adult organism. Erythropoietin is synthesised by the kidney and stimulates the proliferation and maturation of bone marrow erythroid precursor cells. The protein is found in the plasma and regulates red cell production by promoting erythroid differentiation and initiating hemoglobin synthesis.

von Willebrand Factor (vWF) is a multimeric glycoprotein that is found in endothelial cells, plasma and platelets. It acts as a carrier protein for Factor VIII and promotes platelet adhesion and aggregation. vWF undergoes a variety of posttranslational modifications that influence the affinity and availability for Factor VIII, including cleavage of the propeptide and formation of N-terminal disulfide bonds. This antibody helps to establish the endothelial nature of some lesions of disputed histogenesis, e.g. Kaposi's sarcoma and cardiac myxoma. It is widely used for differentiating vascular lesions from those of other tissue differentiation within a panel of other vascular markers although not all tumors of endothelial differentiation contain this antigen.

Recognises a 485 amino acid protein (55 kDa monomer/110 kDa homodimer), identified as platelet-derived endothelial growth factor (PD-ECGF), also called Thymidine Phosphorylase (TP, Tymp) or Gliostatin. In the presence of inorganic orthophosphate, it catalyses the reversible phospholytic cleavage of thymidine and deoxyuridine to their corresponding bases and 2-deoxyribose-1-phosphate. It is both chemotactic and mitogenic for endothelial cells and a non-heparin binding angiogenic factor present in platelets. Its enzymatic activity is crucial for angiogenic activity (metabolite is angiogenic). Higher levels of serum TP/PD-ECGF are observed in cancer patients. It is also involved in transformation of fluoropyrimidines, cytotoxic agents used in the treatment of a variety of malignancies, into active cytotoxic metabolites (e.g. 5 -deoxy-5-fluorouridine to 5-FU). High intra-cellular levels of TP/PD-ECGF are associated with increased chemosensitivity to such antimetabolites.

The HM40-3 monoclonal antibody reacts with mouse CD40, a 48 kDa type I glycoprotein known as Bp50. CD40 is part of the TNFR family and expressed on a subset of T cells, B cells, dendritic cells, and macrophages. It has been shown to be involved in the protection of B cells from apoptosis, B cell differentiation, co-stimulation, and isotype class-switching.
