Description:
Cerberus 1 is a secreted glycoprotein that forms disulfide-linked homodimers. It is a cytokine member of the DAN domain family of BMP antagonists that includes DAN (DAND1), Gremlin/Drm (DAND2), PRDC (Protein Related to Dan and Cerberus, DAND3), and COCO/Dante (DAND5). DAN family members contain a cysteine knot domain that is homologous to that found in other TGF-beta superfamily ligands. At the onset of gastrulation, Cerberus 1 is transiently expressed in anterior endodermal structures in response to Nodal and Shh. Cerberus 1 binds BMP-4 and Nodal and inhibits their activities. The inhibitory functions of Cerberus favor mesodermal development in the anterior region of the gastrula and suppresses posterior mesodermal differentiation. In chick and Xenopus, Cerberus 1 also regulates, but is not required for embryonic left-right polarization, neurulation, and head and heart induction.

GITRL (Glucocorticoid-induced TNF receptor ligand) is expressed on dendritic cells (DC), monocytes, macrophages, B cells, activated T cells, endothelial cells, osteoclasts and various healthy non-lymphoid tissues (e. g. testis). GITRL is constitutively expressed and released as soluble form by solid tumours and various hematopoietic malignancies. GITRL causes differentiation of osteoclasts, activation of macrophages, but also alteration of carcinoma and leukaemia cells and influences apoptosis. Binding to GITR is important in regulating T cell proliferation and TCR-mediated apoptosis. GITRL is implicated in development of autoimmune diseases and in the immune response against infectious pathogens and tumours.

Apolipoprotein M is a secreted protein which belongs to the Lipocalin family. ApoM often presents in high density lipoprotein (HDL) and to a lesser extent in triglyceride-rich lipoproteins (TGRLP) and low density lipoproteins (LDL). The ApoM gene encoded protein is expressed in liver and kidney, secreted through the plasma membrane but remains membrane-bound. ApoM probably involved in lipid transport. ApoM can bind sphingosine-1-phosphate, myristic acid, palmitic acid and stearic acid, retinol, all-trans-retinoic acid and 9-cis-retinoic acid. The expression of ApoM could be regulated by platelet activating factor (PAF), Transforming Growth Factors (TGF), Insulin-Like Growth factor (IGF) and Leptin.

Mannan-Binding Lectin Serine Protease 1 (MASP-1) belongs to the peptidase S1 family. MASP1 contains two CUB domains, one EGF-like domain, one peptidase S1 domain and two Sushi (CCP/SCR) domains. MASP1 is primarily expressed in liver. MASP1 involved in the lectin pathway of the complement, performs a key role in innate immunity by recognizing pathogens through patterns of sugar moieties and neutralizing them. MASP1 is synthesized as a zymogen and activated when it complexes with the pathogen recognition molecules of lectin pathway, the mannose-binding lectin and the ficolins. MASP1 is not directly involved in complement activation but may act as an amplifier of complement activation by cleaving complement C2 or by activating another complement serine protease, MASP2. MASP1 is also able to cleave fibrinogen and factor XIII and may may be involved in coagulation. MASP1 is inhibited by SERPING1 and A2M.

CD99 is a type I transmembrane glycoprotein and the founding member of the CD99 family of molecules. The extracellular domain of CD99 contains no identifiable motifs, its cytoplasmic region, although short, does have signal transduction capability. Cells known to express CD99 include fibroblasts, neutrophils, T cells, double positive thymocytes, CD34+ stem cells, monocytes and endothelial cells. Two types of CD99 isoforms have been classified. Native human CD99 is referred to as the long, or type I isoform. The best studied type II isoform shows an Asp-Gly substitution for the C terminal 27 amino acids. The type I and II isoforms have distinctive signal transduction pathways (FAKsrc for type I PI3K plus srcERK1/2 for type II), and mediate clearly different biological outcomes. Homophilic interaction between CD99 on the neutrophil and CD99 on the endothelial cell regulates the transendothelial migration of neutrophils during inflammation. Human CD99 has 48% aa sequence identity to mouse CD99.
