Neuroligin 4, X-Linked (NLGN4X) is a single-pass type I membrane protein that belongs to the type-B carboxylesterase/lipase family. NLGN4X is detected at higher levels in heart and at lower levels in the liver, skeletal muscle, and pancreas. NLGN4X is a putative neuronal cell surface protein involved in cell-cell-interactions. NLGN4X may act as splice site-specific ligands for beta -neurexins. It has been shown that NLGN4X is involved in the formation and remodeling of central nervous system synapses. NLGN4X also interacts with discs, large (Drosophila) homolog 4 (DLG4). Defects in NLGN4X have been associated with autism and Asperger syndrome.

tumour necrosis factor receptor superfamily member 1B is a 461 amino acids protein that belongs to the TNFR (tumour necrosis factor receptor) superfamily Characterised by cysteine-rich extracellular domains. It contains 4 TNFR-Cys repeats. TNFRII is expressed in fetal brain. TNFRII is strongly expressed at the cartilage-pannus junction, and plays a major role in a subset of families with multiple cases of rheumatoid arthritis (RA). This receptor mediates most of the metabolic effects of TNF-alpha. Isoform 2 blocks TNF-alpha-induced apoptosis, which suggests that it regulates TNF-alpha function by antagonizing its biological activity.

TIP60 is the catalytic subunit of the NuA4 histone acetyltransferase complex which is involved in transcriptional activation of selected genes principally by acetylation of nucleosomal histones H4 and H2A. This modification may both alter nucleosome-DNA interactions and promote interaction of the modified histones with other proteins which positively regulate transcription. This complex is required for the activation of transcriptional programs associated with oncogene and proto-oncogene mediated growth induction, tumor suppressor mediated growth arrest and replicative senescence, apoptosis and DNA repair. NuA4 may also play a direct role in DNA repair when recruited to sites of DNA damage. TIP60 directly acetylates and activates ataxia-telangiectasia mutated (ATM). In case of HIV-1 infection, interaction with the viral Tat protein leads to KAT5 polyubiquitination and targets it to degradation.

PARP-1 (ARTD1) is involved in the base excision repair (BER) pathway, by catalyzing the poly(ADP-ribosyl)ation of a limited number of acceptor proteins involved in chromatin architecture and in DNA metabolism. This modification follows DNA damages and appears as an obligatory step in a detection/signalling pathway leading to the reparation of DNA strand breaks. PARP-1 positively regulates the transcription of MTUS1 and negatively regulates the transcription of MTUS2/TIP150. It forms a complex with EEF1A1 and TXK that acts as a T-helper 1 (Th1) cell-specific transcription factor and binds the promoter of IFN-gamma to directly regulate its transcription, and is thus involved importantly in Th1 cytokine production.

Interleukin-12 (IL-12), also known as Natural Killer Cell Stimulatory Factor (NKSF) or Cytotoxic Lymphocyte Maturation Factor (CLMF), is a heterodimeric pleiotropic cytokine made up of a 40 kDa (p40) subunit and a 35 kDa (p35) subunit. IL-12 is produced by macrophages and B lymphocytes and has been shown to have multiple effects on T cells and Natural Killer (NK) cells. Some of these IL-12 activities include the induction of IFN-gamma and TNF in resting and activated T and NK cells; the enhancement of cytotoxic activity of resting NK and T cells, the stimulation of resting T cell proliferation in the presence of a comitogen; and the enhancement of NK cell proliferation. Current evidence indicates that IL-12 is a key mediator of cellular immunity and induces the differentiation of Th1 cells from precursor T helper cells. Based on its activities, it has been suggested that IL-12 may have therapeutic potential as a vaccine adjuvant that promotes cellular immunity and as an antitumour and anti viral agent.

Angiopoietin-1 (Ang-1) and Angiopoietin-2 (Ang-2) are closely related secreted ligands which bind with similar affinity to Tie-2. Tie-2 and angiopoietins have been shown to play critical roles in embryogenic angiogenesis and in maintaining the integrity of the adult vasculature. Ang-1 activates Tie-2 signalling on endothelial cells to promote chemotaxis, cell survival, cell sprouting, vessel growth and stabilisation. Ang-2 has been identified as a secreted protein ligand of Tie-2 and has alternatively been reported to be an antagonist for Ang-1 induced Tie-2 signalling as well as an agonist for Tie-2 signalling, depending on the cell context.

Description:
Cochlin is an extracellular matrix protein expressed in the cochlea and vestibule of the inner ear. Mutations in the cochlin gene are linked to the autosomal-dominant DFNA9 syndrome, a disorder characterised by hearing loss. Cochlin is also expressed in follicular dendritic cells of the spleen and the lymph nodes. The N-terminus of cochlin contains a LCCL domain also present in Factor C, a serine protease of the horseshoe crab Limulus, involved in activating the coagulation cascade in response to LPS. A recent study showed that in response to inflammation, LPS or bacterial infection, cochlin is cleaved by the proteases aggrecanases and secreted into blood circulation. The secreted cochlin (LCCL domain) seems to have a role in local cytokine production, recruitment of immune effector cells and bacterial clearance. Cochlin is secreted as two forms p8 and p18, whereas p18 being the glycosylated form of p8.

TLR11, an innate sensor for profilin (Toxoplasma gondii), is an intracellular receptor that resides in the endoplasmic reticulum. The 12 membrane-spanning endoplasmic reticulum-resident protein UNC93B1 interacts directly with TLR11 and regulates the activation of dendritic cells in response to profilin (Toxoplasma gondii) and parasitic infection in vivo. TLR12, a previously uncharacterised TLR, also recognises profilin from Toxoplasma gondii. TLR12 is sufficient for recognition of profilin by plasmacytoid dendritic cells (pDCs), whereas TLR11 and TLR12 are both required in macrophages and conventional DCs. In contrast to TLR11, TLR12-deficient mice succumb rapidly to T. gondii infection.

Irisin activates beige fat cells (beige cells have a gene expression pattern distinct from either white or brown fat and are preferentially sensitive to the polypeptide hormone irisin).
