Pleckstrin Homology-Like Domain Family A Member 2 (PHLDA2) is a peripheral membrane protein that belongs to the PHLDA2 family. PHLDA2 is expressed in the placenta and adult prostate gland. In the placenta, it is present in all cells of the villous cytotrophoblast. PHLDA2 plays a role in regulating placenta growth. PHLDA2 may act via its PH domain that competes with other PH domain-containing proteins, thereby preventing their binding to membrane lipids.

GDNF family receptor alpha-2 is a glycosylphosphatidylinosito l (GPI)-linked cell surface receptor. It is part of the GDNF receptor family. Glial cell line-derived neurotrophic factor (GDNF) and neurturin (NTN) are two structurally related, potent neurotrophic factors that play key roles in the control of neuron survival and differentiation. GFRA2 mediates the NRTN-induced autophosphorylation and activation of the RET receptor. It also able to mediate GDNF signalling through the RET tyrosine kinase receptor. It acts preferentially as a receptor for NTN compared to its other family member, GDNF family receptor alpha 1.

Alpha-2-HS-glycoprotein (AHSG) is a glycoprotein that is composed of two subunits, the A and B chains, belongs to the Cystatin family of proteases inhibitors. It is highly expressed in embryonic cells and adult hepatocytes, and is expressed to a lesser extent in monocytes/macrophages. AHSG is an important circulating inhibitor of calcification in vivo, and is downregulated during the acute-phase response. It is involved in several functions, such as endocytosis, brain development and the formation of bone tissue. In addition, AHSG may influence the resolution of inflammation by modulating the phagocytosis of apoptotic cells by macrophages. ASHG blocks TGF-beta-dependent signalling in osteoblastic cells.

Platelet-derived growth factor receptors (PDGFR) are cell surface tyrosine kinase receptors for members of the platelet-derived growth factor (PDGF) family. The PDGF family consists of PDGF-A, -B, -C and -D, which form either homo- or heterodimers (PDGF-AA, -AB, -BB, -CC, -DD). The four PDGFs are inactive in their monomeric forms. PDGFs bind to the protein tyrosine kinase receptors PDGF receptor- alpha and - beta . These two receptor isoforms dimerise upon binding the PDGF dimer, leading to three possible receptor combinations, namely - alpha alpha, - beta beta and - alpha beta . PDGFR alpha and PDGFR beta are members of the class III RTK family. Inappropriate PDGFR alpha and PDGFR beta signalling has been linked to a number of proliferative disorders.

Interleukin 23 receptor (IL23R) is a type I cytokine receptor for IL23. IL23 receptor complex is comprised of two subunits, the IL12R beta 1 subunit, which is shared with several cytokines, and a subunit that is unique to IL-23. IL23, after binding to IL23R, activates memory T cells and mediates pro-inflammatory activities in part by the production of IL17 through activation of TH17 lymphocytes. IL23R is expressed on T cells, NK cells, dendritic cells, and macrophages. In fact, polymorphisms of the IL23R gene were reported to be associated with susceptibility to inflammatory diseases and autoimmune diseases such as psoriasis, multiple sclerosis, Graves's ophtalmopathy and inflammatory bowel diseases. The IL23R is known to be critically involved in the carcinogenesis of different malignant tumour.

Cytoplasmic Protein NCK1 (NCK1) is a cytoplasmic protein that contains one SH2 domain and three SH3 domains. NCK1 is a member of the adapter family, which associates with tyrosine-phosphorylated growth factor receptors, such as KDR and PDGFRB, or their cellular substrates. NCK1 maintains low levels of EIF2S1 phosphorylation by promoting its dephosphorylation by PP1. NCK1 plays a role in the DNA damage response, but not in the detection of the damage by ATM/ATR. Estrogen Receptor Beta (ESR2) is a nuclear protein that belongs to the nuclear hormone receptor family of NR3 subfamily. It contains one nuclear receptor DNA-binding domain and is expressed in many tissues at a lower level. ESR2 is a nuclear hormone receptor. It binds estrogens with an affinity similar to that of ESR1 and activates expression of reporter genes containing estrogen response elements (ERE) in an estrogen-dependent manner. DNA-binding by ESR1 and ESR2 is rapidly lost at 37 degrees Celsius in the absence of ligand while in the presence of 17 beta-estradiol and 4-hydroxy-tamoxifen loss in DNA-binding at elevated temperature is more gradual.

SMAD Family Member 4 (SMAD4) is a cytoplasmic protein that belongs to the Dwarfin/SMAD family. SMAD4 contains one MH1 (MAD homology 1) domain and one MH2 (MAD homology 2) domain. It is the component of the heterotrimeric SMAD2/SMAD3-SMAD4 complex that forms in the nucleus and is required for the TGF-mediated signaling. SMAD4 promotes binding of the SMAD2/SMAD4/FAST-1 complex to DNA and provides an activation function required for SMAD1 or SMAD2 to stimulate transcription. SMAD4 may act as a tumor suppressor. It positively regulates PDPK1 kinase activity by stimulating its dissociation from the 14-3-3 protein YWHAQ which acts as a negative regulator. Mutations or deletions in SMAD4 have been shown to result in pancreatic cancer, juvenile polyposis syndrome, and hereditary hemorrhagic telangiectasia syndrome.

Human Chemokine (C-X-C motif) Ligand 4 (CXCL4) is expressed in megakaryocytes and stored in the alpha-granules of platelets. CXCL4 contains several heparin-binding sites at the C-terminal region and binds heparin with high affinity. The active CXCL4 protein is a tetramer. Human and mouse CXCL4 share 64% sequence identity. CXCL4 is chemotactic for neutrophils, fibroblasts and monocytes and plays a critical role in inflammation and wound repair. CXCL4 functions via a splice variant of the chemokine receptor CXCR3, known as CXCR3B. The major physiologic role of CXCL4 appears to be neutralisation of heparin-like molecules on the endothelial surface of blood vessels, thereby inhibiting local antithrombin III activity and promoting coagulation. In contrast to other CXC chemokines, CXCL4 lacks chemotactic activity for polymorphonuclear granulocytes.

It is expressed in Expressed at high levels in erythrocytes and testis, and, at lower levels, in skeletal muscle, tonsils, lymph node, spleen and appendix. Vaspin (Visceral Adipose-Specific SERPIN) is a newly described adipokine. Vaspin has three beta -sheets, nine alpha-helices, and one central loop; the structure is part of the set of distinctive features that are descriptive of Serpin family members. Vaspin is also a unique insulin sensitizing adipocytokine in obesity. A recent publication indicates that Vaspin mRNA expression in visceral fat is positively correlated with BMI and percent of body fat. and could be associated with parameters of obesity, insulin resistance, and glucose metabolism. These findings suggest a potential clinical use for Vaspin in ameliorating certain aberrations seen in the obesity metabolic syndrome.

Amiloride-sensitive amine oxidase (AOC1) belongs to the copper/topaquinone oxidase family. The protein exists as homodimer by disulfide and mainly located in placenta and kidney. AOC1 catalyses the degradation of compounds such as putrescine, histamine, spermine, and spermidine, substances involved in allergic and immune responses, cell proliferation, tissue differentiation, tumour formation, and possibly apoptosis. Placental DAO is thought to play a role in the regulation of the female reproductive function. The activity of this protein can be inhibited by amiloride in a competitive manner. It is inhibited by amiloride, a diuretic that acts by closing epithelial sodium ion channels.
