Recognizes a protein of 33-55 kDa, identified as CD53 (Workshop V; Code CD53.1). CD53 is expressed on monocytes, and macrophages, granulocytes, dendritic cells, osteoblasts and osteoclasts, NK cells, and on T- and B-cells from every stage of differentiation but is absent from platelets, erythrocytes, and non-haemopoietic cells. CD53 is a member of a family of tetraspan transmembrane proteins, including CD9, CD37, CD63, CD81, and CD82. It associates with integrins, MHC class II molecules, and a tyrosine phosphatase and plays a role in cellular activation as part of a signal transduction complex involving other membrane glycoproteins. Defects of CD53 expression on neutrophils appear to be related with recurrent infectious diseases. Cross-linking CD53 using CD53 antibodies led to cytoplasmic calcium fluxes in B cells, monocytes, and granulocytes and activation of the monocyte oxidative burst.

Importin 7 is a 1,038 amino acid protein encoded by the human gene IPO7. Importin-7 belongs to the importin b family and contains one importin N-terminal domain. Importin-7 functions in nuclear protein import, either by acting as an autonomous nuclear transport receptor or as an adapter-like protein in association with the Importin b subunit KPNB1. Acting autonomously, Importin-7 is thought to serve itself as receptor for nuclear localization signals (NLS) and to promote translocation of import substrates through the nuclear pore complex (NPC) by an energy requiring, Ran-dependent mechanism. At the nucleoplasmic side of the NPC, Ran binds to Importin-7, the Importin-7/substrate complex dissociates and Importin-7 is re-exported from the nucleus to the cytoplasm where GTP hydrolysis releases Ran. Importin-7 is a nuclear protein that is expressed in most tissues.

Glucosylceramide synthase (GCS), also designated ceramide glucosyltransferase, belongs to the glycosyltransferase 2 family. It is a widely expressed integral membrane protein encoded by UGCG. The enzyme can be found in the plasma membrane of all eukaryotic cells, and a significant concentration of glucosylceramide synthase activity has been reported in the Golgi complex. Glucosylceramide synthase catalyzes the first glycosylation step in glycosphingolipid biosynthesis and functions as a glucosyltransferase and flippase in the transfer of glucose to ceramide. Glucosylceramide synthase operates in cell recognition, cell proliferation and differentiation, immune recognition and signal transduction. The regulation of ceramide levels through glucosylceramide synthase has been associated with the induction of apoptosis and notable research implicates this relationship with drug-induced apoptosis in a variety of cell types.

The finding that mutations in DNA mismatch repair genes are associated with hereditary nonpolyposis colorectal cancer (HNPCC) has resulted in considerable interest in the understanding of the mechanism of DNA mismatch repair. Initially, inherited mutations in the MSH2 and MLH1 homologs of the bacterial DNA mismatch repair genes MutS and MutL were demonstrated at high frequency in HNPCC and were shown to be associated with microsatellite instability. The demonstration that 10 to 45% of pancreatic, gastric, breast, ovarian and small cell lung cancers also display microsatellite instability has been interpreted to suggest that DNA mismatch repair is not restricted to HNPCC tumors but is a common feature in tumor initiation or progression. Two additional homologs of the prokaryotic MutL gene, designated PMS1 and PMS2, have been identified and shown to be mutated in the germline of HNPCC patients.
