Niemann-Pick disease type C2 protein (NPC2) is also known as epididymal secretory protein E1 and human epididymis-specific protein 1 (HE1). NPC2 is a intracellular cholesterol transporter which acts in concert with NPC1 and plays an important role in the egress of cholesterol from the endosomal/lysosomal compartment. Both NPC1 and NPC2 function as the cellular 'tag team duo' (TTD) to catalyze the mobilization of cholesterol within the multivesicular environment of the late endosome (LE) to effect egress through the limiting bilayer of the LE. Also,NPC2 binds unesterified cholesterol that has been released from LDLs in the lumen of the late endosomes/lysosomes and transfers it to the cholesterol-binding pocket of the N-terminal domain of NPC1. Furthermore,the secreted form of NCP2 regulates biliary cholesterol secretion via stimulation of ABCG5/ABCG8-mediated cholesterol transport.

Neuropilin-1 (NRP1) is also known as Vascular endothelial cell growth factor 165 receptor (VEGF165R), CD antigen CD304, which belongs to the neuropilin family. The membrane-bound isoform 1 is a receptor involved in the development of the cardiovascular system, in angiogenesis, in the formation of certain neuronal circuits and in organogenesis outside the nervous system. It mediates the chemorepulsant activity of semaphorins. It binds to semaphorin 3A, The PLGF-2 isoform of PGF, The VEGF-165 isoform of VEGF and VEGF-B. Coexpression with KDR results in increased VEGF-165 binding to KDR as well as increased chemotaxis. It may regulate VEGF-induced angiogenesis. Interleukin-12 (IL12) is also known as natural killer cell stimulatory factor (NKSF), cytotoxic lymphocyte maturation factor (CLMF) , is a heterodimeric cytokine encoded by two separate genes, IL-12A (p35) and IL-12B (p40). IL12 is naturally produced by dendritic cells, macrophages and human B-lymphoblastoid cells (NC-37) in response to antigenic stimulation. IL-12 is involved in the differentiation of naive T cells into Th0 cells and plays an important role in the activities of natural killer cells and T lymphocytes.IL-12 also has anti-angiogenic activity, which means it can block the formation of new blood vessels.
Interleukin-12 subunit beta (IL12B) also known as NKSF2, CLMF2 and P40. Interleukin-12 subunit beta has been shown to interact with IL23. A large excess of monomeric IL12B is also secreted by the cells producing IL12, and exhibits no demonstrable biological activity. Overexpression of IL12B gene has been shown to be associated with the pathogenesis of multiple sclerosis. In addition, studies have revealed that the promoter polymorphism of this gene is implicated in the severity of atopic and non-atopic asthma in children.

Mouse Ifng is a secreted protein which belongs to the type I I (or gamma) interferon family. IFNG is produced by lymphocytes and activated by specific antigens or mitogens. In addition to having antiviral activity, IFNG also has important immunoregulatory functions. It is a potent activator of macrophages and has antiproliferative effects on transformed cells. It can potentiate the antiviral and antitumor effects of the type I interferons. Genetic variation in IFNG is associated with the risk of aplastic anaemia (AA) which is a rare disease in which the reduction of the circulating blood cells results from damage to the stem cell pool in bone marrow. In most patients, the stem cell lesion is caused by an autoimmune attack. T-lymphocytes, activated by an endogenous or exogenous, and most often unknown antigenic stimulus, secrete cytokines, including IFN-gamma, which would in turn be able to suppress haematopoiesis.
