MATN3 Antibody: Matrilins (MATNs) are a family of non-collagenous extra-cellular matrix (ECM) proteins consisting of four known members that have been proposed to play key roles in modulating cellular phenotypes during chondrogenesis of mesenchymal stem cells (MSCs). MATN1 and MATN3 are expressed specifically in cartilage and are among the most up-regulated ECM proteins during chondrogenesis. MATN3 is composed of a single N-terminal von Willebrand Factor A (vWFA) domain followed by four epidermal growth factor (EGF) repeats and a coiled-coil domain whereas MATN1 is composed of two vWFA domains separated by one EGF-like domain. MATN1 or MATN3 may play a role in modulating chondrogenesis during the chondrocyte differentiation process. Mutations of this gene have been associated with variety of inherited chondrodysplasias. Recent studies show that aberrant expression and processing of MATN3 are hallmarks of conventional cartilaginous neoplasms.

CD3e molecule, epsilon is also known as CD3E, is a T-cell surface single-pass type I membrane glycoprotein. CD3E contains 1 Ig-like (immunoglobulin-like) domain and 1 ITAM domain. CD3E, together with CD3-gamma, CD3-delta and CD3-zeta, and the T-cell receptor alpha/beta and gamma/delta heterodimers, forms the T cell receptor-CD3 complex. This complex plays an important role in coupling antigen recognition to several intracellular signal-transduction pathways. The genes encoding the epsilon, gamma and delta polypeptides are located in the same cluster on chromosome 11. The epsilon polypeptide plays an essential role in T-cell development. CD3E plays an essential role in T-cell development, and defects in CD3E gene cause severe immunodeficiency. CD3E gene has also been linked to a susceptibility to type I diabetes in women. CD3E has been shown to interact with TOP2B, CD3EAP and NCK2.

The RPA-T4 monoclonal antibody specifically binds to the CD4 receptor for the human immunodeficiency virus (HIV). CD4 is a 59 kDa single-chain transmembrane glycoprotein that expressed on the surface of most of the thymocytes, T-helper cells, and in low levels on monocytes and macrophages. CD4 is a co-receptor in the antigen-induced T cell activation, together with the MHC class II. The RPA-T4 antibody is capable of blocking HIV binding and inhibiting syncytium formation, by binding to the D1 domain of the CD4 antigen. GALNT4 catalyzes the initial reaction in O-linked oligosaccharide biosynthesis, the transfer of an N-acetyl-D-galactosamine residue to a serine or threonine residue on the protein receptor. It has a highest activity toward Muc7, EA2 and Muc2, with a lowest activity than GALNT2. This gene encodes a member of the UDP-N-acetyl-alpha-D-galactosamine:polypeptide N-acetylgalactosaminyltransferase (GalNAc-T) family of enzymes. GalNAc-Ts initiate mucin-type O-linked glycosylation in the Golgi apparatus by catalyzing the transfer of GalNAc to serine and threonine residues on target proteins. They are characterized by an N-terminal transmembrane domain, a stem region, a lumenal catalytic domain containing a GT1 motif and Gal/GalNAc transferase motif, and a C-terminal ricin/lectin-like domain. GalNAc-Ts have different, but overlapping, substrate specificities and patterns of expression. In vitro, the encoded protein can complement other GalNAc-Ts in the complete O-glycosylation of the mucin-1 tandem repeat and can O-glycosylate the P-selectin glycoprotein ligand-1 molecule. The coding region of this gene is contained within a single exon.

This gene encodes a common acute lymphocytic leukemia antigen that is an important cell surface marker in the diagnosis of human acute lymphocytic leukemia (ALL). This protein is present on leukemic cells of pre-B phenotype, which represent 85% of cases of ALL. This protein is not restricted to leukemic cells, however, and is found on a variety of normal tissues. It is a glycoprotein that is particularly abundant in kidney, where it is present on the brush border of proximal tubules and on glomerular epithelium. The protein is a neutral endopeptidase that cleaves peptides at the amino side of hydrophobic residues and inactivates several peptide hormones including glucagon, enkephalins, substance P, neurotensin, oxytocin, and bradykinin. This gene, which encodes a 100-kD type II transmembrane glycoprotein, exists in a single copy of greater than 45 kb. IL6ST /gp130 /CD130 can be found in tissues such as brain, heart, thymus, spleen, kidney, lung and liver and found in all the cell lines tested except BaF-B03.The expression of IL-6ST /gp130 is not restricted to IL6-responsive cells. The receptor systems for IL6, LIF, OSM, CNTF, IL11, CTF1 and BSF3 can utilize gp130 for initiating signal transmission. IL6ST /CD130 can bind to IL6 /IL6R (alpha chain) complex, resulting in the formation of high-affinity IL6 binding sites, and transduce the signal. IL6ST /GP130 does not bind IL6 and may have a role in embryonic development.

Livin Antibody: Apoptosis, or programmed cell death, is related to many diseases, such as cancer. Apoptosis is triggered by a variety of stimuli including members in the TNF family and prevented by the inhibitor of apoptosis (IAP) proteins. IAP proteins form a conserved gene family that binds to and inhibits cell death proteases. A novel member in the IAP protein family was recently identified and designated Livin and KIAP for kidney IAP. Livin/XIAP contains a single baculoviral IAP repeat (BIR) domain and a RING finger domain and has two isoforms termed Livin-alpha and Livin-beta. Transfection of Livin in cells resulted in protection from apoptosis induced by FADD, BAX, RIP, RIP3 and DR6. Livin has direct interaction with several caspases including caspase-3, -7, and -9. Livin inhibits the activation of caspase-9 induced by Apaf-1, cytochrome c, and dATP. The two isoforms of Livin appear to have different functions and tissue distributions.

Interferon alpha/ beta Receptor 2 (IFN- alpha/ beta R2) is a single-pass type I membrane protein which belongs to the type II cytokine receptor family. It complexes with IFN- alpha/ beta R1 to form the signaling receptor complex for the family of alpha and beta IFN subtypes. By alternative splicing, IFN- alpha/ beta R2 can exist as a secreted soluble protein or as a type I membrane protein. IFN- alpha/ beta R2 is the principal ligand binding subunit of the receptor. Ligand binding is stabilized by the subsequent association with IFN- alpha/ beta R1, resulting in the formation of a signaling ternary receptor complex. IFNAR2 was detected in most lymphocytes, monocytes, and granulocytes, although IFNAR2 expression was higher in the monocytes and granulocytes than in the lymphocytes. Among the lymphocyte subsets, IFNAR2 showed high expression in natural killer (NK) cells and low expression in T lymphocytes. Isoform 1 and isoform 3 of IFNAR2 are directly involved in signal transduction due to their interaction with the TYR kinase, JAK1. Isoform 1 also interacts with the transcriptional factors, STAT1 and STAT2. Both forms are potent inhibitors of type I IFN activity.

Tumor necrosis factor receptor superfamily member 12A(Tnfrsf12a) is a single-pass type I membrane protein and contains 1 TNFR-Cys repeat. It is weak inducer of apoptosis in some cell types.It promotes angiogenesis and it is the proliferation of endothelial cells. It may modulate cellular adhesion to matrix proteins.TNFR binds specifically to tumor necrosis factor (TNF) and blocks its interaction with cell surface TNF receptors. TNF is a naturally occurring cytokine that is involved in normal inflammatory and immune responses. It plays an important role in the inflammatory processes of rheumatoid arthritis (RA), polyarticular-course juvenile rheumatoid arthritis (JRA), and ankylosing spondylitis and the resulting joint pathology. In addition, TNF plays a role in the inflammatory process of plaque psoriasis. Elevated levels of TNF are found in involved tissues and fluids of patients with RA, psoriatic arthritis, ankylosing spondylitis (AS), and plaque psoriasis. Two distinct receptors for TNF (TNFRs), a 55 kilodalton protein (p55) and a 75 kilodalton protein (p75), exist naturally as monomeric molecules on cell surfaces and in soluble forms. Biological activity of TNF is dependent upon binding to either cell surface TNFR.
