Tumor necrosis factor receptor superfamily member 21 (TNFRSF21) is also known as death receptor 6 (DR6), which is a member of the TNF-receptor superfamily. TNFRSF21 contains one death domain and four TNFR-Cys repeats. TNFRSF21 / DR6 has been shown to activate NF-κB and MAPK8/JNK, and induce cell apoptosis. Through its death domain, this receptor interacts with TRADD protein, which is known to serve as an adaptor that mediates signal transduction of TNF-receptors.

SERPINE2 is also known as Glia-derived nexin (GDN), Peptidase inhibitor 7 (PI7), Protease nexin 1(PN1). SERPINE2 is a secreted glycoprotein which belongs to the serpin family. SerpinE1 is the primary physiological inhibitor of the two plasminogen activators urokinase (uPA) and tissue plasminogen activator (tPA). PAI-1 / GDN is also implicated in adipose tissue development. It suggests that PAI-1 inhibitors serve in the control of atherothrombosis. Defects in Serpin E1 / PN1 are the cause of plasminogen activator inhibitor-1 deficiency (PAI-1 deficiency) which is characterized by abnormal bleeding due to SerpinE1 defect in the plasma.

SUMO-Conjugating Enzyme UBC9 (UBC9) belongs to the ubiquitin-conjugating enzyme family. UBC9 is homologous to ubiquitin-conjugating enzymes (E2s). However, instead of conjugating ubiquitin, UBC9 conjugates a ubiquitin homologue, Small Ubiquitin-Like Modifier 1 (SUMO-1). The conjugation of ubiquitin requires the activities of ubiquitin-activating (E1) and conjugating (E2) enzymes. It is suggested that UBC9 might play a role in DNA repair and perhaps even in aging.

C-C Motif Chemokine 18 (CCL18) is secreted protein that belongs to the intercrine beta (chemokine CC) family. CCL18 is expressed at high levels in the lung, lymph nodes, placenta, bone marrow, and dendritic cells. CCL18 is a chemotactic factor that attracts lymphocytes but not monocytes or granulocytes. CCL18 may be involved in B-cell migration into B-cell follicles in lymph nodes. CCL18 attracts naive T-lymphocytes toward dendritic cells and activated macrophages in lymph nodes. It has chemotactic activity for naive T-cells, CD4+ and CD8+ T-cells and thus may play a role in both humoral and cell-mediated immunity responses.

Gamma-Aminobutyric Acid Receptor-Associated Protein (GABARAP) is a ligand-gated chloride channel protein that mediates inhibitory neurotransmission and belongs to the MAP1 LC3 family. GABARAP is highly positively charged in its N-terminus and shares sequence similarity with light chain-3 of microtubule-associated proteins 1A and 1B. GABARAP clusters neurotransmitter receptors by mediating interaction with the cytoskeleton. Autophagy is the process by which cells recycle cytoplasm and dispose of excess or defective organelles. This process is suggested to be involved development, differentiation, growth regulation and tissue remodeling in multicellular organisms. An important inhibitory neurotransmitter, GABA, acts on GABA receptors that are ubiquitously expressed in the CNS. GABARAP has been shown to play a important role in intracellular transport of GABA(A) receptors and its interaction with the cytoskeleton.

Cluster of differentiation 14 (CD14), is a cell surface glycoprotein, and is a is a component of the innate immune system. CD14 is a myelomonocytic differentiation antigen preferentially expressed on monocytes, macrophages, and activated granulocytes. CD14 exists in two forms. Either it is anchored into the membrane by a glycosylphosphatidylinositol tail (mCD14) or it appears in a soluble form (sCD14). Soluble CD14 either appears after shedding of mCD14 (48 kDa) or is directly secreted from intracellular vesicles (56 kDa). CD14 acts as a co-receptor (along with the Toll-like receptor TLR 4 and MD-2) for the detection of bacterial lipopolysaccharide (LPS). CD14 can bind LPS only in the presence of lipopolysaccharide-binding protein (LBP). CD14 has been proposed to be involved in various biological processes, including transportation of other lipids, cell-cell interaction during different immune responses, as well as recognition of apoptotic cells. Although LPS is considered its main ligand, CD14 also recognizes other pathogen-associated molecular patterns. CD14+ cells are monocytes that can differentiate into a host of different cells. CD14 has been shown to interact with Lipopolysaccharide-binding protein.

Cytotoxic Tlymphocyte 4(CTLA-4,CD152), is a type I transmembrane T cell inhibitory molecule that is a member of the Ig superfamily. Human or mouse CTLA4 cDNA encodes 223 amino acids (aa) including a 35 aa signal sequence, a 126 aa extracellular domain (ECD) with one Ig-like V-type domain, a 21 aa transmembrane (TM) sequence, and a 41 aa cytoplasmic sequence. It is widely expressed with highest levels in lymphoid tissues. CD28 and CTLA-4, together with their ligands, B7-1 and B7-2, constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD28 and CTLA-4 are structurally homologous molecules that are members of the immunoglobulin (Ig) gene superfamily. CTLA4 transmits an inhibitory signal to T cells, whereas CD28 transmits a stimulatory signal. Intracellular CTLA4 is also found in regulatory T Cells and may play an important role in their functions. Tcell activation through the Tcell receptor and CD28 leads to increased expression of CTLA4.

Ly6/PLAUR domain containing3 (LYPD-3) is a GPI-linked protein. The structure of LYPD-3 is similar to the urokinasetype plasminogen activator receptor (uPAR). The interaction of LYPD-3 with Laminin 1 and 5 on neighboring cells promotes the adhesion, spreading, and migration of tumor cells. CD27 (TNFRSF7) is a member of the TNF-receptor superfamily limited to cells of the lymphoid lineage and exists as both a dimeric glycoprotein on the cell surface and as a soluble protein in serum. As a T and B cell co-stimulatory molecule, the activity of CD27 is governed by its TNF-like ligand CD70 on lymphocytes and dendritic cells. The CD27-CD70 interaction is required for Th1 generation responses to differentiation signals and long-term maintenance of T cell immunity, and meanwhile, plays a key role in regulating B cell differentiation, activation and immunoglobulin synthesis. The CD27 receptor transduces signals and subsequently leads to the activation of NF-kappaB and MAPK8/JNK, mediated by the adaptor proteins TRAF2 and TRAF5. In addition, the proapoptotic protein SIVA is capable of binding the cytoplasmic tail of CD27 and exerts action in the process of apoptosis.

CCL4, (C-C motif chemokine 4; macrophage inflammatory protein 1beta) is a chemokine secreted at sites of inflammation by activated leukocytes, lymphocytes, vascular endothelial cells and pulmonary smooth cells. CCL4 attracts a variety of immune cells to sites of microbial infection as well as to pathologic inflammation, such as allergic asthma and ischemic myocardium. Full-length and truncated CCL4 exert biological activity through the receptor CCR5. The truncated form of CCL4 interacts additionally with CCR1 and CCR2b. The ability of CCL4 to bind CCR5 inhibits the cellular entry of M-tropic HIV. Both CCL4 forms block HIV-1 infection of T cells by inducing the downregulation of CCR5.
