Description:
Lipocalin-2, also known as Neutrophil Gelatinase-Associated Lipocalin (NGAL), is a secretory protein of the lipocalin superfamily. Lipocalin-2 contains a signal peptide that enables it to be secreted and form complexes with matrix metalloproteinase-9 (MMP-9) through disulphide bonds. Similar to other lipocalin family members, Lipocalin-2 is involved in diverse cellular processes, including the transport of small hydrophobic molecules, protection of MMP-9 from proteolytic degradation, and cell signalling. Furthermore, Lipocalin-2 can tightly bind to bacterial siderophore through a cell surface receptor, possibly serving as a potent bacteriostatic agent by sequestering iron, regulating innate immunity and protecting kidney epithelial cells from ischemia–reperfusion injury. This protein is mainly expressed in neutrophils and in lower levels in the kidney, prostate, and epithelia of the respiratory and alimentary tracts. Recent evidence also suggests its role as a biomarker for renal injury and inflammation.

Leptin is a hormone secreted from white adipocytes and plays important role in the regulation of food intake and energy balance. Leptin functions via signalling pathways involving OB-R in hypothalamus. In mammals, leptin is mainly produced by the adipose tissue and encodes body fat reserves, acting as a short-term satiety signal. In fish, the presence of a leptin-like peptide was first evidenced by immuno-cross-reactivity [14], and its existence was certainly demonstrated after the finding by synteny of a leptin sequence in the pufferfish.

The bone morphogenetic protein receptor, type IA is also known as BMPR1A or ALK3 is a protein which in humans is encoded by the BMPR1A gene. BMPR1A has also been designated as CD292 (cluster of differentiation 290). The bone morphogenetic protein (BMP) receptors are a family of transmembrane serine/threonine kinases that include the type I receptors BMPR1A (this protein) and BMPR1B and the type II receptor BMPR2. These receptors are also closely related to the activin receptors, ACVR1 and ACVR2. Alk3-mediated BMP signaling in AV endocardial/mesenchymal cells plays a central role during cushion morphogenesis.

EGR-1 belongs to the EGR family of C2H2-type zinc finger proteins. It is a nuclear protein and functions as a transcriptional regulator. EGR-1 recognises and binds to the DNA sequence 5'-CGCCCCCGC-3'(EGR-site).The products of target genes it activates are required for differentiation and mitogenesis. Studies suggest this is a tumour suppressor gene. EGR-1 has a distinct pattern of expression in the brain, and its induction has been shown to be associated with neuronal activity. Several studies suggest it has a role in neuronal plasticity. EGR-1 has also been found to regulate the expression of synaptobrevin II (a protein important for synaptic exocytosis).

Resistin known as adipose tissue-specific secretory factor (ADSF) or C/EBP-epsilon-regulated myeloid-specific secreted cysteine-rich protein (XCP1) that seems to suppress insulin ability to stimulate glucose uptake into adipose cells. The length of the resistin pre-peptide in human is 108 amino acid residues and in the mouse and rat it is 114 aa; the molecular weight is ~12.5 kDa. Resistin is a cytokine whose physiologic role has been the subject of much controversy regarding its involvement with obesity and type II diabetes mellitus (T2DM). Resistin has been shown to cause "high levels of 'bad' cholesterol (low-density lipoprotein or LDL), increasing the risk of heart disease, resistin increases the production of LDL in human liver cells and also degrades LDL receptors in the liver. Potentially links obesity to diabetes.

ATG5 is an E2 ubiquitin ligase which is necessary for autophagy. Its expression is a relatively late event in the apoptotic process, occurring downstream of caspase activity, dramatically highly expressed in apoptotic cells. It is activated by ATG7, conjugates to ATG12 and associates with isolation membrane to form cup-shaped isolation membrane and autophagosome. The conjugate complex detaches from the membrane immediately before or after autophagosome formation is completed. ATG5 plays an important role in the apoptotic process, possibly within the modified cytoskeleton.

Tnfrsf1a is one of the major receptors for the tumour necrosis factor-alpha. It can activate the transcription factor NF-κB, mediate apoptosis, and function as a regulator of inflammation. Antiapoptotic protein BCL2-associated athanogene 4 (BAG4/SODD) and adaptor proteins TRADD and TRAF2 have been shown to interact with this receptor, and thus play regulatory roles in the signal transduction mediated by the receptor. Germline mutations of the extracellular domains of this receptor were found to be associated with the human genetic disorder called tumour necrosis factor associated periodic syndrome (TRAPS) or periodic fever syndrome

UBE2M is a member of the E2 ubiquitin-conjugating enzyme family. Ubiquitination involves at least three classes of enzymes: ubiquitin-activating enzymes, or E1s, ubiquitin-conjugating enzymes, or E2s, and ubiquitin-protein ligases, or E3s. This protein is linked with a ubiquitin-like protein, NEDD8, which can be conjugated to cellular proteins, such as Cdc53/culin. UBE2M accepts the ubiquitin-like protein NEDD8 from the UBA3-NAE1 E1 complex and catalyses its covalent attachment to other proteins. The specific interaction with the E3 ubiquitin ligase RBX1, but not RBX2, suggests that the RBX1-UBE2M complex neddylates specific target proteins, such as CUL1, CUL2, CUL3 and CUL4. It involved in cell proliferation and is an important cellular mechanism for targeting abnormal or short-lived proteins for degradation.

Calcitonin is a secreted protein which belongs to the calcitonin family. Calcitonin is cleaved into the following two chains: Calcitonin and Katacalcin. Katacalcin is a potent plasma calcium-lowering peptide. Calcitonin is a 32-amino acid linear polypeptide hormone. Calcitonin acts to reduce blood calcium (Ca2+), opposing the effects of parathyroid hormone (PTH). Its importance in humans has not been as well established as its importance in other animals, as its function is usually not significant in the regulation of normal calcium homeostasis. Calcitonin causes a rapid but short-lived drop in the level of calcium and phosphate in blood by promoting the incorporation of those ions in the bones.

RAC-alpha serine/threonine-protein kinase (AKT1) is also known PKB, Protein kinase B alpha, PKB alpha, Proto-oncogene c-Akt and RAC-PK-alpha, which belongs to the protein kinase superfamily, AGC Ser/Thr protein kinase family and RAC subfamily and is expressed in prostate cancer and levels increase from the normal to the malignant state (at protein level). AKT1 is one of 3 closely related serine/threonine-protein kinases (AKT1, AKT2 and AKT3) called the AKT kinase, and which regulate many processes including metabolism, proliferation, cell survival, growth and angiogenesis. AKT is responsible of the regulation of glucose uptake by mediating insulin-induced translocation of the SLC2A4/GLUT4 glucose transporter to the cell surface. AKT regulates also the storage of glucose in the form of glycogen by phosphorylating GSK3A at 'Ser-21' and GSK3B at 'Ser-9', resulting in inhibition of its kinase activity. Phosphorylation of GSK3 isoforms by AKT is also thought to be one mechanism by which cell proliferation is driven. AKT regulates also cell survival via the phosphorylation of MAP3K5 (apoptosis signal-related kinase).

The cytokine thymic stromal lymphopoietin receptor (TSLPR) is consisting of a common gamma receptor–like chain (TSLPR- gamma ) and a common interleukin 7 (IL-7) R alpha chain that belongs to the type 1 cytokine receptor family. Transfection of TSLPR cDNA result in only low affinity binding, while cotransfection of the IL-7R alpha chain cDNA shows high affinity binding. TSLP and TSLPR play a critical role in the initiation of allergic diseases in mice. The TSLP R cDNA encodes a transmembrane receptor containing 370 amino acids (aa) with two potential N-linked glycosylation sites and a cytoplasmic domain of 104 aa including a single tyrosine residue. TSLPR can mediate signalling of the signal transducer and activator of transcription 5 (Stat5) by TSLP. TSLP R is broadly expressed in the immune and hematopoietic cells, particularly in hematopoietic progenitors and myeloid cells.

The tetrameric receptor complex for IFN gamma consists of two subunits, IFNGR1 (IFN gamma R alpha) and IFNGR2 (IFN gamma R beta ), through which the dimeric IFN- gamma exerts its biological functions, including antiviral, antiproliferation and immune-modulatory activity in mammals. Both IFNGR1 and IFNGR2 are single transmembrane proteins belonging to the class II cytokine family. FNGR1, widely expressed in most host cells, is essential for IFN gamma binding, receptor trafficking, and signal transduction. IFNGR1 possesses an intracellular Janus tyrosine kinase (JAK) 1 binding site, a signal transducer and activator of transcription 1 (STAT1) binding site. The resulting STAT1 homodimers translocate from the cytoplasm to the nucleus and bind to the interferon-gamma activated sequence (GAS) promoter to induce expression of downstream interferon stimulated genes (ISGs).

Transforming growth factor beta 2 (TGF- beta 2) is a member of TGF-beta superfamily that shares a characteristic cysteine knot structure. Mice with TGF- beta 2 gene deletion show defects in development of cardiac, lung, craniofacial, limb, spinal column, eye, inner ear and urogenital systems. All TGF- beta isoforms signal via the same heteromeric receptor complex, consisting of a ligand binding TGF- beta receptor type II (T beta R-II), and a TGF- beta receptor type I (T beta R-I). Signal transduction from the receptor to the nucleus is mediated via SMADs. TGF- beta expression is found in cartilage, bone, teeth, muscle, heart, blood vessels, haematopoitic cells, lung, kidney, gut, liver, eye, ear, skin, and the nervous system.

Proprotein convertase subtilisin/kexin type 9 (PCSK9) is also known as NARC1 (neural apoptosis regulated convertase), is a newly identified subtilase belonging to the peptidase S8 subfamily. Mouse PCSK9 is synthesized as a soluble zymogen, and undergoes autocatalytic intramolecular processing in the endoplasmic reticulum, resulting in the cleavage of its propeptide that remains associated with the secreted active enzyme with a broad alkaline pH optimum. This protein plays a major regulatory role in cholesterol homeostasis. PCSK9 binds to the epidermal growth factor-like repeat A (EGF-A) domain of the low-density lipoprotein receptor (LDLR), inducing LDLR degradation. PCSK9 may also have a role in the differentiation of cortical neurons. Mutations in this gene have been associated with a rare form of autosomal dominant familial hypercholesterolemia (HCHOLA3).

Programmed cell death 1 ligand 1 (PD-L1) is also known as cluster of differentiation (CD274) or B7 homolog 1 (B7-H1), is a member of the growing B7 family of immune molecules and is involved in the regulation of cellular and humoral immune responses. B7-H1 is a cell surface immunoglobulin superfamily with two Ig-like domains within the extracellular region and a short cytoplasmic domain. PD-L1 is highly expressed in the heart, skeletal muscle, placenta and lung and weakly expressed in the thymus, spleen, kidney and liver. PD-L1 is expressed on activated T-cells, B-cells, dendritic cells, keratinocytes and monocytes. PD-L1 is up-regulated on T- and B-cells, dendritic cells, keratinocytes and monocytes after LPS and IFNG activation and up-regulated in B-cells activated by surface Ig cross-linking. PD-L1 involve in the costimulatory signal, essential for T-cell proliferation and production of IL10 and IFNG, in an IL2-dependent and a PDCD1-independent manner.
