Description:
Tissue inhibitor of metalloproteinases 1 (TIMP1) and Tissue inhibitor of metalloproteinases 2 (TIMP 2) have similar properties, specifically in inhibiting enzymes of the matrix metalloproteinase family, and are thought to be of great importance in the maintenance of connective tissue integrity. TIMP1 forms a complex of 1:1 stoichiometry with activated interstitial collagenases, activated stromelysin, active form of 72kDa Type IV collagenase (also known as MMP2 or gelatinase A), and latent and active forms of 92kDa Type IV collagenase (also known as MMP0 or gelatinase B). TIMPs inhibit the proteolytic invasiveness of tumour cells and normal placental trophoblast cells. TIMP1 is produced in low (pg/mL) levels by most cell types. Treatment of cells with the phorbol ester TPA stimulates production of TIMP1 in some cell types, but the low protein levels produced often require concentration of cell culture media to visualize the bands by Western Blotting.

C-X-C chemokine receptor type 4 is also known as fusin or CD184 (cluster of differentiation 184), CXCR4, CD184, D2S201E, FB22, HM89, HSY3RR, LAP3, LCR1, LESTR, NPY3R, NPYR, NPYRL, NPYY3R or WHIM. CXCR-4 is an alpha-chemokine receptor specific for stromal-derived-factor-1 (SDF-1 also called CXCL12), a molecule endowed with potent chemotactic activity for lymphocytes. This receptor is one of several chemokine receptors that HIV isolates can use to infect CD4+ T cells. HIV isolates that use CXCR4 are traditionally known as T-cell tropic isolates. Typically, these viruses are found late in infection. It is unclear as to whether the emergence of CXCR4 using HIV is a consequence or a cause of immunodeficiency.CXCR4 is upregulated during the implantation window in natural and hormone replacement therapy cycles in the endometrium, producing, in presence of a human blastocyst, a surface polarization of the CXCR4 receptors suggesting that this receptor is implicated in the adhesion phase of human implantation. SDF-1 and CXCR4 were believed to be a relatively "monogamous" ligand-receptor pair (other chemokines tend to use several different chemokine receptors in a fairly "promiscuous" manner). Recent evidence demonstrates ubiquitin is also a natural ligand of CXCR4. Chronic exposure to THC increased T lymphocyte CXCR4 expression on both CD4+ and CD8+ T lymphocytes. Drugs that block the CXCR4 receptor appear to be capable of "mobilizing" hematopoietic stem cells into the bloodstream as peripheral blood stem cells.

Sialate O-Acetylesterase (SIAE) belongs to the family of hydrolases, specifically those acting on carboxylic ester bonds. SIAE is widely expressed with high expression levels in the testis, prostate, and colon. SIAE catalyses N-acetyl-O-acetylneuraminate and H2O to N-acetylneuraminate and acetate. SIAE removes O-acetyl ester groups from position 9 of the parent sialic acid, N-acetylneuraminic acid. SIAE down-regulates B lymphocyte antigen receptor signaling (involving CD22), and is required for immunological tolerance. Loss of function mutations in SIAE are much more frequently found in humans with autoimmune diseases especially rheumatoid arthritis and type 1 diabetes.

Dentin Matrix Acidic Phosphoprotein 1 (DMP-1) is an extracellular matrix protein and a member of the small integrin binding ligand N-linked glycoprotein family. DMP-1 is expressed in teeth particularly in odontoblast, ameloblast, and cementoblast. DMP-1 is critical for proper mineralization of bone and dentin. DMP-1 may have a dual function during osteoblast differentiation. In the nucleus of undifferentiated osteoblasts, the unphosphorylated form of DMP-1 acts as a transcriptional component for activation of osteoblast-specific genes like osteocalcin. During the osteoblast to osteocyte transition phase, DMP-1 is phosphorylated and exported into the extracellular matrix, where it regulates nucleation of hydroxyapatite. DMP-1 mutations have also been shown to cause rickets hypophosphatemic autosomal recessive type 1 (ARHR1).

Interleukins (ILs) are a large group of cytokines that are produced mainly by leukocytes, although some are produced by certain phagocytes and auxiliary cells. ILs have a variety of functions, but most function to direct other immune cells to divide and differentiate. Each IL acts on a specific, limited group of cells through a receptor specific for that IL. Human IL10 is a non glycosylated polypeptide consisting of 178 amino acids. There is 73% homology between the human and mouse IL10 proteins, however, the human IL10 acts on both human and mouse target cells, while the mouse IL10 has species specific activity. The cellular sources of IL10 are CD4+ T cells and T cell clones, thymocytes, B cells and B cell lymphomas, macrophages, mast cell lines and keratinocytes. IL10 will stimulate the growth of stem cells, mast cells and thymocytes. IL10 enhances cytotoxic T cell development, and costimulates B cell differentiation and immunoglobulin secretion. SLAM Family Member 6 (SLAMF6) is a 60 kD single-pass type I membrane protein that belongs to the SLAM subgroup of the CD2 family. Human SLAMF6/ NTB-A contains a 205 amino acid extracellular domain (ECD) with one Ig-like V-set and one Ig-like C2-set domain, a 21 amino acid transmembrane segment and an 84 amino acid cytoplasmic domain, with two immunoreceptor tyrosine-based switch motifs. SLAMF6 is a homodimer. SLAMF6 can interact with PTN6 and, upon phosphorylation, with PTN11 and SH2D1A/SAP. BCL2 family members form hetero- or homodimers and act as anti- or pro-apoptotic regulators that are involved in a wide variety of cellular activities. This protein forms a heterodimer with BCL2, and functions as an apoptotic activator. This protein is reported to interact with, and increase the opening of, the mitochondrial voltage-dependent anion channel (VDAC), which leads to the loss in membrane potential and the release of cytochrome c. BAX expression is regulated by the tumor suppressor P53 and has been shown to be involved in P53-mediated apoptosis.
