CDKN2B's gene lies adjacent to the tumor suppressor gene CDKN2A in a region that is frequently mutated and deleted in a wide variety of tumors. CDKN2B is a cyclin-dependent kinase inhibitor, which forms a complex with CDK4 or CDK6, and prevents the activation of the CDK kinases, thus the encoded protein functions as a cell growth regulator that controls cell cycle G1 progression. The expression of this gene was found to be dramatically induced by TGF beta, which suggested its role in the TGF beta induced growth inhibition.

RIG-I (retinoic acid-inducible gene I; Ddx58) and MDA5 (melanoma differentiation-associated gene 5, also known as Ifih1 or Helicard) are proteins that sense viral replication intermediates, such as double-stranded RNA and triggers the host antiviral programs. These molecules signal the downstream activation of NF-kappaB and IFN regulatory factor (IRF) -3, which coordinately regulate the expression of type-I interferons. Cardif (also called VISA/IPS-1/MAVS) is a new CARD (caspase activation and recruitment domain)-containing adaptor protein that interacts with the CARD domain of RIG-I and MDA5, leading to the activation of NF-kappaB and IRF3. Cardif is located to the mitochondrial outer membrane. Removal of the mitochondrial-targeting domain of cardif abolishes its ability to induce IFNs. Cardif is cleaved and inactivated by NS3-4A, a serine protease from hepatitis C virus known to block interferon-beta production.

SGKL/SGK3, also known as CISK for cytokine-independent survival kinase, is a SGK type protein kinase. The protein contains a lipid-binding phox homology (PX) domain, which is required for targeting SGKL/SGK3 protein to the endosomal compartment. SGKL/SGK3 functions downstream of the phosphoinositide 3-kinase (PI3K) cascade and has been implicated in cell survival. SIRT5 is a human member of a family of proteins called Sirtuins (Sir2-like proteins) and are present in prokaryotes and eukaryotes. All Sir2-like proteins have a sirtuin core domain, which contains a series of sequence motifs conserved in organisms ranging from bacteria to humans. Bacterial, yeast and mammalian sirtuins are able to metabolize NAD and possibly at as mono-ADP-ribosyltransferases. The enzymatic function of sirtuins is not yet completely understood but recent reports of histone-activated Sir2-mediated NAD metabolism and NAD-activated Sir2-mediated histone deacetylation suggest a possible coupled reciprocal activation mechanism involving interactions of Sir2 with NAD and the N epsilon-acetyl-lysine groups of acetylated histones.

In mammals, there are several identified isoforms of p21-activated protein kinases or PAKs: alpha-PAK (also known as PAK-1) and beta-PAK (also known as PAK-3) are mostly brain-specific, while -PAK (also known as PAK-2) is expressed ubiquitously (Jakobi et al., 2003). Mutations of the gene coding for PAK-3 are associated with X-linked mental retardation and recent work indicates that PAK-3 is a key regulator of synapse formation and plasticity in the hippocampus (Boda et al., 2004). PAK-3 is thought to play a key role in regulation of cell shape and motility as well as cell death (Jakobi et al., 2003; Walter et al., 1998). IKK epsilon Antibody: Nuclear factor kappa B (NF-kappa B) is a ubiquitous transcription factor and an essential mediator of gene expression during activation of immune and inflammatory responses. NF-kappa B mediates the expression of a great variety of genes in response to extracellular stimuli. NF-kappa B is associated with I kappa B proteins in the cell cytoplasm, which inhibit NF-kappa B activity. I kappa B is phosphorylated by I kappa B kinase (IKK) complex that contains IKK alpha , IKK beta , and IKK gamma. A novel molecule in the IKK complex was recently identified and designated IKK epsilon /IKK-i. IKK epsilon is required for the activation of NF-kappa B by PMA and T cell receptors but not by TNF alpha and IL-1. IKK epsilon /IKK-i message is expressed in a variety of tissues and is inducible by TNF alpha , IL-1, and LPS.
