CD152 (CTLA-4) and CD28, together with their ligands B7-1 and B7-2, constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD152 and CD28 are structurally
homologous molecules that are members of the immunoglobulin (Ig) gene superfamily. Both CD152 and CD28 are composed of a single Ig V-like extracellular domain, a transmembrane domain and an intracellular domain. CD152 and CD28 are both expressed on the cell surface as disulfide-linked homodimers or as monomers. CD152 was originally identified as a gene that was specifically expressed by cytotoxic T lymphocytes. However, CD152 transcripts have since been found in both Th1 and Th2, and CD4+ and CD8+ T cell clones. Whereas, CD28 expression is constitutive on the surfaces of 95% of CD4+ T cells and 50% of CD8+ T cells and is down regulated upon T cell activation, CD152 expression is upregulated rapidly following T cell activation and peaks approximately 24 hours following activation. Although both CD152 and CD28 can bind to the same ligands, CD152 binds to B71 and B72 with 20-100-fold higher affinity than CD28.

CD200, also known as OX-2, is a 45 kDa transmembrane immunoregulatory protein that belongs to the immunoglobulin superfamily. CD200 is widely but not ubiquitously expressed. Its receptor (CD200R) is restricted primarily to mast cells, basophils, macrophages, and dendritic cells, which suggests myeloid cell regulation as the major function of CD200. CD200 knockout mice are characterised by increased macrophage number and activation and are predisposed to autoimmune disorders. In T cells, CD200 functions as a costimulatory molecule independent of the CD28 pathway. Several viruses encode CD200 homologs which are expressed on infected cells during the lytic phase. Like CD200 itself, viral CD200 homologs also suppress myeloid cell activity, enabling increased viral propagation.

The most prominent members of the interleukin-1 (IL-1) superfamily are IL-1alpha and IL-1beta. They lack a signal peptide and are secreted by an unconventional, endoplasmic reticulum-Golgi-independent mechanism. IL-1alpha was reported to be more widely and constitutively expressed and has intracellular functions, but also acts locally in a membrane-bound form by activating IL-1R1. Additionally, passive release of IL-1alpha upon cell death can trigger a sterile inflammatory response to dying cells. The cleavage of IL-1alpha is not mediated by caspase-1 and is not required for binding to IL-1R1. Recently it has been observed that all activators of the inflammasome NLRP3/NALP3 induce the simultaneous secretion of IL-1alpha and IL-1beta. Although most activators fully rely on the inflammasome for IL-1alpha secretion, some induce the processing and secretion of IL-1alpha in an inflammasome-independent manner.

B18R, also known as B19R in the Copenhagen strain of Vaccinia, is a type I interferon (IFN)-binding protein encoded by the B18R open reading frame in the Western Reserve strain of vaccinia virus. B18R exists in two forms, soluble and membrane-bound form and has a broad species specificity. B18R has high affinity for human IFN-alpha and binds rabbit, bovine, rat, pig, and mouse IFN-alpha and IFN-beta. Secreted B18R binds to uninfected and infected cells. B18R presents at the cell surface and protects cells from the antiviral state. It has shown that binding of soluble recombinant B18R protects cultured cells from IFN and allows vaccinia virus replication.

Complement Factor H-Related 1 (CFHR1) is a 43 kDa secreted member of the factor H family of glycoproteins. The human Complement Factor H protein family consists of the complement and immune regulators factor H, the factor H-like protein 1 (FHL-1) and five factor H-related proteins (CFHR-1 to -5). Members of the H-related protein family are exclusively composed of individually folded protein domains, termed short consensus repeats (SCRs) or complement control modules. FHR1 is produced by hepatocytes and circulates as two differentially glycosylated isoforms (37 kDa and 43 kDa). Mature human FHR1 is 312 amino acids in length. It contains five, approximately 60 aa SCRs that basically constitute the entire molecule. In response to the presence of allergens, CCL11 selectively recruits eosinophils, a prominent feature of allergic inflammatory reactions. The effects of CCL11 are mediated by its binding to a G-protein-linked receptor known as a chemokine receptor. Chemokine receptors for CCL11 include CCR2, CCR3 and CCR5. Limitin has approximately 30% sequence homology with IFN- alpha, IFN- beta , and IFN-ω and binds to the IFN- alpha/ beta receptors. Activin Receptor Type-2A is a protein that in humans is encoded by the ACVR2A gene. ACVR2A is an activin type 2 receptor. This gene encodes activin A type II receptor. Activins are dimeric growth and differentiation factors which belong to the transforming growth factor-beta (TGF-beta) superfamily of structurally related signaling proteins. Activins signal through a heteromeric complex of receptor serine kinases which include at least two type I (I and IB) and two type II (II and IIB) receptors. These receptors are all transmembrane proteins, composed of a ligand-binding extracellular domain with cysteine-rich region, a transmembrane domain, and a cytoplasmic domain with predicted serine/threonine specificity. Tumour necrosis factor receptor superfamily member 9(TNFRSF9), also known as CD137 and 4-1BB, is an inducible T cell surface protein belonging to the tumour necrosis factor receptor superfamily. It is a single-pass type I membrane protein which contains 4 TNFR-Cys repeats. The human and mouse proteins share 60% amino acid sequence identity. CD137 is expressed by mesenchymal cells, including endothelial cells, chondrocytes, and cells of the central nervous system. CD137 is also broadly expressed by cells of the human immune system, is broadly expressed by cells of the human immune system, including activated CD8+ and CD4+ T cells, activated natural killer (NK) cells, follicular dendritic cells (FDCs) and monocytes.
