Description:   Mouse ULBP1, also known as RAET1I and NKG2DL1, is a member of the ULBP/RAET1 gene family. ULBP1 plays an important role in immune responses, especially in cancer and infectious diseases, and is well-known to bind to NKG2D together with at least ULBP 2 and 3. These proteins are distantly related to major histocompatibility class I (MHC I) molecules, possessing the alpha 1 and alpha 2 Ig-like domains, but lacking the alpha 3 domain. Unlike MHC Class I, they have no capacity to bind peptide or interact with beta2-microglobulin. It can activate multiple signalling pathways in primary NK cells, gamma delta T cells, and CD8+ alpha beta T cells, resulting in the production of cytokines and chemokines.ULBP1 is expressed in wide range of tissues including heart, brain, lung, liver, bone marrow and some tumour cells, T-cells, B-cells, As an unconventional member of the MHC class I family, ULBP1 is able to interact with soluble CMV glycoprotein UL16 in CMV infected cells. The interaction with UL16 blocked the interaction with the NKG2D receptor, and thus might escape the immune surveillance. Furthermore, UL16 also causes ULBP1 to be retained in the ER and cis-Golgi apparatus so that it does not reach the cell surface. The ULBP1 regulation may have implications for development of new therapeutic strategies against cancer cells.

Description:   Mouse Legumain,also known as LGMN, is a cysteine protease belonging to peptidase family C13 and is expressed in kidney and placenta abundantly. LGMN has a strict specificity for hydrolysis of asparaginyl bonds. It can also cleave aspartyl bonds slowly, especially under acidic conditions. The mammalian legumain is involved in the processing of proteins for MHC class II antigen presentation in the lysosomal/endosomal system. It plays a role in the regulation of cell proliferation via its role in EGFR degradation. Legumain deficiency causes the accumulation of pro-Cathepsins B, H and L, another group of lysosomal cysteine proteases. Overexpression of Legumain in tumours is significant for invasion/metastasis. Mammalian legumain is inhibited by iodoacetamide and maleimides. Legumain activation appears to be autocatalytic and can be triggered by acidic pH.

Description:   Carcinoembryonic antigen-related cell adhesion molecules (CEACAMs) belong to a group of mammalian immunoglobulin related glycoproteins. They play critical roles in cell–cell recognition. CEACAM5, also called CEA and CD66e, is characterised by having seven extracellular Ig domains and a glycosylphosphatidylinositol (GPI) anchor. CEACAM5 is expressed primarily by epithelial cells, and functions as a calcium-independent adhesion molecule through homophilic and heterophilic interactions with CEACAM1. Studies have shown that CEACAM5 is overexpressed in a majority of carcinomas, and its overexpression can protect tumour cells from apoptosis. It is commonly used as a cancer marker.

Description:   Human serum albumin (HSA), the most abundant protein in human blood plasma, is essential for maintaining osmotic pressure. It is produced in the liver, consists of a single polypeptide chain, with 585 amino acids with 17 tyrosil residues and one tryptophan located in position 214. HSA is organised in three domains, I, II and III, each consisting of two subdomains, A and B. In the physiological states, HSA occurs in two forms – the non-modified HSA with a free thiol group of Cys-34, and the modified (oxidised) form (oHSA), also called human mercaptoalbumin (HMA) and human nonmercaptoalbumin (HNA), respectively. HMA and HNA are in equilibrium, depending on the redox state of Cys-34, and their ratio also varies depending on age and the diseased state. HSA functions primarily as a carrier protein for drugs, steroids, fatty acids, and thyroid hormones, and plays a role in stabilising extracellular fluid volume.

Description:   Pre-B cell colony enhancing factor (PBEF) was originally identified as a cytokine that potentiated the clonal expansion and differentiation of pre-B cells, but it is also acknowledged to be the ubiquitous intracellular enzyme nicotinamide phosphoribosyltranferase (NAMPT) and the adipokine “visfatin”. PBEF is constitutively expressed in the fetal membranes where its greatest expression is in the amnion. It has intracellular and extracellular forms. Most of the intracellular functions of PBEF are due to its role as a Nampt which can induce angiogenesis through upregulation of VEGF and VEGFR and secretion of MCP-1. Extracellular PBEF has been shown to increase inflammatory cytokines, such as TNF- alpha, IL-1 beta , IL-16, and TGF- beta 1. PBEF also increases the production of IL-6, TNF- alpha, and IL-1 beta in CD14+ monocyctes, macrophages, and dendritic cells, enhances the effectiveness of T cells.

Description:   Interleukin-16 (IL-16) is a CD8+ T cell-derived cytokine that induces chemotaxis of CD4+ T cells and CD4+ monocytes and eosinophils. Analysis by gel filtration suggests that, under physiological conditions, human IL-16 exists predominantly as a noncovalently linked multimer, but that some IL-16 may exist as a monomer. However, only the multimeric form appears to possess chemotactic activity, suggesting that receptor cross-linking may be required for activity. IL-16 also induces expression of IL-2 receptor (IL-2R) and MHC class II molecules on CD4+ T cells. Human and murine IL-16 show significant cross-species reactivity.

Description:   Killer cell immunoglobulin-like receptor 2DL4(KIR2DL4) is a Single-pass type I membrane protein and contains 2 Ig-like C2-type (immunoglobulin-like) domains.It belongs to the immunoglobulin superfamily. KIR2DL4 is expressed in all NK cells and some T cells. KIR2DL4 activates the cytotoxicity of NK cells, despite the presence of an immunoreceptor tyrosine-based inhibition motif (ITIM) in its cytoplasmic tail. The ITIM was not necessary for activation of lysis by KIR2DL4. The activation signal of KIR2DL4 was sensitive to inhibition by another ITIM-containing receptor. The activation-deficient mutant of KIR2DL4 inhibited the signal delivered by the activating receptor CD16.
