ZNHIT1 belongs to the ZNHIT1 family and contains one HIT-type zinc finger. It can be phosphorylated on Thr by MAPK11 or MAPK14. ZNHIT1 is a component of the chromatin-remodeling SRCAP complex, which is composed of at least SRCAP, DMAP, RUVBL1, RUVBL2, ACTL6A, YEATS4, ACTR6 and ZNHIT1. ZNHIT1 may play a role in p53-mediated apoptosis induction.

Description:
ST2, also called IL-1 R4, is an Interleukin-1 receptor family glycoprotein that plays a role in Th2 immune responses. ST2 is expressed on the surface of mast cells, activated Th2 cells, macrophages, and cardiac myocytes. This receptor is very similar to the IL-1 receptor type I and the IL-18 receptor alpha chain in that ST2 also has three extracellular Ig domains and an intracellular Toll domain. ST2 binds IL-33, enhances inflammatory cytokines by activating nuclear factor-κB (NF-κB) and mitogen activated protein (MAP) kinases. ST2 exists as either a membrane bound form (ST2L) or as a soluble form (sST2). ST2L acts as a transmembrane signalling receptor for IL-33 by mediating the effect of IL-33 on the inflammatory process, while sST2 can suppress IL-33 activity.

Thymic stromal lymphopoietin (TSLP) is a novel member of the hemopoietic cytokine family that promotes the development of B cells and shares overlapping activity with IL-7. The human TSLP protein comprises a 28 amino acids (aa) signal sequence and 131 aa mature region. Human TSLP has two isoforms lfTSLP (159 aa) and sfTSLP (63 aa) produced by alternative splicing . lfTSLP (159aa) is expressed in a number of tissues including heart, liver and prostate, and sfTSLP (63aa) is predominantly expressed in keratinocytes of oral mucosa, skin and in salivary glands. In aa sequence level, Human TSLP displays about 43% identity with mouse TSLP. Thymic stromal lymphopoietin (TSLP) is a cytokine that functions mainly on myeloid cells; it induces the release of T cell-attracting chemokines from monocytes and enhances the maturation of CD11c(+) dendritic cells . TSLP has proliferative effects on the myeloid cell line and may initiate asthma or atopic dermatitis responses by directly activating mast cells . TSLP signals cells via the interleukin-7 receptor- alpha chain (IL-7R alpha), shared with IL-7, together with the TSLP receptor (TSLPR) subunit. Recent studies indicate that TSLP and its receptor are novel therapeutic targets for rheumatoid arthritis , for increased intraarticular TSLP concentrations in patients has caused chemotaxis and activation of arthritogenic T cells.

TNF-related apoptosis-inducing ligand (TRAIL; Apo2L;CD253; TNFSF10) is a type II transmembrane protein of about 34kDa. Like most members of the tumor necrosis factor (TNF) superfamily of cytokines TRAIL can be cleaved at the cell surface by metalloproteases to form a soluble molecule. FGF-2 (Basic fibroblast growth factor) belongs to the FGF superfamily that is made up of 22 different fibroblast growth factor genes. FGF-2 and FGF-1 (acidic FGF) are unique in that they do not follow the conventional signal sequence for secretion. FGF-2 is found in five isoforms via alternative initiation of translation. Signalling of FGF-2 occurs through the high-affinity tyrosine kinase receptors FGFR1-4. FGF-2 is a potent wide-spectrum mitogen whose overexpression is associated with immortalisation and unregulated cell proliferation in many tumors. FGF-2 functions in angiogenesis, cell survival, tissue repair, embryonic development and inflammation. FGF-2 is widely used to maintain the self-renewal of human embryonic stem cells (ESCs) and to induce pluripotent stem cells. FGF-2 is an established neurogenic factor for proliferation and differentiation of multipotent neural stem cells both during development and in the adult mouse brain as well.

Folate receptor alpha(FOLR) belongs to the folate receptor family, and is primarily expressed in tissues of epithelial origin. It is also expressed in kidney, lung and cerebellum. The secreted form is derived from the membrane-bound form either by cleavage of the GPI anchor, or/and by proteolysis catalyzed by a metalloprotease. FOLR1 binds to folate and reduced folic acid derivatives and mediates delivery of 5-methyltetrahydrofolate and folate analogs into the interior of cells. It has high affinity for folate and folic acid analogs at neutral pH. Exposure to slightly acidic pH after receptor endocytosis triggers a conformation change that strongly reduces its affinity for folates and mediates their release. It is required for normal embryonic development and normal cell proliferation.
