Galectin-3 belongs to the lectin family of carbohydrate binding proteins. Galectin-3 is expressed by a wide range of cell types including activated T cells, tumor cells, macrophages, osteoclasts, fibroblasts, and epithelial cells. Galectin-3 has specific binding affinity for beta-galactoside sugar moieties and has functional roles during development, innate immunity, cell apoptosis, and tumor metastasis. Galectin-3 is associated with cancer, heart failure, stroke, and inflammation. The amino acid sequences of human and mouse Galectin-3 proteins share 80% homology.

Macrophage inflammatory protein-3 beta (MIP-3 β), also called CCL19, is a chemokine that is expressed in the thymus, lymph nodes, and activated bone marrow stromal cells. MIP-3 β signals through the G protein-coupled receptor CCR7 to regulate normal lymphocyte recirculation. MIP-3 β also functions during T cell trafficking to the thymus, and in T cell and B cell homing to the lymph nodes and secondary lymphoid organs.

Human Kallikrein 7 is a member of the tissue kallikrein family of extracellular serine proteases that is made up of 15 members. It is predominantly expressed in the skin. A major physiological function of Kallikrein 7 is to regulate the desquamation process (the shedding of corneocytes from the outer layer of the epidermis) through proteolysis of the intercellular adhesive structures between corneocytes. Dysregulation of Kallikrein 7 has been linked to several inflammatory skin diseases including atopic dermatitis, psoriasis, and Netherton syndrome. Studies have shown that Kallikrein 5 is a potential physiological activator for Kallikrein 7. The proform of Kallikrein 7 can be activated by thermolysin.

Mouse Protein Tnfrsf14, is a type I transmembrane protein belonging to the TNF receptor superfamily. It is tumour necrosis factor receptor superfamily member 14 and expressed on the surface of T cells during the resting state. Interaction of HVEM with TNF family member LIGHT co-stimulates T cells and promotes inflammation. HVEM also triggers inhibitory signalling cascade in effector T (Teff) cells and regulatory T cells (Tregs) as a ligand of B and T lymphocyte attenuator. Tnfrsf14 is detected in peripheral blood T cells, B cells, monocytes and in various tissues enriched in lymphoid cells. It has demonstrated that HVEM Ig is able to exert a significant antiviral effect against HSV-1 infection in vivo.

The human CD36 gene encodes a single chain 472 amino acid residue protein containing both an N- and a C-terminal cytoplasmic tail and an extracellular loop.CD36 is found on platelets, erythrocytes, monocytes, differentiated adipocytes, mammary epithelial cells, spleen cells and some skin microdermal endothelial cells. CD36 is a multiligand pattern recognition receptor that interacts with a large number of structurally dissimilar ligands, including long chain fatty acid (LCFA), advanced glycation end products (AGE), thrombospondin-1, oxidized low-density lipoproteins (oxLDLs), high density lipoprotein (HDL), phosphatidylserine, apoptotic cells, beta-amyloid fibrils (fAβ), collagens I and IV, and Plasmodium falciparum infected erythrocytes. CD36 is required for the anti-angiogenic effects of thrombospondin1 In the corneal neovascularisation assay. On binding a ligand the protein and ligand are internalized. This internalisation is independent of macropinocytosis and occurs by an actin dependent mechanism requiring the activation Src-family kinases, JNK and Rho-family GTPases. CD36 ligands have also been shown to promote sterile inflammation through assembly of a Toll-like receptor 4 and 6 heterodimer.

Cluster of Differentiation 86 (CD86) is also known as B-lymphocyte activation antigen B7-2, is a type I membrane protein that is a member of the immunoglobulin superfamily, and is constitutively expressed on interdigitating dendritic cells, Langerhans cells, peripheral blood dendritic cells, memory B cells, and germinal center B cells. Additionally, B72 is expressed at low levels on monocytes and can be upregulated through interferon γ. CD86 is the ligand for two different proteins on the T cell surface: CD28 (for autoregulation and intercellular association) and CTLA-4 (for attenuation of regulation and cellular disassociation). CD86 works in tandem with CD80 to prime T cells. Recent study has revealed that B7-2 promotes the generation of a mature APC repertoire and promotes APC function and survival. Furthermore, the B7 proteins are also involved in innate immune responses by activating NF-κB-signaling pathway in macrophages. CD86 thus is regarded as a promising candidate for immune therapy. CD86+ macrophages in Hodgkin lymphoma patients are an independent marker for potential nonresponse to firstline-therapy.
