CD274, also known as B7-H1 or programmed death ligand 1 (PD-L1), is a 40 kD type I transmembrane protein and a member of the B7 family within the immunoglobulin receptor superfamily. Programmed death-1 ligand-1 (PD-L1, CD274, B7-H1) has been identified as the ligand for the immunoinhibitory receptor programmed death-1(PD1/PDCD1) and has been demonstrated to play a role in the regulation of immune responses and peripheral tolerance. By binding to PD1 on activated T-cells and B-cells, PD-L1 may inhibit ongoing T-cell responses by inducing apoptosis and arresting cell-cycle progression. Accordingly, it leads to growth of immunogenic tumour growth by increasing apoptosis of antigen specific T cells and may contribute to immune evasion by cancers. Transforming growth factor- beta (TGF- beta ) is an essential regulator in the processes of development, cell proliferation, and extracellular matrix deposition. TGF- beta regulates cellular processes by binding to three high-affinity cell surface receptors: TGF- beta receptor type I (TGF- beta -RI), TGF- beta receptor type II (TGF- beta -RII), and TGF- beta beta receptor type III (TGF- beta -RIII). TGF- beta RII is consists of a C-terminal protein kinase domain and an N-terminal ectodomain and belongs to transforming growth factor-beta (TGF- beta ) receptor subfamily. Messenger RNA (mRNA) degradation plays a key role in the control of gene expression in all organisms by limiting the number of times that each mRNA molecule can be used as a template for protein synthesis. RNA pyrophosphohydrolase, also called RppH, is a master regulator of 5'-dependent mRNA decay. It accelerates the degradation of transcripts by removing pyrophosphate from the 5'-end of triphosphorylated RNA, leading to a more labile monophosphorylated state that can stimulate subsequent ribonuclease cleavage. RppH preferentially hydrolyses diadenosine penta-phosphate with ATP as one of the reaction products, and can be able to hydrolyse diadenosine hexa- and tetra-phosphate. NADE Antibody: The p75 neurotrophin receptor (p75NTR) is a member of the tumor necrosis receptor superfamily and can mediate cell death and cell survival in response to nerve growth factor (NGF). The p75NTR-associated cell death executor (NADE) mediates apoptosis by interacting with the cell death domain of p75NTR following the binding of NGF by p75NTR. Recent studies have shown that NADE also interacts with second mitochondria-derived activator of caspase (Smac). Co-expression of NADE and Smac promotes TRAIL-induced apoptosis and inhibits XIAP-mediated Smac ubiquitization. It has been suggested that it is this interaction between NADE and Smac that allows apoptosis to proceed. Finally, although initially discovered as an mRNA expressed in ovarian granulosa cells, NADE has been suggested to play a role in the neuronal death seen in epileptic brain damage.
