NANOG Antibody: Expression of NANOG is required for the maintenance of pluripotency in epiblast and embryonic stem (ES) cells as well as for the ability to maintain ES self-renewal independently of LIF/Stat3. The role of NANOG in embryonic development suggested that it might be useful in the creation of stem cells that might be useful in cell replacement therapies in the treatment of several degenerative diseases. Artificial stem cells, termed induced pluripotent stem (iPS) cells, can be created by expressing POU5F1 (also known as Oct-4), another germline-specific transcription factor, and the transcription factors Sox2, Klf4 and Lin28 along with c-Myc in mouse fibroblasts. More recently, experiments have demonstrated that iPS cells could be generated using expression plasmids expressing NANOG, Sox2, KlfF4 and c-Myc, eliminating the need for virus introduction, thereby addressing a safety concern for potential use of iPS cells in regenerative medicine.

ITGB3BP is a transcription coregulator that can have both coactivator and corepressor functions. Isoform 1, but not other isoforms, is involved in the coactivation of nuclear receptors for retinoid X (RXRs) and thyroid hormone (TRs) in a ligand-dependent fashion. ITGB3BP acts as a transcriptional corepressor via its interaction with the NFKB1 NF-kappa-B subunit, possibly by interfering with the transactivation domain of NFKB1. It induces apoptosis in breast cancer cells, but not in other cancer cells, via a caspase-2 mediated pathway that involves mitochondrial membrane permeabilization but does not require other caspases. ITGB3BP may also act as an inhibitor of cyclin A-associated kinase. ITGB3BP may be involved in incorporation of newly synthesized CENPA into centromeres via its interaction with the CENPA-NAC complex.

AIFM3 Antibody: Apoptosis, also known as programmed cell death, plays major roles in development and normal tissue turnover in addition to tumor formation. Recently a protein similar to the apoptosis-inducing factor (AIF) was cloned and designated AIFL (also known as AIFM3). AIFM3 is expressed ubiquitously and is predominantly localized to the inner membranes of mitochondria. Unlike AIF, AIFM3 does not translocate to the nucleus upon induction of apoptosis. However, overexpression of AIFM3, like AIF, induced cytochrome c release from the mitochondria, cleavage of caspase 3, and ultimately apoptosis, indicating AIFM3 induces apoptosis through caspase activation. Multiple isoforms of AIFM3 are known to exist.

IRGM Antibody: Autophagy, the process of bulk degradation of cellular proteins through an autophagosomic-lysosomal pathway is important for normal growth control and may be defective in tumor cells. It is involved in the preservation of cellular nutrients under starvation conditions as well as the normal turnover of cytosolic components. Two of the strongest hits implicate genes IRGM and ATG16L1, which encode proteins thought to be critical to the autophagy pathway and being significantly associated with Crohn's disease. IRGM induces autophagy and generates large autolysosomal organelles as a mechanism for the elimination of intracellular Mycobacterium tuberculosis. In mouse, IRGM belongs to a family of gamma-interferon-induced GTP-binding proteins of approximately 48 kDa that also includes IRGM2 and IRGM3; this antibody may also recognize these proteins.
