This gene encodes a member of the E2F transcription factor protein family. E2F family members play a crucial role in control of the cell cycle and of the action of tumor suppressor proteins. Many E2F proteins contain several evolutionarily conserved domains: a DNA binding domain, a dimerization domain which determines interaction with the differentiation regulated transcription factor proteins (DP), a transactivation domain enriched in acidic amino acids, and a tumor suppressor protein association domain which is embedded within the transactivation domain. The encoded protein of this gene is atypical because it lacks the transactivation and tumor suppressor protein association domains. It contains a modular suppression domain and is an inhibitor of E2F-dependent transcription. The protein is part of a multimeric protein complex that contains a histone methyltransferase and the transcription factors Mga and Max. Multiple transcript variants have been reported for this gene, but it has not been clearly demonstrated that they encode valid isoforms. Glioma Pathogenesis-Related Protein 1 (GLIPR1) is a member of the CRISP family. It is a single-pass membrane protein with 266 amino acids. GLIPR1 is highly expressed in the lung and kidney and lowly expressed in the heart and liver. It is also highly expressed in cell lines that are derived from nervous system tumors arising from glia; conversely, it is lowly expressed in non-glial-derived nervous system tumor cell lines. GLIPR1 is only expressed in brain tumor glioblastoma multiforme/astrocytoma and not expressed in other nervous system tumors nor normal adult or fetal tissues.

Fc Receptor-Like Protein 1 (FCRL1) is a single-pass type I membrane protein that may function as an activating coreceptor in B cells. FCRL1 is primarily expressed in secondary lymphoid tissues by mature subsets of B cells. It can be detected in the spleen, lymph node, heart, skeletal muscle, kidney, liver and placenta. It is specifically expressed by mature B lineage cells with higher expression at the protein level in naive B cells compared to memory B cells. FCRL1 contains three extracellular C2-like immunoglobulin domains, a transmembrane domain, and a cytoplasmic domain with two immunoreceptor-tyrosine activation motifs and may play a role in the regulation of cancer cell growth.

Poliovirus Receptor (PVR) is a 70 kDa type I transmembrane single-span glycoprotein that belongs to the nectin-like (Necl) family and was originally identified based on its ability to mediate the cell attachment and entry of poliovirus (PV), an etiologic agent of the central nervous system disease poliomyelitis. PVR contains three Ig-like extracellular domains, a transmembrane segment, and a cytoplasmic tail. The normal cellular function of PVR maybe the involvement of intercellular adhension between epithelial cells. Alternate splicing of the PVR mRNA yields four different isoforms ( alpha, beta , gamma , and delta) with identical extracellular domains.

PPP1R14A binds directly to protein kinase C and casein kinase I. Meantime, PPP1R14A is a phosphorylation-dependent inhibitor of smooth muscle myosin phosphatase. PPP1R14A is the inhibitor of PPP1CA. When phosphorylated, PPP1R14A has over 1000-fold higher inhibitory activity, creating a molecular switch for regulating the phosphorylation status of PPP1CA substrates and smooth muscle contraction. In addition, inhibition of PPP1R14A also enhances contraction of smooth muscle in the absence of increment of intracellular Ca2+ concentration.
