Cystatin SA is a member of family 2 of the Cystatin superfamily. Together with Cystatins S and SN, it is produced by the salivary gland and secreted largely in the submandibular/sublingual saliva. Cystatin SA inhibits Cathepsin L, but may not inhibit Cathepsins B and H. The functions of Cystatin SA are largely unknown. However, it has been suggested to play a role in the control of periodontal tissue destruction.

Description:
Endothelial Protein C Receptor (EPCR) is a Vitamin K-dependent Serine Protease that plays a major role in blood coagulation. Binding of Protein C to EPCR leads to the proteolytic activation of PAR1 (Protease-Activated Receptor 1) on endothelial cells and subsequent up-regulation of Protein C-induced genes. EPCR is a type I transmembrane glycoprotein in the CD1/MHC family. It is expressed most strongly in the endothelial cells of arteries and veins in heart and lung. Membrane bound EPCR is released by metalloproteolytic cleavage to generate the soluble receptor. The extracellular domain of human and mouse EPCR shares approximately 61% amino acid sequence homology. EPCR plays an important role in augmenting Protein C activation by the Thrombin-Thrombomodulin complex and in regulating blood coagulation and inflammation. EPCR is found primarily on endothelial cells. Deletion of EPCR function results in embryonic death, at least in part due to placental thrombosis.

Junctional Adhesion Molecule A (JAM-A) is a single-pass type I membrane protein that belongs to the immunoglobulin superfamily. JAM-A contains 2 Ig-like V-type (immunoglobulin-like) domains and Interacts with the ninth PDZ domain. JAM-A is localized to the tight junctions of both epithelial and endothelial cells. JAM-A seems to be involved in epithelial tight junction formation. JAM-A appears early in primordial forms of cell junctions and recruits PARD3. The association of the PARD6-PARD3 complex may prevent the interaction of PARD3 with JAM-A, thereby preventing tight junction assembly. JAM-A plays a role in regulating monocyte transmigration involved in regulating integrity of the epithelial barrier. In the case of orthoreovirus infection, JAM-A serves as receptor for the virus.

Regulator of G protein signaling (RGS) family members are regulatory molecules that act as GTPase activating proteins (GAPs) for G alpha subunits of heterotrimeric G proteins. RGS proteins are able to deactivate G protein subunits of the Gi alpha, Go alpha and Gq alpha subtypes. They drive G proteins into their inactive GDP-bound forms. Regulator of G protein signaling 10 belongs to this family. All RGS proteins share a conserved 120-amino acid sequence termed the RGS domain. This protein associates specifically with the activated forms of the two related G-protein subunits, G-alphai3 and G-alphaz but fails to interact with the structurally and functionally distinct G-alpha subunits. Human Glycoprotein A33 (GPA33) is a single-pass type I membrane protein, belongs to the CTX family of cell adhesion molecular within the immunoglobulin family, can be expressed in normal gastrointestinal epithelium and in 95% of colon cancers. GPA33 consists of one Ig-like C2-type domain and one Ig-like V-type domain. The predicted mature protein includes a single transmembrane domain, a extracellular region and a intracellular tail. Intracellular traffic and recycling to the cell surface appear to play an important role in GPA33 function and to have an influence on its surface density superseding translation regulation. GPA33 has become a promising target of immunologic therapy strategies. GPA33 may also play a important role in cell-cell recognition and signaling.
