Sumo Antibody: The sumo family of proteins is related both structurally and functionally to ubiquitin in that they are post-translationally attached to the e-amino group of a lysine residue of the substrate protein. This sumoylation plays a number of roles in DNA replication and repair, protein targeting to various subnuclear structures, and the regulation of numerous cellular processes including the inflammatory response in mammalian cells. Sumo was initially identified as a covalent modification of RanGAP1 in studies on nuclear import in mammalian cells. More recently, sumo has been shown to be involved in the regulation of transcription factors, possibly by enhancing their interactions with co-repressors. Sumo is also thought to play some role in the modulation of ubiquitin-mediated degradation of proteins by acting as an inhibitor. At least four different isoforms of sumo are known to exist; Sumo antibody will only recognize isoform 1.

Lipe Antibody: Although initially described as an adipocyte-specific triacylglycerol lipase, lipe (also known as hormone-specific lipase or HSL) is expressed in multiple tissues and cell lines. It plays multiple roles in lipid metabolism, including hormone-stimulated lipolysis in adipose tissue and the hydrolysis of cholesterol esters. Lipe is expressed as a long and a short form, generated by use of alternative translational start codons. The long form is expressed in steroidogenic tissues such as testis, where it converts cholesterol esters to free cholesterol for steroid hormone production. The short form is expressed in adipose tissue, among others, where it hydrolyzes stored triglycerides to free fatty acids. Recently, it was observed that the lack of lipe in genetically obese leptin-null mice inhibited obesity and adipogenesis, suggesting that lipe plays a major role in adipocyte proliferation.

Mouse myeloid cell surface antigen CD33(CD33) is a member of the immunoglobulin superfamily and SIGLEC (sialic acid binding Ig-like lectin) family. CD33 contains one Ig-like C2-type domain and one Ig-like V-type domain. CD33 is a putative adhesion molecule of myelomonocytic-derived cells that mediates sialic-acid dependent binding to cells. CD33 preferentially binds to alpha-2,6-linked sialic acid. The sialic acid recognition site may be masked by cis interactions with sialic acids on the same cell surface. In the immune response, CD33 may act as an inhibitory receptor upon ligand induced tyrosine phosphorylation by recruiting cytoplasmic phosphatase(s) via their SH2 domain(s) that block signal transduction through dephosphorylation of signalling molecules. CD33 induces apoptosis in acute myeloid leukemia. CD33 is becoming increasingly important as a target of antibody-mediated therapy in acute myeloid leukaemia (AML).

MUC1 is a membrane bound, glycosylated phosphoprotein. The protein is anchored to the apical surface of many epithelia by a transmembrane domain, with the degree of glycosylation varying with cell type. It also includes a 20 aa variable number tandem repeat (VNTR) domain, with the number of repeats varying from 20 to 120 in different individuals. The protein serves a protective function by binding to pathogens and also functions in a cell signaling capacity. Overexpression, aberrant intracellular localization, and changes in glycosylation of this protein have been associated with carcinomas.

Decoy receptor 3 (DcR3; TNFRSF6B; M68) is a member of the TNF receptor superfamily. DcR3 shares sequence identity with OPG (31%), TNF-RII (29%) and Fas (17%) and is expressed in a variety of different tissues and at high levels in many malignant tumours. Ligands of DcR3 include Fas ligand, homologous to lymphotoxin showing inducible expression and competing with HSV glycoprotein D for herpes virus entry mediator (a receptor expressed by T lymphocytes), TNF-like molecule 1A and heparan sulfate proteoglycans. Programmed Death-1 (PD-1, CD279) is a type I transmembrane protein belonging to the CD28/CTLA-4 family of immunoreceptors that mediate signals for regulating immune responses. Members of the CD28/CTLA-4 family have been shown to either promote T cell activation (CD28 and ICOS) or downregulate T cell activation (CTLA-4 and PD-1). CD279 is expressed on activated T-cells, B-cells, myeloid cells and on a subset of thymocytes. In vitro, ligation of CD279 inhibits TCR-mediated T-cell proliferation and production of IL-1, IL-4, IL-10 and IFN-gamma. In addition, CD279 ligation also inhibits BCR mediated signallling. CD279 deficient mice have a defect in peripheral tolerance and spontaneously develop autoimmune diseases.
