Description:
The assembly and stability of the RNA polymerase II transcription pre-initiation complex on a eukaryotic core promoter involves the effects of TFIIA on the interaction between TATA-binding protein (TBP) and DNA. ALF is a germ cell-specific counterpart of the large (alpha/beta) subunit of general transcription factor TFIIA that is able to stabilize the binding of TBP to DNA and may be uniquely important to testis biology.

Description:
Mouse interleukin-16(IL-16) is a single chain non-glycosylated polypeptide. IL-16 is widely expressed in human tissues including spleen, thymus, lymph nodes, peripheral leukocytes, bone marrow and cerebellum. IL-16 plays an important role instimulating a migratory response in CD4+ lymphocytes, monocytes, and eosinophils, inducing T-lymphocyte expression of interleukin 2 receptor. It was originally identified as a CD8+ T cell-derived chemoattractant for CD4+ cells. In addition to its chemotactic properties, IL-16 has also been shown to suppress HIV-1 replication in vitro and appears to be involved in transcriptional regulation of SKP2 and is probably part of a transcriptional repression complex on the core promoter of the SKP2 gene. It may act as a scaffold for GABPB1 (the DNA-binding subunit the GABP transcription factor complex) and HDAC3 thus maintaining transcriptional repression and blocking cell cycle progression in resting T-cells.

Description:
Mouse Interleukin-4(IL-4) is a monomeric, Th2 cytokine that shows pleiotropic effects during immune responses. It is a glycosylated polypeptide that contains three intrachain disulfide bridges and adopts a bundled four alphahelix structure. IL4 exerts its effects through two receptor complexes, Participates in at least several B-cell activation processes as well as of other cell types. IL4 is primarily expressed by Th2biased CD4+T cells, mast cells, basophils, and eosinophils. It promotes cell proliferation, survival, and immunoglobulin class switch to IgG1 and IgE in mouse B cells, acquisition of the Th2 phenotype by naïve CD4+T cells, priming and chemotaxis of mast cells, eosinophils, and basophils, and the proliferation and activation of epithelial cells. IL4 plays a dominant role in the development of allergic inflammation and asthma. It also regulates the expression of the low affinity Fc receptor for IgE (CD23) on both lymphocytes and monocytes.

Description:
HOXD4 belongs to the homeobox family of genes. The homeobox genes encode a highly conserved family of transcription factors that play an important role in morphogenesis in all multicellular organisms. Mammals possess four similar homeobox gene clusters, HOXA, HOXB, HOXC and HOXD, located on different chromosomes, consisting of 9 to 11 genes arranged in tandem. This gene is one of several homeobox HOXD genes located at 2q31-2q37 chromosome regions. Deletions that removed the entire HOXD gene cluster or 5' end of this cluster have been associated with severe limb and genital abnormalities. The protein encoded by this gene may play a role in determining positional values in developing limb buds. Alternatively spliced variants have been described but their full length nature has not been determined. This gene belongs to the homeobox family of genes. Description:
Kynurenine aminotransferases (KATs) are pyridoxal-5’-phosphate-dependent enzymes that catalyse the conversion of L-kynurenine into kynurenic acid, a neuroactive metabolite whose unbalancing is associated with a number of brain disorders. Biochemical and structural investigations revealed that L-kynurenine (L-KYN) recognition by hKAT II is achieved by exploiting structural features that are peculiar of this isoform, thus offering the possibility to select/design inhibitor molecules specifically targeting hKAT II to be used as modulators of kynurenic acid synthesis in the CNS. hKAT II is one of the aminotransferases involved in the pyridoxal 5'-phosphate (PLP)-dependent irreversible transamination of L-kynurenine (L-KYN) to kynurenic acid (KYNA) in the central nervous system. When assayed in vitro the protein also displays beta-elimination activity.

Description:
Human nicotinate phosphoribosyltransferase (NAPRTase) is localised in the cytoplasm and is involved in biological processes such as NAD biosynthetic and metabolic processes, nicotinamide metabolic process, nicotinate nucleotide salvage, response to oxidative stress and water-soluble vitamin metabolic process. It functions by catalyzing the conversion of nicotinic acid (NA) to NA mononucleotide (NaMN) and is essential for NA to increase cellular NAD levels and prevent oxidative stress of the cells. It is a crucial factor in the NAD+ biosynthesis pathway. Catalytic activity: Beta-nicotinate D-ribonucleotide + diphosphate = nicotinate + 5-phospho-alpha-D-ribose 1-diphosphate.

Description:
M13 endopeptidases are single-pass, transmembrane zinc-metallopeptidases responsible in human for the regulation of the biological activity of many hormones and peptides and are involved in many important processes such as blood pressure regulation (Neprilysin or NEP1), cardiovascular development (Endothelin Converting Enzyme-1 or ECE-12), prevention of haemolytic reaction (KELL) and phosphate homeostasis (PHEX). Zmp1 is a soluble enzyme member of the M13 endopeptidases family that plays a key role in Mycobacterium tuberculosis pathogenicity with a still unknown mechanism. Recently, it was shown to suppress inflammasome activation by inhibiting caspase-1 activation, thus preventing processing of pro-IL-1beta into IL-1beta and the consequent phagosome maturation.
