NFYA founctions as a heterotrimeric transcription factor , which is composed of three components, NF-YA, NF-YB and NF-YC, binds to CCAAT motifs in the promoter regions in a variety of genes. Lumican is a 40 kD secreted protein which belongs to the small leucine-rich repeat proteoglycans (SLRPs) and the class II subfamily. Human Lumican is synthesized as a 338 amino acid precursor then cut the 18 aa signal sequence. The mature Human Lumican contains 12 leucine-rich repeats (LRRs), 4 potential sites of N-linked glycosylation, and a C- terminal with two conserved cyst-eines. Lumican can be existed in extracellular matrix of human articular cartilage. Lumican participates in the maintenance of tissue homeostasis and regulates cellular functions in vivo, such as cell proliferation, adhesion, migration, and differentiation. The overexpression of lumican has been correlated to colorectal tumor, breast, neuroendocrine, and pancreatic cancers.

Surfactant Pulmonary-Associated Protein D (SP-D) is a 43 kDa member of the collectin family of innate immune modulators. Its principal components consist of a collagen-like region and a C-terminal carbohydrate recognition domain (CRD), a structure that places it in a subset of pattern recognition proteins termed defense collagens. SP-D is constitutively secreted by alveolar lining cells and epithelium associated with tubular structures and induced in cardiac smooth muscle and endothelial cells. It binds both secreted and transmembrane proteins that transduce its function. It binds human neutrophil defensins, modulating influenza anti-viral defense. It binds MD-2/LY96, a secreted protein that cooperates with Toll-like receptors (TLRs) in the response of macrophages to bacterial lipopolysaccharides (LPS) or cell wall components. It also binds macrophage CD14 and TLRs directly, blocking binding of LPS and down-regulating TNF- alpha secretion. SP-D binding of both SIRP alpha and the calreticulin/CD91 complex on macrophages allows for a graded response to environmental challenge.

Granulocyte-macrophage colony-stimulating factor (GM-CSF) is also known as Colony stimulating factor 2 (granulocyte-macrophage), is a cytokine initially characterized by its ability to induce colonies of granulocytes and macrophages from myeloid progenitor cells, and is secreted by macrophages, T cells, mast cells, endothelial cells and fibroblasts. GM-CSF is a cytokine that functions as a white blood cell growth factor. GM-CSF stimulates stem cells to produce granulocytes (neutrophils, eosinophils, and basophils) and monocytes. Monocytes exitthe circulation and migrate into tissue, whereupon they mature into macrophages and dendritic cells. Thus, it is part of the immune/inflammatory cascade, by which activation of a small number of macrophages can rapidly lead to an increase in their numbers, a process crucial for fighting infection. The active form of the protein is found extracellularly as a homodimer. Human GM-CSF glycosylated in its mature form. As a part of the immune/inflammatory cascade, GM-CSF promotes Th1 biased immune response, angiogenesis, allergic inflammation, and the development of autoimmunity, and thus worthy of consideration for therapeutic target. GM-CSF has also recently been evaluated in clinical trials for its potential as a vaccine adjuvant in HIV-infected patients. The preliminary results have been promising. GM-CSF is also used as a medication to stimulate the production of white blood cells following chemotherapy.

Leukocyte-Associated Immunoglobulin-Like Receptor 2 (LAIR2) is a secreted, 131 amino acid protein that contains one Ig-like C2 type domain, making it a member of the Ig superfamily. When compared to LAIR-1, its transmembrane counterpart, it shares 83% amino acid identity across the signal sequence and extracellular domains; although one is secreted and one is membrane-bound, the two LAIR proteins are thought to have arisen from a common gene ancestor and appear to share similar adhesion profiles. This suggests that LAIR-2 may compete with LAIR-1 for ligand binding. A 114 amino acid alternate splice form of LAIR-2 is truncated at the C terminus, but retains the entire Ig domain. The expression profile of these splice forms, and the presence of orthologs in other species, have not been reported.

Cellular repressor of E1A genes (CREG) is an evolutionarily conserved lysosomal protein, and an important new factor in regulating tissue homeostasis that has been shown to antagonise injury of tissues or cells. CREG contains three mannose 6-phosphate (M6P) markers, and depends on interactions with M6P receptors for efficient delivery to lysosomes, which is implicated in the regulation of lysosomal functions. This protein shares limited sequence similarity with E1A and binds both the general transcription factor TBP and the tumor suppressor pRb in vitro. CREG plays an important role in the control of cell growth and differentiation. It has been shown that CREG antagonises transcriptional and cellular transformation by the adenoviral E1A oncoprotein, induces differentiation while attenuating cellular proliferation, regulates the levels of the signaling kinases ERK1/2, and mediates glucocorticoid-induced proliferation of ileal epithelial cells. CREG is widely expressed in adult tissues, such as the brain, heart, lungs, liver, intestines and kidneys in mice, but is not markedly expressed in pluripotent embryonic stem cells.

CTGF causes a variety of cellular responses including reduced cell adhesion and enhanced cell migration and proliferation. CTGF has also been shown to be essential for epithelial to mesenchymal transition (EMT), a process whereby normal functioning cells morph into ones that produce mainly scar tissue (of which collagen in the major protein component).
