Stimulating T cells with CMV (IE-2) Peptide pool releases downstream cytokines and upregulates activation markers, enabling antigen-specific T cells to be detected or isolated for analysis. CMV (IE-2) Peptide pool is a lyophilised mixture of 143 peptides from immediate early protein 2 (IE2) of human cytomegalovirus (CMV; strain AD169), and consists of 15-mer peptides with 11-amino-acid overlaps that cover amino acids 1 to 580 on IE2. IE2 is critical for viral infection and for the expression of CMV early genes (Marchini <i>et al.</i>). It also controls transcription initiation for a subset of early-late and late viral promoters via RNA polymerase II (Li <i>et al.</i>). IE2, along with IE1, may contribute to the survival of CMV-infected cells by blocking tumor necrosis factor alpha (TNF-α)-induced apoptosis (Zhu <i>et al.</i>). Viral peptide pools are useful for a broad range of applications, including vaccine development, immunological research, and diagnostic assay development.

Fibroblast growth factor 7 (FGF-7) is a member of the FGF family, and acts exclusively through a subset of FGF receptor isoforms expressed predominantly by epithelial cells (Finch and Rubin). FGF-7 seems to act specifically on epithelial cells and stimulates proliferation, migration, and differentiation of these cells, and also participates in epithelial protection and repair both in vitro and in vivo (Finch and Rubin; Werner). In contrast, FGF-7 is produced solely by cells of mesenchymal origin, and functions as a paracrine mediator of mesenchymal-epithelial communication (Rubin <i>et al.</i>). FGF-7 has also been shown to supplement several wound-healing properties of bioengineered skin (Erdag <i>et al.</i>) and to induce autophagy in human keratinocytes (Belleudi <i>et al.</i>). Dickkopf-related protein 1 (DKK-1) is a member of the Dickkopf family and is a secreted protein that inhibits the canonical WNT pathway by competitive binding to low-density lipoprotein receptors (LRP)-5 and 6 with high affinity, thereby decreasing β-catenin protein stability (Niehrs). DKK-1 regulates embryonic development and contains two conserved cysteine-rich domains separated by a linker region and an N-terminal signal peptide (Krupnik <i>et al.</i>; Lieven <i>et al.</i>). A family of human DKK-related genes composed of DKK-1, DKK-2, DKK-3, and DKK-4 have been characterized together with a unique DKK-3 related protein termed Soggy (Krupnik <i>et al.</i>). DKK-1 has been shown to support the generation of myeloid-derived suppressor cells (MDSCs) and thus is a negative regulator of antitumor immune responses (D’Amico <i>et al.</i>). DKK-1 from thrombocytes is an important regulator of leukocyte infiltration and induces Th2 cell polarization and potentiates Th2 cell cytokine expression (Chae <i>et al.</i>). DKK-1 has also been shown to drive cardiac and retinal differentiation from induced pluripotent stem (iPS) cells (Lian <i>et al.</i>). SARS-CoV-2 Recombinant Spike Protein, aa319-541 is expressed in <i>Pichia pastoris</i> and is one of four structural proteins encoded by the SARS-CoV-2 genome. The Spike Protein plays a key role in attachment to host cells allowing invasion through clathrin-mediated endocytosis. The spike protein can be cleaved by host cell proteases after aa685 to yield the N-terminal S1 subunit, and C-terminal S2 region. The S1 subunit is responsible for interacting with the host cell receptor (angiotensin-converting enzyme II) through a receptor-binding domain that is highly conserved with SARS-CoV. The S1 subunit has two conformations: a ‘down’ conformation in which the receptor is inaccessible, and an ‘up’ conformation in which the receptor is accessible. These conformational changes are key for monoclonal antibody drugs and vaccine development. At the amino terminus of the polypeptide chain, SARS-CoV-2 Recombinant spike protein contains a polyhistidine tag and a SUMOstar site.

Human Recombinant Angiotensin-Converting Enzyme 2 (ACE2) Protein, aa18-740, is expressed in HEK293 cells. ACE2 activity results in vasodilation by degrading vasoconstrictor angiotensin II to produce vasodilator angiotensin. Decreased expression of this enzyme is associated with cardiovascular disease and possibly also glomerular injury in diabetic nephropathy. ACE2 contains a peptidase domain at the N-terminus and a single transmembrane helix. ACE2 is also the primary human receptor for the surface spike glycoprotein of SARS-CoV-2 and SARS-CoV. This receptor is expressed in human lung and small intestine epithelia as well as the heart, kidney, and testes. The peptidase domain of ACE2 binds to the protein-binding domain of the S1 protein subunit of the spike protein; this binding results in a cleavage site becoming exposed. Cleavage, or ‘protein priming’, is done by TMPRSS2, a cellular serine protease, and by cathepsins. This cleavage activates the S2 subunit of the spike protein, allowing the fusion of the viral membrane to the host cell membrane. At the carboxy terminus, Human Recombinant ACE2 Protein contains a TEV site and a human IgG1 Fc tag.

Interleukin 1 receptor antagonist (IL-1RA) is a member of the IL-1 family that binds to IL-1 receptors but does not induce any intracellular signaling (Arend <i>et al.</i>). IL-1RA is a natural regulator of IL-1’s biological activity. It binds to the IL-1 receptors with similar affinity as IL-1, thereby inhibiting the proinflammatory activities of IL-1ɑ and IL-1ꞵ (Kinne <i>et al.</i>). In particular, mesenchymal stem cells have been shown to enhance tissue repair in an IL-1RA-dependent manner through the suppression of IL-1ꞵ production in dermal macrophages and enhanced expansion of immunosuppressive regulatory T cells in the skin (Harrell <i>et al.</i>). IL-1RA has also been shown to help in the treatment of rheumatoid arthritis (Cutolo), sepsis, asthma (Mao <i>et al.</i>), and inflammatory bowel diseases (Dosh <i>et al.</i>). Ciliary neurotrophic factor (CNTF) is a neurotrophic factor that belongs to the four-helix bundle cytokine family and is structurally related to interleukin 6 (IL-6), interleukin 11 (IL-11), leukemia inhibitory factor (LIF), and oncostatin M (OSM). CNTF binds to its receptor CNFTRα and induces formation of a heterodimer of the signal transducing IL-6 receptor gp130 and LIF receptor (LIFR)-β, which triggers JAK/STAT, ERK, and PI3K signaling cascades (Schuster <i>et al.</i>). CNTF plays an important role in neurogenesis and the differentiation of neural stem cells and has been suggested to possess a therapeutic role in treating neurological disorders (Ding <i>et al.</i>; Oppenheim <i>et al.</i>). CNTF has also been shown to protect rod photoreceptors from light-induced damage and have therapeutic effects on retinal degenerative diseases caused by genetic defect or damage induced by toxins, autoantibodies, or strong light (Pernet <i>et al.</i>; Rhee <i>et al.</i>). Another therapeutic role of CNTF has been reported in protecting oligodendrocytes from death induced by apoptosis (Louis <i>et al.</i>). Additionally, CNTF is commonly used to differentiate human pluripotent stem cell (hPSC)-derived neural progenitor cells into astrocytes (Krencik and Zhang). This product is animal component-free.

Leukemia inhibitory factor (LIF) is an interleukin 6 class cytokine that regulates a broad variety of developmental functions. After LIF binds to LIF receptor (LIFR), LIFR associates with gp130 and activates JAK/STAT and MAPK signaling (Auernhammer and Melmed; Suman <i>et al.</i>). LIFR activation of STAT3 is essential for maintaining the mouse embryonic stem cell phenotype (Niwa <i>et al.</i>). Produced by the endometrium, LIF plays an important autocrine and paracrine role in implantation by regulating proliferation, invasion, and differentiation of trophoblasts following blastocyst attachment (Auernhammer and Melmed; Suman <i>et al.</i>). Human LIF can be used for the maintenance of mouse embryonic stem cells, however mouse LIF cannot bind to the human receptor, thus rendering mouse LIF inactive (Dahéron <i>et al.</i>). LIF is produced by CD4+ and activated regulatory T cells, and promotes Foxp3 expression, while repressing Th17 lineage-specific genes (Metcalfe). LIF is also secreted by mesenchymal stromal cells, where it supports hematopoiesis and immune modulation (Nasef <i>et al.</i>). This product is animal component-free.
